Abstract. Prolactin in chicken (cPRL) is a polypeptide hormone that plays a crucial role in incubation and brooding behavior. This hormone is coded by a gene located at chromosome 2. The objective of this study was to investigate 24-bp indel polymorphism in promoter prolactin gene of Papua local chickens. A number of 60 DNA samples came from 60 Papua local chickens (25 males and 35 females) were used in this study. PCR (Polymerase Chain Reaction) method with a pair of specific primer was applied and a specific DNA fragment with 130 and/or 154 bp contained 24-bp indel was successfully amplified. Results of electrophoresis run on 1.5 % agarose gel showed three genotypes: II, ID and DD with the frequency of 6,7%, 48.3% and 45% respectively. The frequency of I allele and D allele was 0.69 and 0.31, respectively. Results indicated that the I allele frequency was medium in Papua local chickens population. Results therefore suggest that incubation behavior in hen population of Papua local chickens is easy to eliminate by increasing the I allele frequency in the population.Key words: 24-bp Indel, Papua local chicken, polymorphism, promoter prolactin gene.
PE goat is a small ruminant crossed between Etawah goat and bean goat, which introduced in Manokwari since 2007. Livestock can well produce depend on environment comfortable temperature in the area of tropical temperature on the day is radiated in high sun light. If the goat is expose to the blazing sun, it supposed experiencing heat stress and caused to the physiological and haematological condition. The aim of this study is to know the physiological and haematological response of PE goat to heat stress. This design of research used is Split Plot (RPT). 8 goats will be a sample which devide in two groups, of unexposed groups and Groups exposed to sunlight. Each group consists of young males, young female, adult male, adult female. The variables observed included physiological aspects (heart rate, rectal temperature, respiratory rate) and haematological aspects (hemoglobin level, hematocrit value, erythrocyte count, leukocyte count). The results showed that interaction between goats and heat stress status did not influence physiological aspect variable (heart rate, rectal temperature, respiratory rate), but very significant effect on some hematological aspect variable such as hemoglobin and erythrocytes. Penelitian bertujuan untuk mengetahui respon produksi rumput Brachiaria decumbens akibat perlakuanpemberian dekamon sebagai zat pengatur tumbuh pada berbagai frekuensi penyemprotan pupuk daungandasil. Penelitian berlangsung selama 4,5 bulan, bertempat di Balai Pembibitan Ternak dan HijauanMakanan Ternak (BPT-HMT) Dinas Peternakan dan Kesehatan Hewan Provinsi Papua Barat, KampungSumber Boga, Distrik Masni, Kabupaten Manokwari dan Laboratorium Nutrisi dan Makanan Ternak FapetUNIPA Manokwari. Rancangan eksperimen yang digunakan adalah rancangan acak lengkap (RAL) dengandua perlakuan. Perlakuan dekamon (D) dengan 6 taraf yaitu D0 (tanpa pemberian), D1= 0,4 ml/250 ml air,D2= 0,5 ml/250 ml air, D3=0,6 ml/250 ml air, D4=0,7 ml/250 ml air dan D5= 0,8 ml/250 ml air. Masingmasingtaraf perlakuan diulang 5 kali. Selanjutnya menggunakan 3 petak percobaan dengan ukuran 2,4 ×2 m2 /petak untuk perlakuan frekuensi penyemprotan pupuk daun dengan Gandasil D, masing-masing petak I (S0) untuk tanpa penyemprotan, petak II (S1) untuk penyemprotan 1 kali/minggu dan petak III (S2) untuk enyemprotan 2 kali/minggu. Data dianalisis secara statistik dengan analisis ragam dan perbedaan antara taraf perlakuan dianalisis dengan uji t. Hasil penelitian menunjukkan bahwa pemberian dekamon sebagai zat pengatur tumbuh tidak berpengaruh terhadap produksi rumput bede, namun frekuensi penyemprotan pupuk daun 2 kali/minggu berpengaruh sangat nyata terhadap jumlah tunas rumput bede pada dosis dekamon 0 ml dan 0,6 ml. Dosis dekamon yang tepat bagi produksi rumput bede adalah tanpa dekamon dengan frekuensi penyemprotan 2 kali/minggu. Kata kunci: Brachiaria decumbens, produksi, dekamon, pupuk daun
Weaning weights and yearling weights are economical characteristics of production to be used as selection criteria in animal. The study aimed to evaluate selection response of weaning weight and yearling weights directly and to evaluate selection response of yearling weights indirectly through weaning weights. A total of 190 data from 23 sires and 133 dams were used to estimate the heritability of weaning weights. Data to estimate heritability of yearling weights were about 231 data from 20 sires and 101 dams. Data to estimate genetic correlation between weaning weights and yearling weights were about 194 data from 26 sires and 125 dams. The heritability was estimated by using a variance analysis of paternal half-sib correlation. The results showed that heritability of weaning weights and yearling weights were 0.63±0.291 and 0.68±0.342, respectively. The genetic correlation between weaning weights and yearling weights was 0.78. The fewer proportions of animal that selection for, the higher selection response for weaning weights and yearling weights both directly and indirectly selection.
Penelitian ini bertujuan untuk mengidentifikasi genotip dari lokus growth hormone-AluI (GH-AluI) pada populasi sapi Bali di Pleihari, Kalimantan Selatan. Sampel darah diambil dari 46 ekor sapi Bali (25 jantan dan 21 betina) di lokasi Pleihari, Kalimantan Selatan. Identifikasi genotip dari lokus ini dilakukan dengan mendigesti fragmen DNA berukuran 221 bp yang merentang dari daerah intron 4 (49 bp) hingga exon 5 (162 bp) menggunakan enzim restriksi AluI. Hasil penelitian menunjukkan bahwa lokus GH-AluI pada populasi sapi Bali di Pleihari, Kalimantan Selatan hanya memiliki satu macam alel, yaitu alel L, dan tidak ditemukan alel V atau alel lainnya pada lokus ini. Disuimpulkan bahwa lokus GH-AluI pada populasi sapi Bali di Pleihari, Kalimantan Selatan bersifat monomorfik.
