PENENTUAN TOTAL FENOLIK DAN UJI AKTIVITAS ANTIOKSIDAN PADA BIJI DAN KULIT BUAH PINANG YAKI (Areca vestiaria Giseke)ABSTRAK Telah dilakukan penelitian untuk menentukan kandungan total fenolik dan menguji aktivitas antioksidan pada biji dan kulit buah pinang yaki. Ekstrak biji dan kulit buah pinang yaki diperoleh dengan maserasi tanpa sokletasi dan maserasi setelah sokletasi. Total fenolik diuji dengan metode Folin-Ciocalteu dan uji aktivitas antioksidan dengan metode DPPH. Hasil penelitian yang diperoleh, kandungan total fenolik Maserasi Biji Setelah Sokletasi, Maserasi Kulit Setelah Sokletasi berturut-turut memiliki nilai sebagai berikut : 85,92 mg/kg, 3,16 mg/kg. sedangkan aktivitas antioksidan Maserasi Biji Setelah Sokletasi, Maserasi Kulit Setelah Sokletasi berturut-turut memiliki nilai sebagai berikut : 88,16%, 54,11%. The samples of the seed and skin were prepared separetely by maceration and soxhlet extraction followed by maceration. Folin-Ciocalteu method was used to measure the total phenolic compounds of while DPPH (1,1-diphenyl-2-pikrilhidrazil) method was used to measure the antioxidant activity. The total phenolic compounds of the seed by soxhlet extraction followed by maceration, skin by soxhlet extraction followed by maceration and were 85,92 mg/kg, 3,16 mg/kg respectively. The antioxidant activity of the seed by soxhlet extraction followed by maceration, skin by soxhlet extraction followed by maceration and were 88,16%, 54,11% respectively. ABSTRAK Penelitian ini dilakukan untuk mengetahui senyawa aktif yang terkandung melalui pengujian fitokimia dan aktivitas antioksidan pada tumbuhan obat di pulau Tidore. Uji aktivitas antioksidan menggunakan metode DPPH dilanjutkan dengan perhitungan Inhibition Concentration 50% (IC50). Hasil yang diperoleh adalah biji buah mojoi terkandung senyawa alkaloid dan saponin, buah coro terkandung alkaloid, flavonoid, dan saponin, pada daun ofo terkandung alkaloid, tanin, flavonoid, steroid, dan saponin dan pada rimpang kuso mafola terkandung alkaloid, tanin, flavonoid, steroid, dan saponin. Nilai IC50 sebagai berikut biji rimpang kuso mafola 37,30 ppm, buah coro 250,17 ppm, daun ofo 976,10 ppm dan buah mojoi 1001, 07 ppm. Kata kunci :Tumbuhan obat, DPPH, antioksidan dan uji fitokimia. PHYTOCHEMICALS TEST AND DETERMINATION Inhibition Concentration 50% ON SOME MEDICINAL PLANTS IN THE TIDORE ISLAND ABSTRACT This study was conducted to determine the active compounds contained in an assessment of phytochemical and antioxidant activity in the medicinal plants of Tidore island. The test antioxidant activity was used DPPH method. In the test results to the phytochemical , that mojoi fruit seeds contained alkaloids and saponins, fruit coro (alkaloids, flavonoids, and saponins), ofo leaves (alkaloids, tannins, flavonoids, steroids, and saponins) and ethanol extract of rhizome kusomafola (alkaloids, tannins, flavonoids, steroids, and saponins. In calculation of IC50 values for rhizome kusomafola 37.30 ppm, 250.17 ppm coro fruit, 976.10 ppm ofo leaf extract, and fruit seed extract mojoi 1001.07 ppm. Penelitian ini bertujuan untuk menentukan total phenolic content (TPC), total flavonoid content (TFC), condensed tannin content (CTC) dan nilai toksisitas dari daun leleng merah. Daun leleng merah diekstraksi dengan metode maserasi dan dipartisi menggunakan pelarut n-heksana, etil asetat, dan air. Penentuan nilai TPC menggunakan metode Folin-Ciocalteu. Penentuan nilai TFC menggunakan metode AlCl3. Penentuan nilai CTC menggunakan metode vanillin-HCl. Penentuan nilai toksisitas menggunakan metode BSLT. Nilai TPC yang diperoleh pada fraksi n-heksana (FH), fraksi air (FA), dan fraksi etil asetat (FEA) daun Leleng Merah berturut-turut, adalah: 20,48; 31,7; dan 37,57 (mg/g). Nilai TFC diperoleh pada FH, FA, dan FEA berturut-turut, adalah: 0,64; 0,99; dan 1,27 (mg/g). Nilai CTC diperoleh pada FH, FA, dan FEA berturut-turut, adalah: 0,56; 2,17; dan 6,52 (mg/g). Nilai toksisitas diperoleh pada FEA, FA, FH berturut-turut, adalah: 21,93; 100,7; dan 117,22 (mg/L). FEA memiliki nilai toksisitas dan kandungan fitokimia tertinggi.
