1. Global pollinator declines as a result of emerging infectious diseases are of major concern. Managed honeybees Apis mellifera are susceptible to numerous parasites and pathogens, many of which appear to be transmissible to sympatric non-Apis taxa. The ectoparasitic mite Varroa destructor is considered to be the most significant threat to honeybees due to its role in vectoring RNA viruses, particularly Deformed wing virus (DWV). Vector transmission of DWV has resulted in the accumulation of high viral loads in honeybees and is often associated with colony death. DWV has two main genotypes, A and B. DWV-A was more prevalent during the initial phase of V. destructor establishment. In recent years, the global prevalence of DWV-B has increased, suggesting that DWV-B is better adapted to vector transmission than DWV-A.2. We aimed to determine the role vector transmission plays in DWV genotype prevalence at a colony level.3. We experimentally increased or decreased the number of V. destructor mites in honeybee colonies, and tracked DWV-A and DWV-B loads over a period of 10 months. 4. Our results show that the two DWV genotypes differ in their response to mite numbers. DWV-A accumulation in honeybees was positively correlated with mite numbers yet DWV-A was largely undetected in the absence of the mite. In contrast, colonies had high loads of DWV-B even when mite numbers were low. DWV-B loads persisted in miticide-treated colonies, indicating that this genotype has a competitive advantage over DWV-A irrespective of mite numbers.
In a wind tunnel we compared the colour preference for western flower thrips to four types of colour plates (clear, white, blue and yellow) applied with two types of glue (diffuse Stikem versus clear D41). Further the results for blue and yellow preference were validated in two greenhouses. In the wind tunnel, we found a clear preference of yellow over blue when a clear glue (D41) was used. However, with a more diffuse (whitish) glue (Stikem) the preference for yellow over blue disappeared, whereby the attraction to yellow decreased (58%) while the attraction to blue increased (65%). In the greenhouses, we found similar effects as in the wind tunnel with a decrease in attraction to yellow (35%) and increase in attraction to blue (32%) for Stikem compared to D41. Light measurements showed an increase of 18% of blue, 21% of violet light, 8% of yellow and 9% of green light reflected on the yellow Stikem trap versus the yellow D41 trap. On blue plates there was only 4% increase of blue light, 8% decrease of yellow light reflected when Stikem glue was used compared to D41 glue. It is not yet clear if the change of light reflection ratio blue/yellow caused by the glue type plays a role in the change of attraction. The reflective properties of glue are so far an unknown factor in colour choice and may explain partially the different results on colour preference. A small review on thrips colour preference is discussed to determine possible other factors of influence on colour choice.
The objective of the present study was to establish whether exposure to temperatures of 55-70 °C results in eradication of the pathogen Clavibacter sepedonicus (Cs) in colonised potato tissue, in order to evaluate the efficiency of composting for disinfection of Cs-infected potato waste. Pre-sprouted potato tubers were inoculated and planted to produce Cs-colonised stem and tuber material. After incubation in temperature-controlled water baths, the infected potato tissue was analysed for the presence of culturable and pathogenic Cs. Additional experiments were performed with Cs-colonised potato stem tissue crushed and deaerated, thus simulating macerated stem tissue in the compost heap. To enable a comparison with bacterial cells that are not enclosed by their natural organic matrix, temperature treatments were applied to non-infected stem tissue crushed and deaerated, and spiked with freshly prepared Cs-suspensions. Cs settled inside colonised potato tissue, as well as Cs present as Cs-suspensions supplemented to potato tissue, was eradicated by exposure to heat, even after a treatment of 1 h at 55 °C, with the exception of one case in which the pathogen present in intact stem material escaped a treatment of 6 h at 60 °C, indicating that incidentally stems may provide niches in which the pathogen is protected against heat.
A potential cause of the dissemination of the potato ring rot bacterium Clavibacter sepedonicus (Cs) is the use of automated seed potato cutters. The present study focuses on the question of whether disinfection practices are sufficient to prevent the transmission of Cs from contaminated machine parts to a new tuber lot. The disinfection efficacy was determined by establishing the culturability of Cs that remained after spray application of sodium-p-toluenesulfochloramide solution on clean and fouled specimens of machinery material that had been provided with an imprint of Cs biofilm. Although conventional spraying, with the authorized concentration of sodium-p-toluenesulfochloramide, of inoculated rubber, PVC and lacquered steel led to a substantial decrease of colony forming units, the treatment was insufficient for complete eradication of Cs. The presence of dirt negatively affected the efficacy of the disinfectant.
