【摘要】目的：观察电针不同原穴对急性心肌缺血家兔心交感神经电活动及心功能的影 响，了解心交感神经在针刺调整急性心肌缺血过程中的作用机制。方法：健康青紫蓝家 兔 40 只，随机选 8 只作正常组，其余家兔通过静脉注射垂体后叶素复制急性心肌缺血模 型，将模型复制成功的家兔随机分为模型组、电针"神门"组、电针"太溪"组和电针 非经穴组。采用BIOPAC生物信号采集系统记录心交感神经电信号和心功能指标的变化， 同时接入SKY-A8 生物信号处理系统进行数据处理。结果：急性心肌缺血模型复制后，家 兔的左心室收缩压力峰值(LVSP)及室内压上升段最大变化率(+dp/dt max)明显下降， 室内压下降段最大变化率(-dp/dt max)升高。在停针即刻，电针"神门"组家兔的+dp/dt max、-dp/dt max及LVSP与模型组、电针"太溪"组及电针非经穴组相比较差异有统计学 意义(P<0.05) 。急性心肌缺血模型复制后家兔心交感神经电活动的频率一过性增多后急 剧减少，与正常组比较差异有统计学意义(P<0.05) 。在停针即刻，电针"神门"组家兔 的心交感神经电活动的频率与模型组、电针"太溪"组及电针非经穴组家兔差异均有统 计学意义(P<0.05) ，与正常组比较，差异性无统计学意义。结论：电针"神门"穴可明 显改善急性心肌缺血家兔的心功能及心交感神经电活动，促进机体心脏功能的恢复；电 针不同原穴对急性心肌缺血家兔的心功能及心交感神经电活动的影响存在相对特异性， 其中以"神门"穴的调整作用较为明显，而"太溪"穴及非经穴的作用不明显；电针相 关经穴能调整心交感神经的活动，从而实现抗心肌缺血的效应。交感神经是针刺抗心肌 缺血的主要外周传出途径之一。 【关键词】电针；穴位特异性；心肌缺血；家兔 【Abstract】Objective: To observe the influence of electroacupuncture (EA) of different YuanPrimary acupoints on cardiac sympathetic nerve activity (CSNA) and cardiac function in rabbits with acute myocardial ischemia (AMI), and to understand the interaction mechanisms of cardiac sympathetic nerve in adjusting acute myocardial ischemia. Method: A total of forty healthy rabbits were selected and 8 were randomly chosen as normal control. The rest 32 rabbits were inducted acute myocardial ischemia (AMI) models by intravenous Pituitrin, then the AMI model rabbits were randomly divided into 4 groups, 8 as model group, and 8 as EA on Shenmen (HT 7) group, 8 as EA on Taixi (KI 3) group, and 8 as EA on nonpoint group. 【摘要】目的：从基因水平揭示下丘脑中与针刺心经干预心肌缺血相关的基因表达谱， 探索针刺心经抗干预心肌缺血的中枢作用机制。方法：采用结扎冠状动脉前降支法复制 大鼠急性心肌梗死模型。比较心经组与模型组、模型组与正常组的心脏基因表达谱变化。 结果：与正常组比较，模型组大鼠下丘脑差异表达 2 倍以上基因(包括EST)中，上调表 达 73 个，下调表达 92 个；与模型组比较，电针心经组下丘脑中 2 倍以上差异表达基因 有 190 个上调，34 个下调，涉及细胞代谢、脂质代谢、免疫反应、G蛋白偶联受体、离子 转运、信号转导等多种因子；而电针模型肺经组仅有 57 个基因表达上调，26 个基因表达 下调。结论：心经组、肺经组中大于 2 倍的差异表达基因的数目和类型有较大差异，提 示针刺心经干预心肌缺血作用有相对特异性。 【关键词】针刺；心经；肺经；寡核苷酸序列分析；下丘脑；急性心肌缺血; 大鼠 【Abstract】Objective: To discovery the central mechanism of acupuncture Heart Meridian precondition myocardial ischemia in gene expression pattern, the authors applied gene chip tech to filter variably expressed genes in hypothalamus. Methods: Rats were seperated into normal, model, acupuncture Heart Meridian group and acup Lung Meridian group randomly. Acute myocardial ischemia rat model was made with ligation left anterior descending branch of the coronary artery. After model succeed, select hypothalamus seperately and mixed the same group together of 3 rats. Then applied Rats U230A genechip refered by Affymetrix Co. to compare the variations between these groups. Results: To compare with normal group, differential expression genes and expression sequence tags (ESTs) in model group were 73 with signal log ratio ≥ 1 and 92 with signal log ratio ≤ -1 , mainly included ion channel, calcium/ calmodulin-dependent protein kinase II inhibitor, antigen and so on. Similarly, compared with model group, differential expression genes and expression sequence tags (ESTs) were 190 with signal log ratio ≥1 and 34 with signal log ratio ≤-1 in acupuncture Heart Meridian group, mainly included 5-hydroxytryptamine receptor, cellular metabolism, fatty, immuno reaction, G-protein coupled receptors, ion transport, signal transductions and so on, while in acupucnture Lung Meridian, the number is 57 and 26 correspondly.
