The germinability and viability of mature seeds of five grass (Hordeum spp., Bromus diandrus, Vulpia spp., Avena fatua and Lolium rigidum) and seven broadleaved weed species (Echium spp., Physalis hederifolia, Solanum elaeagnifolium, Raphanus raphanistrum, Marrubium vulgare and Malva parviflora) that were either untreated, ensiled for a minimum of three months, underwent 48 h in sacco digestion in steers or ensiled prior to digestion were tested for germinability and viability. Ensiling and digestion both reduced seed viability, although the extent varied with species. The effect of ensiling was generally greater compared with digestion and differed between years for some species. Ensiling or ensiling plus digestion rendered all seeds of Hordeum spp., B. diandrus, Vulpia spp., A. fatua, Echium spp., P. hederifolia (in one year only), S. elaeagnifolium, R. raphanistrum and M. vulgare nonviable; ensiling and ensiling plus digestion reduced viability of L. rigidum by 74.4% and 92.7% respectively. Viability of M. parviflora displayed the greatest tolerance to damage, with seed viability reduced on average by 31.4%, 27.6% and 27.4% for ensiling, digestion and ensiling plus digestion treatments respectively. These results indicate that ensiling can provide an effective non-chemical weed management option, as a component of an integrated weed management package, for certain weed species responsible for significant crop and pasture production losses in Australian and world temperate and Mediterranean agricultural systems.
A trial was conducted to investigate the survival and digestibility of annual ryegrass (ARG) seed (Lolium rigidum L.) eaten by sheep and cattle. Sheep (n= 8) and cattle (n = 8) were fed a basal diet containing 1:1 lucerne chaff:oaten chaff with (ARG) or without (control) the inclusion of 20% total dry matter of annual ryegrass seed in a changeover design. Intake was restricted to 17 g/kg liveweight. Digestibility of the control diet was lower (P<0.01) for sheep than cattle. Annual ryegrass seed was present (P<0.01) in the faeces of both sheep and cattle within 24 h of first ingestion. Some 10.8 and 32.8% of seed ingested was excreted by sheep and cattle respectively, with 3.9% (sheep) and 11.9% (cattle) remaining germinable. Annual ryegrass seed continued to be excreted by both sheep and cattle up to 5 days after removal from the diet. Dry matter digestibility of the annual ryegrass diet was 53% in cattle.
SUMMARYBerseem clover (Trifolium alexandrinum L.) is an important forage legume and is the primary winter forage crop in Pakistan. There are significant gaps in yield potential among varieties of berseem clover, as well as yields obtained at research stations and on-farm. To address this problem a survey of farmers was undertaken in the districts of Kasur and Okara, Punjab, Pakistan to determine the level of knowledge and understanding of berseem forage cultivation and seed production. The study comprised 44% smallholder (<3 ha), 26% medium (3–5 ha) and 30% large farmers (>5 ha) with average age of 42 years. Most farmers had little or no knowledge of the role of seed quality, inoculation with rhizobium, pollination, fertiliser use, irrigation management and the importance of forage nutritional value in improving livestock productivity. Most farmers (56%) had received no input from the government or private sector to improve forage production, relying instead on traditional knowledge. Knowledge of the importance of land preparation (95%), sowing rate (98%) and insect and pest management (75%) was higher than seed selection and fertilisation. Adoption of improved varieties (3%) and production technologies (14%) was low due to various constraints including ignorance, high cost of inputs, lack of availability of inputs in the market and a perceived high level of financial risk. Almost 100% of the respondents agreed that seed of improved varieties was a pre-requisite for higher forage and seed production as well as essential to start village-based forage seed enterprises.
Several methods are available to extract total lipid and methylate fatty acids from a range of samples including red blood cells (RBC). Fatty acid analysis of human RBC can be undertaken using a two-step extraction and methylation or a combined one-step extraction and methylation procedure. The lipid composition of sheep RBC differs significantly from that of humans and may affect their extraction. The purpose of the current study was to examine the efficiency of extraction of lipid and detection of fatty acids from sheep RBC using a one-step procedure. Fatty acids were analysed using a one-step extraction and methylation procedure using methanol:toluene and acetyl chloride in comparison with a two-step procedure involving extraction of lipid using chloroform:methanol and separate methylation. Concentrations of saturated fatty acids including C16:0 and C18:0 were significantly higher (42.6 and 33.9 % respectively) following extraction using the one-step procedure compared with the two-step procedure. However, concentrations of some polyunsaturated fatty acids, including C20:5n-3 and C22:6n-3 were not significantly different between either procedure. The improved detection of fatty acids may be related to the ability of different solvents to extract different lipid fractions. The differential extraction of lipids and detection of fatty acids from sheep RBC may have important implications in studies examining the effect of dietary treatment on the possible health benefits of fatty acids.
