In this study, the probiotic potential of Lactobacillus strains isolated from traditional Kurdish cheese was investigated. The Lactobacillus strains were examined for resistance to gastric acidity and bile toxicity, antimicrobial activities, autoaggregation, coaggregation, hydrophobicity, adhesion to Caco-2 cells, and antibiotic susceptibility. The results showed that all strains tested tolerate acid gastric conditions (pH 2.0 and 3.0), and all of them were bile resistant (at 0.3 and 1% concentration). Although no antibacterial activity was detected in vitro assay for the treated (neutralized to pH 6.5 and treated with catalase) cell-free culture supernatant (CFCS) of strains, untreated CFCS showed strong antagonistic activity against two known pathogens bacteria. All strains exhibited a strong autoaggregating phenotype and manifested a high degree of coaggregation with pathogens. On the other hand, majority of studied strains were found sensitive to different antibiotics, such as ampicillin, penicillin, ciprofloxacin, chloramphenicol, erythromycin, rifampicin, and tetracycline, and were resistant to vancomycin and streptomycin. Finally, isolated strains showed good hydrophobicity and adherence to Caco-2 cell line, so they could be exploited for food manufacture.
Summary
Burdock root could be a source of inulin, but scarce studies about its extraction exist. To optimise conventional and ultrasound‐assisted extraction of inulin, various combinations were used. The independent variables for ultrasound extraction were sonication amplitude (20–100%), sonication temperature (20–60 °C) and sonication time (5–25 min) while for conventional extraction the following variables were water to solid ratio (5–15 v/w), temperature (40–90 °C) and time (5–40 min). For each response, a second‐order polynomial model was developed using multiple linear regression analysis. The use of high intensity ultrasound significantly improves the extraction of inulin contained within the body of Burdock root. It was shown that increasing the amplitude (from 20% to 85%) and extraction time increased the extraction yield; however, the effect of temperature was minor. Optimum extraction condition was found to be sonication time: 25 min, sonication amplitude 83.22% and temperature: 36.76 °C.
Yogurt-ice cream is a nutritious product with a refreshing taste and durability profoundly longer than that of yogurt. The probiotic Lactobacillus acidophilus (La-5) cells either in free or encapsulated form were incorporated into yog-ice cream and their survivability were studied. Fructooligosaccharide (FOS) as a prebiotic compound at three levels (0, 4 & 8 % w/w) was added to yogurt-ice cream mix and its effects on some chemical properties, overrun and firmness of product were evaluated. The higher the incorporated FOS concentration, the lower were the pH value and higher the total solid content of treatments. FOS incorporation (8 %) significantly increased the overrun of treatments and reduced their firmness. The viable counts of free probiotics decreased from~9.55 to~7.3 log cfu/g after 60 days of frozen storage while that of encapsulated cells merely decreased less than 1 log cycle. Encapsulation with alginate microbeads protected the probiotic cells against injuries in the freezing stage as well as, during frozen storage.
