Macadamia is a kind of evergreen nut trees which belong to the Proteaceae family. The two commercial macadamia species, Macadamia integrifolia and M. tetraphylla, are highly prized for their edible kernels. Catherine et al. reported M. integrifolia genome using NGS sequencing technology. However, the lack of a high-quality assembly for M. tetraphylla hinders the progress in biological research and breeding program. In this study, we report a high-quality genome sequence of M. tetraphylla using the Oxford Nanopore Technologies (ONT) technology. We generated an assembly of 750.54 Mb with a contig N50 length of 1.18 Mb, which is close to the size estimated by flow cytometry and k-mer analysis. Repetitive sequence represent 58.57% of the genome sequence, which is strikingly higher compared with M. integrifolia. A total of 31,571 protein-coding genes were annotated with an average length of 6,055 bp, of which 92.59% were functionally annotated. The genome sequence of M. tetraphylla will provide novel insights into the breeding of novel strains and genetic improvement of agronomic traits.

The M. integrifolia genome was recently sequenced, but the genome of M. tetraphylla has to date not been published, which limits the study of biological research and breeding in this species. This study reports a high-quality genome sequence of M. tetraphylla based on the Oxford Nanopore Technologies technology and high-throughput chromosome conformation capture techniques (Hi-C). An assembly of 750.87 Mb with 51.11 Mb N50 length was generated, close to the 740 and 758 Mb size estimates by flow cytometry and k-mer analysis, respectively. Genome annotation indicated that 61.42% of the genome is composed of repetitive sequences and 34.95% is composed of long terminal repeat retrotransposons. Up to 31,571 protein-coding genes were predicted, of which 92.59% were functionally annotated. The average gene length was 6,055 bp. Comparative genome analysis revealed that the gene families associated with defense response, lipid transport, steroid biosynthesis, triglyceride lipase activity, and fatty acid metabolism are expanded in the M. tetraphylla genome. The distribution of fourfold synonymous third-codon transversion showed a recent whole-genome duplication event in M. tetraphylla. Genomic and transcriptomic analysis identified 187 genes encoding 33 crucial oil biosynthesis enzymes, depicting a comprehensive map of macadamia lipid biosynthesis. Besides, the 55 identified WRKY genes exhibited preferential expression in root as compared to that in other tissues. The genome sequence of M. tetraphylla provides novel insights for breeding novel varieties and genetic improvement of agronomic traits.
Elaeagnus conferta Roxb is a kind of fruit known for its oval shape, bright red colour, and sour and sweet taste. In this study, we assembled and characterized the complete chloroplast genome of E. conferta Roxb as a resource for future genetic studies. With a total length of 151,751 bp, the chloroplast genome has a total of 130 genes, consisting of 88 protein-coding genes, 34 tRNA genes, and 8 rRNA genes. Phylogenetic analysis confirmed the position of E. conferta Roxb within the order Rosales.
Improving drought resistance of rubber trees has become a pressing issue with the extension of rubber plantations and the prevalence of seasonal drought. Root system is vital to water and nutrients uptake of all plants, therefore, rootstocks could play decisive roles in drought resistance of grafted rubber trees on a specific scion clone. To investigate the responses of different clone rootstocks and their grafted trees to water stress and find applicable methods for selecting drought resistant rootstocks, seven related parameters and root hydraulic properties of both seeds originated and grafted saplings of PB86, PR107, RRIM600 and GT1 were measured to assess their drought resistance. It was shown that the rootstock drought resistance and root hydraulic conductance may improve the drought resistance of the grafted rubber trees. Among the four clone rootstocks, GT1, which demonstrated more resistant to drought and higher root hydraulic conductance, was comparatively resistant to drought both for the seed propagation seedlings and grafted saplings. In addition, studies on the grafted saplings with different root hydraulic conductance further validated the possibility of selecting drought resistant rootstocks on the basis of rootstock hydraulic conductance using a high-pressure flow meter.
The chloroplast (cp) genome sequence of Persea americana Mill. has been characterized from Illumina pair-end sequencing. The complete cp genome was 152,732 bp in length, containing a large single copy region (LSC) of 84,632 bp and a small single copy region (SSC) of 18,346 bp, which were separated by a pair of 24,877 bp inverted repeat regions (IRs). The genome contained 126 genes, including 86 protein coding genes, 32 tRNA genes, and eight rRNA genes. Further, phylogenetic analysis of 10 representative sequences within the order Ebenales suggests that Persea americana Mill. is closely related to the species in family Sapotaceae.
