Functional traits offer a rich quantitative framework for developing and testing theories in evolutionary biology, ecology and ecosystem science. However, the potential of functional traits to drive theoretical advances and refine models of global change can only be fully realised when species-level information is complete. Here we present the AVONET dataset containing comprehensive functional trait data for all birds, including six ecological variables, 11 continuous morphological traits, and information on range size and location. Raw morphological measurements are presented from 90,020 individuals of 11,009 extant bird species sampled from 181 countries. These data are also summarised as species averages in three taxonomic formats, allowing integration with a global phylogeny, geographical range maps, IUCN Red List data and the eBird citizen science database. The AVONET dataset provides the most detailed picture of continuous trait variation for any major radiation of organisms, offering a global template for testing hypotheses and exploring the evolutionary origins, structure and functioning of biodiversity.
Botrytis cinerea usually produces grayish mycelia and conidia as well as black-colored sclerotia (BS) due to accumulation of melanin. An isolate (XN-1) of B. cinerea producing orange-colored sclerotia (OS) on agar media was obtained from an orange-colored apothecium of an uncultured soil fungus. Whether or not the OS B. cinerea occurs on plants and how they differ from the BS isolates in melanogensis and ecological fitness remained unknown. This study, for the first time, confirmed the presence of the OS B. cinerea in strawberry and tomato plants that were surveyed in Hubei Province of China. Only five OS isolates were obtained from a total of 2,031 isolates surveyed from the two crops. The OS isolate XN-1 was compared and contrasted with the BS isolate B05.10 in sclerotial melanogenesis and ecological fitness. Sclerotial melanogenesis was evident in B05.10 but was deficient in XN-1. The OS were more susceptible to the two mycoparasites Trichoderma koningiopsis and Clonostachys rosea than the BS. The percentage of viable sclerotia after the mycoparasitism study was significantly (P < 0.01) lower in OS (21%) than in BS (48%). This study also reaffirmed the importance of melanization for survival of B. cinerea sclerotia.
Isolation of high quality RNA from ramie (Boehmeria nivea L. Gaud.) is difficult due to its high levels of polyphenols, polysaccharides, pectin, fat, wax and other secondary metabolites. A modified procedure based on guanidinium isothiocyanate for RNA preparation of ramie was developed in this study. High concentrations (5%, v/v) of guanidinium isothiocyanate, PVP-4000, sodium citrate and sodium lauryl sarcosinate and beta-mercaptoethanol were used in the extraction buffer, together with a low pH sodium acetate (pH 4.0) added to improve the RNA quality. The average yield was about 400 microg RNAg(-1) fresh leaves. One SSH library which was induced by ramie anthracnose was constructed by utilizing the RNA extracted through the present method. These results showed that our protocol was applicable for RNA isolation from recalcitrant ramie tissues.
A phage display peptide library targeting the nucleocapsid (N) protein of porcine reproductive and respiratory syndrome virus (PRRSV) strain CH-1a was generated and used for epitope mapping. After 3 rounds of biopanning with the monoclonal antibody (MAb) N3H2 directed against the N protein, 3 positive phages were screened and sequenced. These phages share a consensus sequence, IQTAFNQGA, which corresponds to the amino acid (AA) 79-87 segment of the CH-1a N protein. A small DNA fragment coding for IQTAFNQGA was expressed as a fusion product, and reacted to N3H2 in Western blots and indirect ELISA. Four truncated peptides (IQTAFNQG, IQTAFNQ, QTAFNQGA, and TAFNQGA) expressed as GST fusion products failed to react with N3H2. The sequences around the N3H2-binding site among the N proteins of 57 PRRSV strains were compared. Our results indicate that the IQTAFNQGA motif is highly conserved among North American and European isolates. We concluded that the precisely defined nona-peptide epitope is a novel conserved Linear B cell epitope on the N protein of PRRSV.
Amblyseius tsugawai is a phytoseiid species distributed in Asia. In the present study, laboratory experiments were conducted to investigate its preference to prey stages when feeding on Tetranychus urticae and Bemisia tabaci, to evaluate its development and reproduction, and to estimate its life table parameters on each of the two prey species. Our results indicate that A. tsugawai shows preference to larvae of T. urticae, and to eggs of B. tabaci. When the two preferred stages were provided together, A. tsugawai randomly selected its prey. No significant difference in developmental times of A. tsugawai was observed when reared on T. urticae (6.75 days) and B. tabaci (7.05 days). In contrast, cumulative fecundity of A. tsugawai was only 14.6% when reared on B. tabaci (4.6 eggs/female) than on T. urticae (31.5 eggs/female). Impact of prey for both females and males on reproduction were also observed. Comparing to the treatment when both parents were reared on T. urticae, A. tsugawai fecundity was reduced by ca. 48.2% when the male was reared on B. tabaci, and was reduced by 86.5% and 88.8% when the female or both parents were reared on B. tabaci, respectively. Amblyseius tsugawai is the first indigenous phytoseiid species of China to show a positive intrinsic rate of population increase (0.013) when reared on B. tabaci as the only prey. The potential of A. tsugawai in greenhouse whitefly control is discussed.
