NF-κB-mediated inflammatory phenotypic switching of vascular smooth muscle cells (VSMCs) plays a central role in atherosclerosis and neointimal formation. However, little is known about the roles of circRNAs in the regulation of NF-κB signaling. Here, we identify the involvement of circ-Sirt1 that was one of transcripts of SIRT1 host gene in VSMC inflammatory response and neointimal hyperplasia. First, in the cytoplasm, circ-Sirt1 directly interacts with and sequesters NF-κB p65 from nuclear translocation induced by TNF-α in a sequence-dependent manner. The inhibitory complex of circ-Sirt1-NF-κB p65 is not dependent on IκBα. Second, circ-Sirt1 binds to miR-132/212 that interferes with SIRT1 mRNA, and facilitates the expression of host gene SIRT1. Increased SIRT1 results in deacetylation and inactivation of the nuclear NF-κB p65. These findings illustrate that circ-Sirt1 is a novel non-coding RNA regulator of VSMC phenotype.
BackgroundRecent research supports a significant role of immune checkpoint inhibitors in the treatment of solid tumors. However, relevant reports for programmed death-ligand 1 (PD-L1) and CD8+ tumor-infiltrating lymphocytes (TILs) in pulmonary neuroendocrine tumors (PNETs) have not been fully studied. Therefore, we investigated PNETs for the expression of PD-L1 and infiltration by CD8+ TILs as well as the prognostic value of both factors.MethodsIn total, 159 specimens of PNETs (35 TC, 2 AC, 28 LCNEC, 94 SCLC) were included in this study. Immunohistochemistry (IHC) was used to detect the expression of PD-L1 in these cases. Cases demonstrating ≥5% tumor cell expression or any expression (> 1%) of PD-L1 on immune cells were considered positive. CD8+ TILs both within stroma and tumor areas of invasive carcinoma were analyzed using whole-slide digital imaging. Manual regional annotation and machine cell counts were performed for each case.ResultsPositive expression of PD-L1 was observed in 72 cases (45.3%), including 9 cases (5.7%) with expression exclusively on tumor cells, 46 cases (28.9%) with expression exclusively on immune cells, and 17 cases (10.7%) with the expression on tumor cells and immune cells. PD-L1 expression was associated with necrosis (p < 0.001), high pathologic grade (p < 0.001) and histologic type (p < 0.001). No correlation was observed with overall survival (OS) (p = 0.158) or progression-free survival (PFS) (p = 0.315). In contrast, higher CD8+ T cell density was associated with the absence of vascular invasion (p = 0.004), histologic type (p = 0.005), negative lymph node metastasis (p = 0.005) and lower clinical staging (p = 0.007). Moreover, multivariate analysis revealed that CD8+ stromal TIL was an independent prognostic factor for improved OS (p = 0.009) and PFS (p = 0.002).ConclusionPD-L1 was expressed in approximately half of the PNETs. The majority of the expression was observed in immune cells. Positive expression of PD-L1 showed no correlation with OS or PFS, while higher CD8+ TILs within stroma was proved to be an independent prognostic factor for favorable OS and PFS of PNETs.
Chlorine is commonly used in disinfection processes in wastewater treatment plants prior to discharge of the effluents into receiving waters. Effluent organic matter and humic substances constitute up to 90% of dissolved organic matter (DOM) in receiving water, which induces photogeneration of reactive species (RS) such as excited triplet state of DOM ( 3 DOM*), singlet oxygen ( 1 O 2 ), and hydroxyl radical ( • OH). The RS plays an important role in the attenuation of trace pollutants. However, the effect of chlorine disinfection on the photoreactivity of the DOM has remained unclear. Here, we investigated the physicochemical properties and subsequent RS variation after chlorination of DOM. Solid-state 13 C cross-polarization/magic angle-spinning NMR and Fourier transform ion cyclotron resonance mass spectrometry verified that the aromaticity, electron-donating capacity (EDC), and average molecular weight of DOM decreased markedly after chlorination. It was found for the first time that the photoproduction of 3 DOM*, 1 O 2 , and • OH increased markedly after chlorination of DOM upon irradiation of simulated sunlight. The quantum yields of 3 DOM*, 1 O 2 , and • OH were positively correlated with E2/E3 (ratio of the absorbance at 254 to 365 nm) while negatively correlated with EDC before and after chlorination. These findings highlight the synergetic effect of chlorine disinfection on the photosensitization of DOM under irradiation of sunlight, which will promote the removal of trace pollutants in surface waters.
Human
flap endonuclease 1 (hFEN1) is instrumental in DNA replication
and repair. It is able to cleave the 5′ single-stranded protrusion
(also known as 5′ flap) resulting from strand displacement
reactions. In light of its crucial functions, hFEN1 is now deemed
as a nontrivial target in the DNA damage response system for anticancer
drug development. Herein, we report that myricetin and some natural
flavonoids are able to inhibit hFEN1. Structure–activity relationship,
inhibitory mechanisms, molecular docking, and cancer cell-based assays
have been performed. Our original findings expand the activity of
flavonoids and may pave the way for flavonoid-assisted targeted cancer
therapy.
