Cobalt, as a trace element, is essential for rumen microorganisms for the formation of vitamin B12. In the metabolism of mammals, vitamin B12 is an essential part of two enzymatic systems involved in multiple metabolic reactions, such as in the metabolism of carbohydrates, lipids, some amino acids and DNA. Adenosylcobalamin and methylcobalamin are coenzymes of methylmalonyl coenzyme A (CoA) mutase and methionine synthetase and are essential for obtaining energy through ruminal metabolism. Signs of cobalt deficiency range from hyporexia, reduced growth and weight loss to liver steatosis, anemia, impaired immune function, impaired reproductive function and even death. Cobalt status in ruminant animals can be assessed by direct measurement of blood or tissue concentrations of cobalt or vitamin B12, as well as the level of methylmalonic acid, homocysteine or transcobalamin in blood; methylmalonic acid in urine; some variables hematological; food consumption or growth of animals. In general, it is assumed that the requirement for cobalt (Co) is expressed around 0.11 ppm (mg/kg) in the dry matter (DM) diet; current recommendations seem to advise increasing Co supplementation and placing it around 0.20 mg Co/kg DM. Although there is no unanimous criterion about milk production, fattening or reproductive rates in response to increased supplementation with Co, in some investigations, when the total Co of the diet was approximately 1 to 1.3 ppm (mg/kg), maximum responses were observed in the milk production.
Context.— Food and Drug Administration–approved TRK inhibitors with impressive overall response rates are now available for patients with multiple cancer types that harbor NTRK rearrangements, yet the identification of NTRK fusions remains a difficult challenge. These alterations are highly recurrent in extremely rare malignancies or can be detected in exceedingly small subsets of common tumor types. A 2-step approach has been proposed, involving a screening by immunohistochemistry (IHC) followed by a confirmatory method (fluorescence in situ hybridization, reverse transcriptase–polymerase chain reaction, or next-generation sequencing) in cases expressing the protein. However, there is no interpretation guide for any of the available IHC clones. Objective.— To provide a pragmatic update on the use of pan-TRK IHC. Selected examples of the different IHC staining patterns across multiple histologies are shown. Data Sources.— Primary literature review with PubMed, combined with personal diagnostic and research experience. Conclusions.— In-depth knowledge of pan-TRK IHC will help pathologists implement a rational approach to the detection of NTRK fusions in human malignancies.
This study aimed to evaluate the postrelease survival and spatial distribution of farm-bred red-legged partridges (Alectoris rufa) that were subject to a prerelease training program based on exemplary behavior and alarm calls given by adults that acted as experienced demonstrators in simulated predator encounters (wooden raptor models and humans). Two groups of partridges were released in this study: trained (intensively reared birds accompanied by tutors) and control (chicks reared without tutors). Releases were conducted in the fall and winter-spring during 2 consecutive years using a total of 44 trained and 40 control radio-tagged partridges. Trained partridges showed statistically higher mean values of survival (72.7 d), home range (13.04 ha), and dispersion (549.58 m) compared with nontrained partridges, with most nontrained birds failing to survive more than 2 wk after release. Trained adult partridges showed the best survival results (105.2 d). Taking all birds into account, causes of death were attributed to terrestrial predators (45%), raptors (18.7%), hunting (11.3%), and unknown causes (25%). Although values of variables reported here were lower than those reported in wild counterparts, survival time and spatial behavior of trained birds were close to that of previous data of parent-reared partridges and higher than that of farm-bred birds. This study aimed to confirm the potential of prerelease training techniques in present-day rearing systems. Farm-bred game birds, which normally suffer from high predation rates after release, could highly benefit from the use of cost-effective training techniques based on learning from experienced adults.
