In this study, efforts were made to utilize hulless barley (variety BHS352) to enhance the nutritive value of chapatti and biscuit made from wheat flour. Barley flour was added to wheat flour in different ratios (5 to 30%). Antioxidant activity, total phenolic content and β-glucan content were determined both in flour blends and their products. Changes in physical quality and taste of chapatti and biscuits after blending of hulless barley flour with wheat flour were measured. The chapatti quality score decreased by 15% and biscuit spread factor by 33% after 30% barley flour blending. Significant increase in β-glucan content and antioxidant activity of flour blends and their products was observed at 30% blending level. The phenolic content increased from 63 to 135 µg for biscuits and 237 to 287 ug GAE/g for chapatti with blending of 30% barley flour.
The study investigates the antioxidant potential of commonly used vegetable oils viz., coconut (CNO), sunflower (SFO), rice bran (RBO), groundnut (GNO), sesame (SESO) and mustard oil (MO), where the oils were extracted with methanol; and these methanolic extracts were used for the antioxidant studies. Fatty acid composition by gas chromatography, physicochemical parameters such as specific gravity, refractive index, acid value, iodine value and peroxide value were also analysed. Total phenolic content (TPC) and antioxidant activities of the oils (DPPH, ABTS, superoxide and nitric oxide radical scavenging assays) were evaluated. Phenolic compounds were identified by high-pressure liquid chromatography (HPLC). Percentage TPC was highest in groundnut oil with 3.09 mg/100 g oil followed by coconut oil, rice bran oil, mustard oil, sunflower oil and sesame oil with 1.8, 0.89, 0.56, 0.49 and 0.33 mg/100 g oil, respectively. The methanolic extracts of oils showed potential antioxidant activity in terms of super oxide and nitric oxide radical scavenging activity.
PRACTICAL APPLICATIONSThe present study indicated that vegetable oils can contribute to the dietary intake of antioxidants, depending on the processing condition. This is the first study of its kind to investigate and compare the phenolic content by HPLC and also the radical scavenging activity (DPPH, ABTS, superoxide and nitric oxide) of different oils. It was observed that the unrefined oils posses better antioxidant activity than the refined oils, which imply the importance of optimizing and/or modifying the existing steps in the vegetable oil processing to retain the polyphenolic content in vegetable oil without compromising other physicochemical parameters. The study has brought out the importance of polyphenolic compounds in vegetable oil as radical scavenging agents that could be beneficial in the management of many degenerative diseases. This may provide vegetable oils a new dimension in health management other than its traditional uses.
BackgroundThe R2 repeat region of GLURP has been reported as a good genetic marker for Plasmodium falciparum genotyping. Proper knowledge of the extent and nature of P. falciparum genetic diversity using highly immunogenic R2 repeat region in malaria-endemic areas is a crucial element to understand various aspects related to immunity acquisition and disease pathogenesis.MethodsPopulation diversity of P. falciparum GLURP and amino acid sequence repeats in GLURP R2 region was studied in malaria-endemic Assam state, northeast India and compared at an interval of five years during 2005 (Group-A) and 2011 (Group-B).ResultsOf the 66 samples, a total of 55 samples showed positive PCR bands for GLURP R2 region and altogether ten types of alleles with size ranging from 501 bp to 1,050 bp (50 bp bin) were observed and coded as genotypes I-X. In Group-A (n = 29), 24 samples were found infected with single, four with double and one with triple P. falciparum genotype, while in Group-B (n = 26), single genotype was found in 23 samples, double in two samples and triple in one sample. Genotype IV showed significant increase (p = 0.002) during 2011 (Group-B). Genotypes I to V were more common in Group-B (62%), however genotypes VI to X were more frequently distributed in Group-A. The expected heterozygosity was found slightly higher in Group-A (HE = 0.87) than Group-B (HE = 0.85), whereas multiplicity of infection (MOI) in Group-A (MOI = 1.21 ± 0.49) and Group-B (MOI = 1.12 ± 0.43) did not display significant variation. The amino acid repeat sequence unit (AAU) DKNEKGQHEIVEVEEILPE (called ‘a’) was more frequent in the well-conserved part of R2 repeat region.ConclusionThe present study is the first extensive study in India which has generated substantial data for understanding the type and distribution of naturally evolved genetic polymorphism at amino acid sequence level in GLURP R2 repeat region in P. falciparum. There was decrease in the PCR amplicon size as well as the number of AAU [amino acid repeat unit] in Group-B displaying the bottleneck effect.
