Reciprocating and low-speed sliding contacts can experience increased friction because of solid boundary interactions. Use of surface texturing has been shown to mitigate undue boundary friction and improve energy efficiency. A combined numerical and experimental investigation is presented to ascertain the beneficial effect of pressure perturbation caused by micro-hydrodynamics of entrapped reservoirs of lubricant in cavities of textured forms as well as improved microwedge flow. The results show good agreement between numerical predictions and experimental measurements using a precision sliding rig with a floating bed-plate. Results show that the texture pattern and distribution can be optimised for given conditions, dependent on the intended application under laboratory conditions. The translation of the same into practical in-field applications must be carried out in conjunction with the cost of fabrication and perceived economic gain. This means that near optimal conditions may suffice for most application areas and in practice lesser benefits may accrue than that obtained under ideal laboratory conditions.
The prevalence of BRCA1 and BRCA2 mutations among breast cancer patients in Peru has not yet been explored. We enrolled 266 women with breast cancer from a National cancer hospital in Lima, Peru, unselected for age or family history. DNA was screened with a panel of 114 recurrent Hispanic BRCA mutations (HISPANEL). Among the 266 cases, thirteen deleterious mutations were identified (eleven in BRCA1 and two in BRCA2), representing 5% of the total. The average age of breast cancer in the mutation-positive cases was 44 years. BRCA1 185delAG represented seven of the eleven mutations in BRCA1. Other mutations detected in BRCA1 included: two 2080delA, one 943ins10, and one 3878delTA. The BRCA2 3036del4 mutation was seen in two patients. Given the relatively low cost of the HISPANEL test, one should consider offering this test to all Peruvian women with breast or ovarian cancer.
AIMTo determine influence of neoadjuvant-chemotherapy (NAC) over tumor-infiltrating-lymphocytes (TIL) in triple-negative-breast-cancer (TNBC).METHODSTILs were evaluated in 98 TNBC cases who came to Instituto Nacional de Enfermedades Neoplasicas from 2005 to 2010. Immunohistochemistry staining for CD3, CD4, CD8 and FOXP3 was performed in tissue microarrays (TMA) sections. Evaluation of H/E in full-face and immunohistochemistry in TMA sections was performed in pre and post-NAC samples. STATA software was used and P value < 0.05 was considered statistically significant.RESULTSHigher TIL evaluated in full-face sections from pre-NAC tumors was associated to pathologic-complete-response (pCR) (P = 0.0251) and outcome (P = 0.0334). TIL evaluated in TMA sections showed low level of agreement with full-face sections (ICC = 0.017-0.20) and was not associated to pCR or outcome. TIL in post-NAC samples were not associated to response or outcome. Post-NAC lesions with pCR had similar TIL levels than those without pCR (P = 0.6331). NAC produced a TIL decrease in full-face sections (P < 0.0001). Percentage of TIL subpopulations was correlated with their absolute counts. Higher counts of CD3, CD4, CD8 and FOXP3 in pre-NAC samples had longer disease-free-survival (DFS). Higher counts of CD3 in pre-NAC samples had longer overall-survival. Higher ratio of CD8/CD4 counts in pre-NAC was associated with pCR. Higher ratio of CD4/FOXP3 counts in pre-NAC was associated with longer DFS. Higher counts of CD4 in post-NAC samples were associated with pCR.CONCLUSIONTIL in pre-NAC full-face sections in TNBC are correlated to longer survival. TIL in full-face differ from TMA sections, absolute count and percentage analysis of TIL subpopulation closely related.
a b s t r a c tAn analytical/numerical methodology is presented to calculate the radiated noise due to internal combustion engine piston impacts on the cylinder liner through a film of lubricant. Both quasi-static and transient dynamic analyses coupled with impact elastohydrodynamics are reported. The local impact impedance is calculated, as well as the transferred energy onto the cylinder liner. The simulations are verified against experimental results for different engine operating conditions and for noise levels calculated in the vicinity of the engine block. Continuous wavelet signal processing is performed to identify the occurrence of piston slap noise events and their spectral content, showing good conformance between the predictions and experimentally acquired signals.
