Methylparaben is a commonly used antimicrobial in cosmetics that has been shown to have negative effects on mammalian cells. Human melanoma M624 cells were treated with 1 and 5 mM methylparaben in the presence and absence of 25 mJ/cm 2 ultraviolet B (UV-B) light. Cell proliferation assays showed that 5 mM methylparaben was toxic to M624 cells after 24 hours. Apoptotic signaling pathways were analyzed via isolation of separate cellular compartments and protein analysis via western blot. Upon 5 mM methylparaben treatment, PARP I was cleaved indicating apoptosis, which was mediated by the TNF-α receptor activated in the lipid rafts of the M624 cells. Upon 25 mJ/cm 2 UV-B radiation, PARP II was activated indicating cellular damage, cytochrome c was released from the mitochondria, and caspase-3 was expressed. Upon combinatory treatment with 5 mM methylparaben and 25 mJ/cm 2 UV-B, apoptosis was induced through mitochondrial release of cytochrome c, expression of caspase-3 and cleavage of PARP I, while methylparaben-induced TNF-α receptor activation and UV-B-induced PARP II activation was inhibited., demonstrating that antimicrobial methylparaben in cosmetics can cause damage to cells.
Objective: To describe the technique and outcomes of the closure of maxillary lip defects using a buccal transposition flap and to identify potential routes of vascular supply to the flap.Animals: Five dogs treated clinically and 1 cadaveric dog head. Study design: Short case series and cadaveric study.Methods: A left maxillary labial defect and a buccal transposition flap were created on a cadaver head. Iodinated contrast was injected into the left common carotid artery and computed tomography was performed to assess the vascular supply. Medical records were reviewed for all dogs that underwent tumor excision with maxillary lip resection, reconstructed with a buccal transposition flap. Results:The buccal transposition flap was perfused by branches of the angularis oris artery and superior labial artery. Five dogs were included in this study. All flaps survived. Three dogs developed postoperative complications, including oronasal fistula (n = 2) and partial flap dehiscence (n = 1). The cosmetic and functional outcomes were considered satisfactory in all cases. Conclusion:Buccal transposition flaps for the closure of large maxillary lip defects provided adequate functional and cosmetic outcomes. The buccal transposition flap had vascular contributions from the angularis oris artery and the superior labial artery.
An 11-year-old male castrated domestic shorthair cat was presented for evaluation due to clinical deterioration and potential extrahepatic biliary obstruction (EHBO). Further investigations confirmed EHBO and revealed severe and previously unreported comorbidities. On initial examination, the cat was markedly icteric with a poor body condition score and severe muscle wasting. Serum chemistry and complete blood count showed evidence of cholestasis and anemia. Primary diagnostics and therapeutics targeted these abnormalities. Abdominal ultrasound revealed peritoneal effusion, multifocal mixed echogenic hepatic and splenic foci, small intestinal thickening, cholelithiasis, choledocholithiasis, and common bile duct and pancreatic duct dilation with evidence of obstruction. Peritoneal effusion cytology confirmed septic peritonitis. Hepatic and splenic cytology was consistent with lymphoma. Based on these results, euthanasia was elected by the owners of the animal. Necropsy confirmed the ultrasound diagnoses, septic peritoneal effusion associated with a duodenal perforation, multiorgan lymphoma, and common bile duct carcinoma. Flow cytometry classified the lymphoma as a double-negative phenotype of T-cell lymphoma (CD3+ and CD5+, but CD4- and CD8-) present in the duodenum and liver and suspected in the spleen which has previously not been reported in cats. This case report documents a cat with EHBO caused by multiple disease processes including a novel T-cell lymphoma phenotype, biliary carcinoma, duodenal perforation and septic abdomen, and choleliths, as well as inflammatory hepatobiliary disease.
