The aim of this work is to develop and validate a dissolution test for glibenclamide tablets. Optimal conditions to carry out the dissolution test are 500 mL of phosphate buffer at pH 8.0, paddles at 75 rpm stirring speed, time test set to 60 min and using equipment with six vessels. The derivative UV spectrophotometric method for determination of glibenclamide released was developed, validated and compared with the HPLC method. The UVDS method presents linearity (r 2 = 0.9999) in the concentration range of 5-14 µg/mL. Precision and recoveries were 0.42% and 100.25%, respectively. The method was applied to three products commercially available on the Brazilian market.
Recebido em 11/9/07; aceito em 22/4/08; publicado na web 17/9/08 VALIDATION OF METHODS FOR DETERMINATION OF FLUOXETINE CAPSULES. A UV spectrophotometric method was developed and validated and a chromatographic method was adapted from the American Pharmacopeia for the analysis of Fluoxetine Hydrochloride capsules. Ethanol was used as solvent for the spectrophotometric method, with detection and determination at 276 nm. The separation for the chromatographic method was carried out using the reversed-phase column LC-8, triethylamine buffer, stabilizer free tetrahydrofuran and methanol (5:3.5:1.5), pH 6.0 as mobile phase and detection at 227 nm. The results obtained for both methods showed to be accurate, precise, robust and linear over the concentration range 100.00 -300.00 µg/mL and 40.00 -80.00 µg/mL of fluoxetine hydrochloride for the spectrophotometric and chromatographic methods, respectively. The accuracy of the methods was evaluated by a recovery test and showed results between 98.89 and 101.10%.Keywords: fluoxetine; UV spectrophotometry; high performance liquid chromatography.
introduçãoA Organização Mundial de Saúde (OMS) prevê que nos próxi-mos 20 anos, a depressão sairá do quarto para o segundo lugar na lista de doenças dispendiosas e fatais e deverá perder apenas para as enfermidades do coração. Os últimos dados revelaram que hoje a depressão atinge 15% da população, em 70 a 80% dos casos é uma doença crônica recidivante, 1 representando uma parcela significativa de despesas com a saúde. Ainda, o mais importante é o fato de que aproximadamente 15% das pessoas que sofrem de depressão cometem suicídio, principalmente se não forem tratadas de modo correto.
Este trabalho descreve um método espectrofotométrico simples, rápido e de baixo custo para análise de metildopa em medicamentos. O método baseia-se na determinação da absorvância do fármaco a 281nm em HCl 0,01M. Os resultados obtidos mostraram que o método proposto é linear na faixa de concentração de 20,0 a 60,0 mg/mL, com precisão intermediária, repetibilidade e exatidão de acordo com as especificações exigidas pela ANVISA. O método proposto foi comparado com o método oficial e aplicado em 4 amostras.
