The majority of the world’s population lives in urban areas, and regions with the highest under-five mortality rates are urbanising rapidly. This 7-year interrupted time series study measured early access to care and under-five mortality over the course of a proactive community case management (ProCCM) intervention in periurban Mali. Using a cluster-based, population-weighted sampling methodology, we conducted independent cross-sectional household surveys at baseline and at 12, 24, 36, 48, 60, 72 and 84 months later in the intervention area. The ProCCM intervention had five key components: (1) active case detection by community health workers (CHWs), (2) CHW doorstep care, (3) monthly dedicated supervision for CHWs, (4) removal of user fees and (5) primary care infrastructure improvements and staff capacity building. Under-five mortality rate was calculated using a Cox proportional hazard survival regression. We measured the percentage of children initiating effective antimalarial treatment within 24 hours of symptom onset and the percentage of children reported to be febrile within the previous 2 weeks. During the intervention, the rate of early effective antimalarial treatment of children 0–59 months more than doubled, from 14.7% in 2008 to 35.3% in 2015 (OR 3.198, P<0.0001). The prevalence of febrile illness among children under 5 years declined after 7 years of the intervention from 39.7% at baseline to 22.6% in 2015 (OR 0.448, P<0.0001). Communities where ProCCM was implemented have achieved an under-five mortality rate at or below 28/1000 for the past 6 years. In 2015, under-five mortality was 7/1000 (HR 0.039, P<0.0001). Further research is needed to elucidate the mechanisms of action and generalizability of ProCCM.
Information on the replication of viral haemorrhagic fever viruses is not readily available and has never been analysed in a comparative approach. Here, we compared the cell culture growth characteristics of haemorrhagic fever viruses (HFV), of the Arenaviridae, Filoviridae, Bunyaviridae, and Flavivridae virus families by performing quantitative analysis of cell culture supernatants by (i) electron microscopy for the quantification of virus particles, (ii) quantitative real time PCR for the quantification of genomes, and (iii) determination of focus forming units by coating fluorescent antibodies to infected cell monolayers for the quantification of virus infectivity.The comparative analysis revealed that filovirus and RVFV replication results in a surplus of genomes but varying degrees of packaging efficiency and infectious particles. More efficient replication and packaging was observed for Lassa virus, and Dengue virus resulting in a better yield of infectious particles while, YFV turned out to be most efficient with only 4 particles inducing one FFU. For Crimean-Congo haemorrhagic fever virus (CCHFV) a surplus of empty shells was observed with only one in 24 particles equipped with a genome. The complete particles turned out to be extraordinarily infectious.
The PICS bags, originally developed for cowpea storage, were evaluated for sorghum (Sorghum bicolor) preservation. Batches of 25 kg of sorghum grain were stored in 50 kg PICS or polypropylene (PP) bags under ambient conditions for 12 months and assessed for the presence of insect pests and their damage, seed viability and, oxygen and carbon dioxide variations. The grain was incubated for 35 days to assess whether any insects would emerge. After six months of storage, oxygen levels decreased in the PICS bags compared to polypropylene bags. After 12 months of storage, only two pests, Rhyzopertha dominica and Sitophilus zeamais were found in the PICS bags. However, in PP bags there were additional pests including Tribolium castaneum and Oryzeaphilus mercator and Xylocoris flavipes. Grain weight loss and damage caused by these insects in the PP bags were significantly higher compared to those stored in PICS bags. Germination rates of sorghum grains stored in PP bags decreased significantly while no changes were observed in grains stored in PICS bags when compared to the initial germination. After the incubation post storage period, there was a resurgence of R. dominica in sorghum grains from PICS bags but the population levels were significantly lower compared to polypropylene bags. PICS bags preserved the quality and viability of stored sorghum grains and protected it from key insect pests. The PICS technology is effective for long-term sorghum storage but the potential resurgence of insects in low-oxygen environment calls for further research.
Abstract. With the advent of transgenic Bt-cowpea, there is a need to identify the feeding preferences of Maruca vitrata Fab., in order to determine which component of the plant the expression of the toxin needs to be the highest in order to ensure the greatest efficacy of the insecticidal Bt proteins. In the current study we examined the feeding preferences of M. vitrata larvae in a naturally infested cowpea field. We also tested, in the laboratory, the suitability of different flower components for the larval development of M. vitrata. Our results indicated that in the field, all types of flowers, regardless of their age, were infested with M. vitrata larvae. The reproductive organs, in the flowers, were the preferred feeding diet for the larvae. Laboratory bioassays confirmed that the reproductive organs were the optimal tissues for M. vitrata larval development. The implications of these findings for transgenic Btcowpea are discussed.
Maruca vitrata is an economically significant insect pest of cowpea in sub-Saharan Africa. Understanding the seasonal population patterns of M. vitrata is essential for the establishment of effective pest management strategies. M. vitrata larval populations on cultivated cowpea and adult flying activities were monitored in addition to scouting for host plants and parasitoids during 2 consecutive years in 2010 and 2011 in southwestern Burkina Faso. Our data suggest that M. vitrata populations overlapped on cultivated cowpea and alternate host plants during the rainy season. During the cowpea off-season, M. vitrata maintained a permanent population on the wild host plants Mucuna poggei and Daniella oliveri. The parasitoid fauna include three species, Phanerotoma leucobasis Kri., Braunsia kriegeri End. and Bracon sp. Implications of these finding for pest management strategies are discussed.
