The antimicrobial peptide LL-37 belongs to the cathelicidin family and is one of the few human bactericidal peptides with potent antistaphylococcal activity. Staphylococcus aureus is one of the main infection bacteria in orthopedic implant therapy. Biofilm formation after bacterial infection brings more and more severe test for clinical antiinfection treatment.However, there are few studies on LL-37 in S. aureus infection of prosthesis. In this work, addition to research the antibacterial activity and the inhibitory effect on bacterial adhesion of LL-37, an in vitro model of S. aureus biofilm formation on titanium alloy surface was established to observe the inhibitory effect of LL-37.The results showed that LL-37 has a strong antibacterial effect on S. aureus in vitro, and the minimum inhibitory concentration (MIC) is about 0.62 μΜ. Moreover, LL-37 has significant impact on the adhesion of S. aureus when the concentration ≥0.16 μM and significant anti-staphylococcal biofilm effects on static biofilm models at the concentration of 0.31 to 10 μM. Additionally, LL-37 at 5 μM had a significant destructive effect on S. aureus biofilm (P < .05) that formed on the titanium alloy surface.This study further confirmed the role of LL-37 in the process of S. aureus infection, including antimicrobial activities, inhibition of bacterial adhesion, and inhibition of mature biofilm. LL-37 can significantly destroy the stable biofilm structure on the titanium alloy surface in vitro, which may provide a new way for refractory infection caused by S. aureus in titanium alloy prosthesis infection.
BackgroundOsteoarthritis in the lateral compartment of the knee joint is more common in middle-aged and elderly patients. Symptoms include pain, swelling, snapping, stiffness, and joint effusion. At present, there are many ways to treat osteoarthritis in the lateral compartment of the knee joint, and most people adopt conservative treatment. However, for patients with more serious conditions, conservative treatment cannot achieve obvious results, but surgical treatment is required. However, surgical treatment often causes psychological stress on patients, especially elderly patients. Therefore, the study focused on the occurrence and prevention of psychological diseases in patients with lateral knee osteoarthritis after postoperative care programs including psychological treatment.Subjects and MethodsA total of 110 patients with osteoarthritis in the lateral compartment of the knee admitted to a hospital from July 2020 to January 2022 were selected as the research objects. The enrolled patients were randomly divided into two groups, the control group and the observation group, with 55 cases in each group. In the control group, there were 29 males and 26 females, aged 37-79 years, with an average of (60.22 ± 14.15) years old. In the observation group, there were 32 males and 23 females, aged from 35 to 80 years old, with an average of (60.83 ± 14.34) years old. There was no significant difference in general data between the two groups (P > 0.05), which were comparable. The control group was given routine postoperative care, and the observation group was given experimental nursing program including psychotherapy.ResultsFrom the perspective of coping style, there was no significant difference (P > 0.05) in the scores of positive and negative coping styles between the two groups before nursing. After nursing, the positive coping style of the observation group was higher than that of the control group (P < 0.05), and the score of negative coping style was lower than that of the control group (P < 0.05). In terms of anxiety and depression scores, there was no significant difference in SAS (Self-Rating Anxiety Scale) and SDS (Self-Rating Depression Scale) scores between the two groups (P > 0.05). Background: At present, there is no systematic evaluation on whether ultrasonic-guided acupotomy is more effective compared with anatomical guidance in knee osteoarthritis. We conducted a protocol for systematic review and meta-analysis to provide a method for evaluating the effectiveness and safety of acupotomy by ultrasound-guided technique.Methods: An all-round retrieval will be performed in the following electronic journal databases from their inception to October 2022, which comprise PubMed, MEDLINE, EMBASE, Cochrane Library, China National Knowledge Infrastructure, Wanfang data, Chinese Scientific Journals Database, and China biomedical literature database. The following key words were used on combination with Boolean operators AND or OR: "acupotomy," "ultrasound," "knee osteoarthritis." Two authors completed the quality assessment using the Cochrane Collaborations risk of bias tool. The meta-analysis was conducted using Review Manager 5.3 software from the Cochrane Collaboration (London, UK).Results: The findings of this study will be submitted to peer-reviewed journals for publication. Conclusion:This systematic review will provide evidence to judge whether acupotomy by ultrasound-guided technique is effective and safe for knee osteoarthritis.
