This study presents self-organizing maps (SOM) as an alternative method to evaluate genetic diversity in plant breeding programs. Twenty-five genotypes were evaluated in two environments for 11 phenotypic traits. The genotypes were clustered according to the SOM technique, with variable topology and numbers of neurons. In addition to the SOM analysis, unweighted pair group method with arithmetic mean clustering (UPGMA) was performed to observe the behavior of the clustering when submitted to these techniques and to evaluate their complementarities. Genotype ordering according to SOM was consistent with UPGMA results, evidenced by the basic structure of UPGMA groups being preserved in each group of the maps. Regarding genotype arrangement and the group neighbors, maps involving five neurons presented inferior organization efficiency compared to the six-map arrangements in both environments. It was observed that the organization pattern among the rice genotypes evaluated by the maps was complementary to the UPGMA approach, as observed in all scenarios. It can be concluded that self-organizing maps have the potential to be useful for genetic diversity studies in breeding programs.
Genome doubling of haploids is one of the major constraints of large-scale doubled haploid (DH) technology. Improving spontaneous haploid genome doubling (SHGD) is an alternative to overcome this limitation. In this study, we aimed to construct a high-density linkage map based on genotyping by sequencing (GBS) of Single Nucleotide Polymorphism (SNPs), to detect QTL and QTL by environment (Q by E) interactions affecting SHGD, and to identify the best trait for mapping and selection of haploid male fertility (HMF). To this end, a bi-parental population of 220 F2:3 families was developed from a cross between A427 (high HMF) and CR1Ht (moderate HMF) to be used as donor. A high-density linkage map was constructed containing 4,171 SNP markers distributed over 10 chromosomes with an average distance between adjacent markers of 0.51 cM. QTL mapping for haploid fertile anther emergence (AE), pollen production (PP), tassel size (TS), and HMF, identified 27 QTL across three environments, and Q by E interactions were significant. A major QTL was identified on chromosome 5. This QTL explained over 45% of the observed variance for all traits across all environments. The introgression of this major QTL, using marker-assisted backcrossing, has great potential to overcome the need of using colchicine in DH line development.
Alfalfa breeding aimed at trait improvement for livestock feed takes longer periods of time, if compared to many other crops. Therefore, better selection methods are necessary for the success of alfalfa breeding programs. Although knowing about selection methods is quite important, there is a notable lack of information, as regards successful solutions. This study aimed to use direct, indirect and simultaneous selection methods for selecting alfalfa cultivars, based on yield traits and nutritive value. The evaluated traits were subdivided into two groups: forage yield and nutritive value. Selection gains were estimated by direct, indirect and simultaneous selection for each group, considering the selection of the 25 % best cultivars. Direct and indirect selections among genotype averages are not efficient to provide the desirable responses to the whole set of traits. The results for simultaneous selection, using the Tai index, provided a more balanced gain distribution to the set of traits in all cuts. The simultaneous selection allowed the identification of the 5681 and Verdor cultivars in the first cut, as well as ProINTA Patricia in the second cut, as superior in the two groups of evaluated traits.
ABSTRACT. Using commercial cultivars to compose crossing blocks in cotton is a promising strategy, because these materials have desirable agronomic and technological characteristics. The objective of this study was to evaluate the genetic diversity among 16 cotton cultivars cultivated in two environments in the State of Mato Grosso, the largest national producer, using agronomical and technological traits. There was significant effect to cultivars for all traits, while genotype x environment interaction was significant only for average boll weight, short fiber index, and maturity of fibers. Therefore, because of the presence of genotype x environment interaction for three traits, we chose to study genetic diversity among cotton cultivars separately in each environment and investigate the interaction impact on the diversity among genotype pairs. Based on agronomical and technological performance and genetic diversity among cultivars in both environments, the most promising cross involves FM 910 and LD CV 02. We also observed that lint percentage and average boll weight presented a higher discrimination capacity in both environments.
