Dengue fever is an important vector-borne disease, mainly transmitted by Aedes aegypti. To date, there are no vaccines or effective drugs available against this arboviral disease. As mosquito control is practically the only method available to control dengue fever, alternative and cost-effective pest control strategies need to be explored. The gram-negative enteric bacteria Xenorhabdus and Photorhabdus are symbiotically associated with nematode parasites, which themselves are highly pathogenic for insect larvae. Here, we evaluate the oral toxicity of these entomopathogenic bacteria in A. aegypti larvae. The susceptibility of larvae (third late or fourth early instars) was assessed by exposing them to suspensions containing Photorhabdus luminescens or Xenorhabdus nematophila, respectively. Two diet treatments were tested with larvae fed on pet food and unfed larvae. After 24 h, larvae began to die when exposed to the bacteria. Exposure to P. luminescens killed 73% of the fed and 83% of the unfed larvae, respectively. In comparison, X. nematophila was less pathogenic, killing 52% of the larvae in the fed and 42% in the unfed treatment. Remarkably, cannibalism was observed in all bioassays after exposing larvae to either of the bacterial species. To our knowledge, this is the first report demonstrating the efficiency of these entomopathogenic bacteria for oral A. aegypti killing. Our results provide a promising basis for using these bacteria as bioinsecticides for mosquito control in the future.
This study investigated the effectiveness of successive bioaugmentation, conventional bioaugmentation, and biostimulation of biodegradation of B10 in soil. In addition, the structure of the soil microbial community was assessed by polymerase chain reaction-denaturing gradient gel electrophoresis. The consortium was inoculated on the initial and the 11th day of incubation for successive bioaugmentation and only on the initial day for bioaugmentation and conventional bioaugmentation. The experiment was conducted for 32 days. The microbial consortium was identified based on sequencing of 16S rRNA gene and consisted as Pseudomonas aeruginosa, Achromobacter xylosoxidans, and Ochrobactrum intermedium. Nutrient introduction (biostimulation) promoted a positive effect on microbial populations. The results indicate that the edaphic community structure and dynamics were different according to the treatments employed. CO2 evolution demonstrated no significant difference in soil microbial activity between biostimulation and bioaugmentation treatments. The total petroleum hydrocarbon (TPH) analysis indicated a biodegradation level of 35.7 and 32.2 % for the biostimulation and successive bioaugmentation treatments, respectively. Successive bioaugmentation displayed positive effects on biodegradation, with a substantial reduction in TPH levels.
-(Agaricomycetes fungi community at different forest systems in the northwest of Rio Grande do Sul State, Brazil: seasonal deciduous forest and eucalypt monoculture). Monocultures differ from native forests in relation to plant heterogeneity, variety of substrates, and soil humidity, possibly altering the diversity of organisms, such as the fungi. This study aimed to compare the richness, abundance, and composition of Agaricomycetes fungi in a native forest and in a eucalypt monoculture. The fungi were collected in a seasonal deciduous forest area and in a Eucalyptus grandis monoculture, in the northwest of Rio Grande do Sul State, Brazil. The abundance and richness of fungi were significantly lower in the eucalypt plantation than in the native forest and the species composition differed between the areas. Thus, it was concluded that the eucalypt monoculture negatively affects the Agaricomycetes fungi community. As coletas dos fungos foram realizadas em uma área de Floresta Estacional Decídua e em uma monocultura de Eucalyptus grandis, no noroeste do Estado do Rio Grande do Sul, Brasil. A abundância e riqueza de fungos na monocultura de eucalipto foram significativamente inferiores às encontradas na mata nativa, assim como a composição de espécies diferiu entre as áreas. Algumas rochas utilizadas na produção de fertilizantes contêm impurezas de baixa solubilidade em água, destacando-se alguns fosfatos de Fe e Al, que poderiam ser aplicados em solos alagados, onde as reações de redução podem aumentar a solubilidade. O objetivo deste trabalho foi estudar a liberação do P de fertilizantes contendo impurezas de fosfatos de Fe com baixa solubilidade em água em diferentes solos de várzea do Rio Grande do Sul (RS) sob condições de alagamento. Para realização do experimento foram utilizados seis solos de várzea (dois Gleisssolos, dois Planossolos e dois Chernossolos) de diferentes locais do Estado do Rio Grande do Sul, os quais foram acondicionados em vasos plásticos de 8 L. Cada solo recebeu duas fontes de P (superfosfato simples e superfosfato simples com resíduos de Fe) e uma testemunha sem adição de P, com três repetições. Nos tratamentos com P foram adicionados ao solo 100 mg kg-1 P no solo. Os solos foram alagados e mantidos, em delineamento inteiramente casualizado e três repetições, com uma lâmina de água de 5 cm de altura acima de sua superfície. A solução do solo foi coletada semanalmente, sendo medidos imediatamente o pH e o potencial redox (Eh); posteriormente, procedeu-se, em laboratório, à determinação dos teores de P e Fe. Os resultados mostram redução do Eh e aumento do pH e dos teores de Fe e P em solução para todos os solos. O teor de P na solução não diferiu quanto à fonte, mostrando que o superfosfato simples com impurezas de fosfato de Fe tem potencial de uso em solos de várzea do Rio Grande do Sul cultivados com arroz irrigado por alagamento, mesmo sem a solubilidade em água prevista na legislação brasileira.
