The SARS-CoV-2 virus is a disease that has mild to severe effects on patients, which can even lead to death. One of the enzymes that act as DNA replication is the main protease, which becomes the main target in the inhibition of the SARS-CoV-2 virus. In finding effective drugs against this virus, Ocimum basilicum is a potential herbal plant because it has been tested to have high phytochemical content and bioactivity. Apigenin-7-glucuronide, dihydrokaempferol-3-glucoside, and aesculetin are polyphenolic compounds found in Ocimum basilicum. Purpose: The purpose of this study was to analyze the mechanism of inhibition of the three polyphenolic compounds in Ocimum basilicum against the main protease and to predict pharmacokinetic activity and the drug-likeness of a compound using the Lipinski Rule of Five. Patients and Methods: The method used is to predict the molecular docking inhibition mechanism using Autodock 4.0 tools and use pkcsm and protox online web server to analyze ADMET and Drug-likeness. Results:The binding affinity for apigenin-7-glucuronide was −8.77 Kcal/mol, dihydrokaempferol-3-glucoside was −8.96 Kcal/mol, and aesculetin was −5.79 Kcal/mol. Then, the inhibition constant values were 375.81 nM, 270.09 nM, and 57.11 µM, respectively. Apigenin-7-glucuronide and dihydrokaempferol-3-glucoside bind to the main protease enzymes on the active sites of CYS145 and HIS41, while aesculetin only binds to the active sites of CYS145. On ADMET analysis, these three compounds met the predicted pharmacokinetic parameters, although there are some specific parameters that must be considered especially for aesculetin compounds. Meanwhile, on drug-likeness analysis, apigenin-7-glucuronide and dihydrokaempferol-3-glucoside compounds have one violation and aesculetin have no violation. Conclusion: Based on the data obtained, Apigenin-7-glucuronide and dihydrokaempferol-3-glucoside are compounds that have more potential to have an antiviral effect on the main protease enzyme than aesculetin. Based on pharmacokinetic parameters and druglikeness, three compounds can be used as lead compounds for further research.
It is important for everyone, regardless of age, to take into account their dental and oral health to enhance their overall well-being. Dentists have the responsibility of educating the public on proper dental and oral care in order to attain the best possible dental and oral health. Nevertheless, there are still numerous individuals who do not prioritize their dental and oral health. With the aim of fulfilling the Tridharma mission in Community Service, the Faculty of Dentistry at Trisakti University has implemented Public Health Services tha provide counseling on oral and dental health for families, staring with the elderly, general dental health, childrens dental health, and safe medications for families. Through the use of a questionnaire, an increase in knowledge was observed among individuals of different age groups regarding oral health care.
<p><strong><em>Background: </em></strong><em>Pulp diseases can be affected by bacterial invasion such as Porphyromonas endodontalis.<strong> </strong>This bacterial invasion can lead to pulp diseases which include irreversible pulpitis and necrotic pulp.<strong> </strong></em><strong><em>Purpose</em></strong><em>: </em><em>To determine Porphyromonas endodontalis difference between teeth with irreversible pulpitis and necrotic pulp. </em><strong><em>Method: </em></strong><em>Samples were taken from root canals fluid of 18 teeth, 9 teeth with irreversible pulpitis, 9 teeth with necrotic pulp, using paper point. The paper point was placed in sterile Eppendorf tube, then DNA were extracted using Heat-Shock DNA Extraction method. Real-time PCR was used to collect the data. The conversion of microbiological data were used standard curve of Porphyromonas endodontalis.<strong> </strong></em><strong><em>Result: </em></strong><em>The average of Real-time PCR on irreversible pulpitis was </em><em>6.9 x 10<sup>-12</sup></em><em> </em><em>ng/mL</em><em> and on necrotic pulp was </em><em>1.3 x 10<sup>-12</sup> ng/mL</em><em>. However, t independent test showed there was no significant difference of Porphyromonas endodontalis between teeth with irreversible pulpitis and necrotic pulp (p>0,05) </em><strong><em>Conclusion: </em></strong><em>There was no difference in the total number of Porphyromonas endodontalis between teeth with irreversible pulpitis and necrotic pulp.<strong> </strong></em></p>
<p>Prevalensi maloklusi di Indonesia masih sangat tinggi yaitu sekitar 80% dari jumlah penduduk dan merupakan salah satu masalah kesehatan gigi dan mulut yang cukup besar. Hal ini ditambah dengan tingkat kesadaran perawatan gigi yang masih rendah dan masyarakat belum menyadari pentingnya pemeliharaan kesehatan gigi dan mulut. Maloklusi sering ditemukan pada anak-anak masa periode gigi campur, yaitu saat gigi sulung dan gigi permanen secara besamaan berada di dalam rongga mulut yang di mulai dari sekitar usia 6 tahun hingga usia 12 tahun. Deteksi kelainan maloklusi dapat dimulai pada usia 7 tahun. Di luar negeri hal ini merupakan bagian dari kampanye yang dilakukan oleh organisasi Orthodontist Amerika Serikat. Tujuan kegiatan ini adalah membiasakan anak-anak untuk menyikat gigi sedini mungkin dan memberikan pengetahuan tentang cara sikat gigi yang baik dan benar dalam rangka pencegahan maloklusi pada seluruh orangtua dan guru anak TK AlHidayah 2 Kelapa Gading Pelaksanaan program merawat gigi pada anak serta mengenali kelainan maloklusi meliputi tahapan proses ceramah, dan tanya jawab melalu media Webinar, Zoom. Materi meliputi alasan pembatasan kunjungan ke dokter gigi di masa pandemi, penyebab terbanyak kunjungan ke dokter gigi (sakit karena gigi berlubang), proses terjadinya, cara pencegahannya, dan tips deteksi dini dalam rangka pencegahan maloklusi. Kegiatan diikuti oleh 34 peserta, keberhasilan kegiatan ini dilihat dari peningkatan pengetahuan. Skor hasil dari pretest dan post test selanjutnya akan dibandingkan dan diketahui apakah ada perubahan pengetahuan sebelum dan sesudah pelatihan. Hasil penyuluhan pelatihan menjaga kesehatan gigi dan mulut bagi orang tua dan guru di TK Alhidayah 2, Kelapa Gading yang dilakukan melalui media webinar menunjukkan hasil yang memuaskan. Terdapat peningkatan nilai Post-test para peserta penyuluhan sebesar 10,5%.</p>
