Understanding how genetic variation contributes to phenotypic differences is a fundamental question in biology. Combining high-throughput gene function assays with mechanistic models of the impact of genetic variants is a promising alternative to genome-wide association studies. Here we have assembled a large panel of 696 Escherichia coli strains, which we have genotyped and measured their phenotypic profile across 214 growth conditions. We integrated variant effect predictors to derive gene-level probabilities of loss of function for every gene across all strains. Finally, we combined these probabilities with information on conditional gene essentiality in the reference K-12 strain to compute the growth defects of each strain. Not only could we reliably predict these defects in up to 38% of tested conditions, but we could also directly identify the causal variants that were validated through complementation assays. Our work demonstrates the power of forward predictive models and the possibility of precision genetic interventions.
O aumento do consumo de produtos de origem natural tem ocasionado problemas de saúde pública devido ao risco da contaminação fúngica e a possível presença de micotoxinas. Objetivou-se, neste estudo, identificar as espécies fúngicas com potencial micotoxigênico e avaliar o efeito da irradiação gama (60Co) na destruição da microbiota fúngica natural de cinco plantas medicinais: Alcachofra (Cynara scolymus L.), Boldo (Peumusboldus Molina), Camomila (Matricaria recutita L.), Chapéu de couro (Echinodorusgrandiflorus Micheli) e Sene (Cassia acutifolia Delile). A quantificação de fungos filamentosos e leveduras foi efetuada pela Técnica de Diluição Seriada em meio DRBC. Em Camomila foram identificados 8 isolados de Aspergillus flavus, sendo 2 (25%) produtores de aflatoxina B1 e B2 e 5 isolados de Aspergillus ochraceus, sendo 2 (40%) produtores de ocratoxina A. Em Alcachofra foi identificado 1 isolado de Aspergillus ostianus produtor de ocratoxina A. Observou-se redução total da contagem de fungos em Boldo a partir de 3 kGy e em Chapéu de couro e Sene a partir de 5 kGy. Em Alcachofra, a contagem inicial de 5,0 x 10(6) UFC/g foi reduzida para 3,5 x 10² UFC com dose de 10 kGy. Nessa mesma dose a contagem fúngica em Camomila foi reduzida de 3,0 x 10(5) UFC/g para 2,2 x 10³ UFC/g.
The viability of the yeast Rhodotorula rubra, isolated from liquid samples of gold-mine effluents, was not affected by the presence of 11.52 mM cyanide. The yeast was able to utilize ammonia, generated from abiotic cyanide degradation in the presence of reducing sugars, in aerobic culture at pH 9.0. These physiological characteristics encourage studies with mixed cultures of cyanide-degrading organisms, using this yeast as an assimilator of ammonia.
