Analytical HPLC methods using derivatized amylose chiral stationary phases were developed for the resolution of enantiomers of methoxy an ethyl tetrahydronaphthalenic derivatives, new agonist and antagonist ligands for melatonin receptors. Separation was by normal phase methodology with a mobile phase of n-hexane-alcohol (methanol, ethanol,l-propanol or 2propanol) in various proportions, and a silica-based, amylose tris-(S)-l-phenylethylcarbamate (Chiralpak AS), or tris-3,5-dimethylphenylcarbamate (Chiralpak AD). The mobile phase and the chiral stationary phase were optimized for best resolution. The effects of concentration of alcohol, various aliphatic alcohols in the mobile phase were studied. The effects of substitution were analysed. Baseline separation (Rs > 1.5) was easily obtained in many cases.
In summary, PTB decreased renal AGE accumulation but did not ameliorate glomerular lesions or proteinuria. Thus, cleavage of AGE by PTB is not sufficient to prevent development of diabetic nephropathy in C57BL/6 mice.
