Zika virus (ZIKV) is an emerging mosquitoborne flavivirus circulating in Asia and Africa. In 2013, a large outbreak was reported on the archipelago of French Polynesia. In this study, we report the detection and molecular characterization of Zika virus for the first time in Chile from an outbreak among the inhabitants of Easter Island. A total of 89 samples from patients suspected of having ZIKV infection were collected between the period from January to May, 2014. Molecular diagnosis of the virus was performed by RT-PCR followed by the sequencing of the region containing the NS5 gene. A comparison of the viral nucleic acid sequence with those of other strains of ZIKA virus was performed using the MEGA software. Fifty-one samples were found positive for ZIKV by RT-PCR analysis. Further analysis of the NS5 gene revealed that the ZIKV strains identified in Easter Island were most closely related to those found in French Polynesia (99.8 to 99.9 % nt and 100 % aa sequence identity). These results strongly suggest that the transmission pathway leading to the introduction of Zika virus on Easter Island has its origin in French Polynesia.Keywords Zika virus Á Easter Island Á Outbreak Á South Pacific Á ZIKV Zika virus (ZIKV) is a mosquito-borne virus belonging to the family Flaviviridae and the genus Flavivirus. The Flaviviruses also include yellow fever, dengue, St. Louis encephalitis, West Nile, and Japanese encephalitis viruses [6]. ZIKV was first isolated from the blood of a sentinel rhesus monkey inhabiting the Zika Forest in Uganda [6]. Later on, ZIKV was isolated from a mosquito of the species Aedes africanus, obtained from the same locality of the Zika Forest. ZIKV has been isolated from several species of Aedes mosquito, notably, A. aegypti and A. albopictus [10]. A. aegypti is widely distributed in tropical and subtropical regions of the world [6,11,12].Clinically diagnosed ZIKV infections in human cases have been reported to include self-limiting acute febrile illnesses with fever, headache, myalgia, retro-orbital pain, arthralgia, conjunctivitis, and rash. These clinical presentations closely resemble those that are caused by dengue virus (DENV) and Chikungunya virus [6]. Serological studies and isolation of ZIKV strains have subsequently indicated that the virus has a wide geographical distribution, including regions in East and West Africa, South and South East Asia, and Micronesia [6], where an outbreak of Zika fever was reported on Yap Island [4]. In 2013, a Zika fever outbreak in French Polynesia was the largest ever reported for an arbovirus other than DENV at this location. Andes virus (ANDV) is the etiological agent of hantavirus cardiopulmonary syndrome in Chile. In this study, we evaluated the profile of the pro-inflammatory cytokines IL-1β, IL-12p70, IL-21, TNF-α, IFN-γ, IL-10 and IL-6 in serum samples of ANDV-infected patients at the time of hospitalization. The mean levels of circulating cytokines were determined by a Bead-Based Multiplex assay coupled with Luminex detection technology, in order to compare 43 serum samples of healthy controls and 43 samples of ANDV-infected patients that had been categorized according to the severity of disease. When compared to the controls, no significant differences in IL-1β concentration were observed in ANDV-infected patients (p = 0.9672), whereas levels of IL-12p70 and IL-21 were significantly lower in infected cases (p = <0.0001). Significantly elevated levels of TNF-α, IFN-γ, IL-10, and IL-6 were detected in ANDV-infected individuals (p = <0.0001, 0.0036, <0.0001, <0.0001, respectively). Notably, IL-6 levels were significantly higher (40-fold) in the 22 patients with severe symptoms compared to the 21 individuals with mild symptoms (p = <0.0001). Using multivariate regression models, we show that IL-6 levels has a crude OR of 14.4 (CI: 3.3–63.1). In conclusion, the serum level of IL-6 is a significant predictor of the severity of the clinical outcome of ANDV-induced disease.
The IL28B SNPs rs12979860 and rs8099917, but not TNF-α SNP rs1800629, are associated with the clinical outcome of ANDV-induced disease, suggesting a possible link between IL28B expression and ANDV pathogenesis.
Glutamic acid decarboxylase activity in the main and accessory olfactory bulbs throughout the sexual cycle of the rat was studied. The effect of male pheromonal secretion on enzyme activity during proestrus and estrus day was also tested. The enzyme activity showed circadian rhythm during the estrous cycle. This rhythm was disrupted during diestrus-2 afternoon in the main bulb and came back during proestrus afternoon. A different pattern of enzyme activity was present in the accessory bulb, since the circadian rhythm was altered during proestrus morning, returning during estrus afternoon. Male odor exposition did not change enzyme profile activity during proestrus day and during estrus morning in the main bulb. In contrast, in the accessory bulb the olfactory stimuli induced opposite changes to that found in rats from the vivarium during proestrus. Comparison of enzyme activity in olfactory stimuli-deprived rats with that of pheromone-stimulated rats during proestrus showed that male odor exposure specifically affects accessory bulb enzyme activity. It is concluded that the changes of the olfactory bulb GABAergic system during proestrus and estrus day, or that evoked by odor stimuli, demonstrate the discriminative response of this system between the accessory olfactory bulb and the main olfactory bulb.Key words: glutamic acid decarboxylase, pheromone, olfactory bulb, GABA, sexual cycle.
