Although the artificial skin has used to skin injury, the problem is that the wound surface is not completely contact with the artificial skin. Now we produce a coating material containing human umbilical cord blood-derived mesenchymal stem cells (UCB-MSCs) for skin repair. UCB-MSCs are a good substitute compared with bone marrow-derived MSCs due to the immaturity of newborn cells.Materials and methods: UCB-MSCs were separated, cultured. The immunocytochemical staining and flow cytometric analysis were used to identify the phenotypes of cells. Then the cells were transfected with pEGFP-N2 plasmid. The total number of 54 nude mice were divided into three groups. A full-thickness skin defect was surgically performed on all mice. The skin defect of MSCs/MC group was covered with methylcellurose containing UCB-MSCs. The skin defect of control group 1 was covered with methylcellurose alone and the control group 2, just covered with saline. The wound size was recorded at days 0, 7, 14, 21, 28, 35, 42. Tissue samples were harvested and examined by histological, immunofluorescent staining.Results: UCB-MSCs could different to chondrogenic and osteogenic-like cells, and exhibit mesenchymal stem cell phenotypes.
