Formalin is a widely used fixative but there is potential public health risks to exposure. Besides, alcoholic fixation is advantageous over formalin fixation because of faster fixation, optimal preservation and safer workplace environment. Following fixation by EMA and 10% neutral buffered formalin (NBF), we analyzed the tissue morphology, antigenic stability, DNA and RNA quantity with quality (OD value). The findings of EMA fixing on both the tissue morphology and molecular characterization, were satisfactory. Specially, EMA was faster in penetration of tissues than NBF, fixed ideally as early as 8 h of fixation whereas improper fixation was evident for NBF. In Hematoxylin and Eosin (H & E) staining, better cellular details with stronger affinity for staining were observed. In immunohistochemistry, better antigenic stability was reported for EMA-fixed tissues. The nucleic acid analysis revealed that total genomic DNA and RNA yield from EMA fixed tissues were significantly higher ( P < 0.05 ) with superior quality than NBF fixed tissues. Our results suggest that EMA could be a potential alternative to NBF for fixation and preservation of tissues. These data provide new insights into an option for a safer working environment to support study and research.
Newcastle disease (ND) is a highly contagious viral disease. This study was conducted to determine the antibody titer in Japanese quails following vaccination against ND and feed-supplementation with vitamin-C, probiotics and antibiotic-growth-promoters. Forty Japanese quails were equally grouped in four groups (A1 to A4) and were vaccinated by LaSota, B1, F and VG/GA strains, respectively, at the age of 10 days. Another forty Japenese quails were equally grouped in four groups (B1 to B4), were vaccinated with the vaccines of the previous groups that showed a higher effect, and were fed with mesh-feed (control-B1), supplemented with vitamin-C (B2), probiotics (B3), and antibiotic growth promoter (B4). Serum samples were obtained on 2, 4, 6, 8, 10 and 12 weeks of age and were subjected to a hemagglutination inhibition test. The mean antibody titer (Log2) had no significant difference between groups A1-A4. The highest titers were recorded in A4 (6.20±0.37) and A3 (6.00±0.71). The mean titer was significantly different among the groups at 4 weeks, but insignificant in the other weeks of the supplemented groups. A higher mean titer was found between 8 and 10 weeks in group B2 (7.40±0.60) and B3 (7.20±0.58). The relative weights (gm/kg) of the spleen, thymus and bursa were significantly higher in group B3 (1.947±0.109, 5.951±0.312 and 2.624±0.083, respectively). They had no significant correlation with antibody titer at 12 weeks. In conclusion, vitamin-C and probiotic supplemented feeds can boost up the antibody titer against ND in Japanese quail when given during VG/GA strain vaccination.
Peste des petits ruminants (PPR), a fatal viral disease of goats causes high mortality and large economic losses, and is considered as one of the major constrains of goat farming worldwide.This study was undertaken to determine the prevalence, alteration of vital signs and effective therapeutics of PPR affected goats inBeraupzilla ofPabna district, Bangladeshduring the period of November 2014 to April 2015. A total number of 465 diseased goats were clinically examined of which 253 (54.41%) were found to be affected with PPR. The highest prevalence (72.27%) was found in Black Bengal goats whereas 35.04% and 27.78% prevalence were found in Jamunapari and Cross breed goats respectively. In case of Black Bengal goats51.78% affected goats were aged between 0-7months. In case ofJamunapari 51% affected goats aged between 8-14 months. Female goats were more susceptible (65%) as compared to males. About 60%Black Bengal goats had a body temperature range of 104-105.9 ?F. Over half of the study populations were in normal range of respiration rate.65%cross breeds heart rate was recorded in 91-110/minute range group that was higher in comparison to other breeds.The response totreatment following parenteral (I/M) administration of Sulphadimidine was higher (58%) than parenteral (I/M) OxytetracyclineHCl (50%) and Gentamycin Sulphate (46%) administration. All these findings revealed that, the prevalence of PPR depends on specific breed and age groupsand Sulphadimidine is the most effective drug choice in PPR treatment.Progressive Agriculture 28 (2): 114-119, 2017
Turkey bird is one of the popular poultry species which is reared primarily for meat production and considered as one of the major sources of animal protein. With such importance of this species, this study was designed to investigate the gross and histomorphology of the ovary and oviduct with especial emphasis on sperm-host glands of the turkey hen involving ten mature female turkeys (Meleagris gallopavo). The present study highlighted the distribution pattern of sperm-host glands (SHGs) in the oviduct of turkey hen that has a potential role in producing a fertile egg in poultry industries. The oviduct of turkey consists of the infundibulum, magnum, isthmus, uterus, and vagina which are sole distributors for making nutrition enriched egg. The tissue samples were collected from the ovary, different segments of the oviduct and especially uterovaginal junction (UVJ) and infundiomagnal junction of the oviduct. The ovaries and the oviducts were dissected and fixed in Bouins solution and processed for a light microscopic study. Histologically, the left ovary of turkey consisted of an outer cortex and inner medulla, with different stages of follicles. In all areas of the oviduct except the infundibulum and vagina, the tunica mucosa epithelium was lined with ciliated pseudo stratified columnar epithelium, and the lamina propria-submucosa contained branched tubular glands. Sperm-storage tubules were observed in the uterovaginal junction and infundibulo-magnum junction. These tubules were mostly branched, slightly coiled and extended into the lamina propria from the bases of the mucosal folds. These glands had proximal and distal parts; the proximal part was lined by pseudostratified columnar epithelium and distal part by non-ciliated simple columnar epithelium. The number of sperm host glands was more at uterovaginal junction than infundibulomagnal junction. The sperm-host glands might play a functional role in the storage and release of spermatozoa from the SHGs in response to oviposition or ovulation. The results would help poultry scientists and farmers in developing effective disease control and growth strategies.
