The aim of our work was to develop PCR test systems for the identification and differentiation of the Piscichlamydia salmonis and Clavochlamydia salmonicola, species, that are known epitheliocystis infection agents of gill and fish skin diseases, characterized by the presence of specific ‘inclusions’ in the epithelial cells of the gills. To date, the diseases of fish associated with chlamydial infections have been detected in more than 90 species of freshwater and marine fish worldwide. For now, there is no available information on the prevalence of Piscichlamydia salmonis and Clavochlamydia salmonicola, which can cause epitheliocystis of commercially important aquaculture species in Ukraine. Identification of these pathogens is possible only using molecular genetic methods. As a result of our research, we got PCR tests for the identification and species differentiation of Piscichlamydia salmonis and Clavochlamydia salmonicola. The use of diagnostics for the identification of Piscichlamydia salmonis and Clavochlamydia salmonicola makes chlamydial infections monitoring among various fish species possible and it will increase the economic efficiency of fish farms.
Products of plant origin, recommended as permanent components of the daily human diet, are rich in vitamins, minerals, plant fibers, as well as various active phytochemicals (polyphenols, flavonoids) and sterols, which have a low calorie content. According to the results of epidemiological and clinical studies, it has been established that the risks of various pathologies are reduced when using a diet that is balanced in terms of nutrients, rich in vegetable fiber and food products of plant origin. The evolution of food technology has led to the appearance on the shelves of Italian supermarkets of products defined as “ready foods” or products – semi-finished products of high quality and safety, which present all the characteristics of freshness, similar to products that have just been harvested. They also include products of vegetable origin, which belong to the fresh-cut category. A limited technological elaboration is applied to this category of products, after which they can be used without further manipulation before consumption. The products of vegetable origin of fresh-cut vegetable have the definition of “potentially dangerous products” due to their possible contamination with pathogenic microorganisms such as Escherichia coli, Salmonella spp. and Listeria monocytogenes. These pathogens very often contaminate fresh-cut vegetable, causing food poisoning in consumers. The main sources of pathogenic contamination by the causative agents of human intestinal infections are the water used for watering plants and exposure to inappropriate temperatures during the storage of plant products. The decisive aspect, without a doubt, remains the sanitary and hygienic characteristics of the product at the time of its consumption. It is fundamental to deepen our knowledge about the transmission, resistance, and growth mechanisms of pathogenic microorganisms in products of the IV range. This will allow the establishment of sampling norms for express diagnostics to reduce the possibility of low-quality products reaching the consumer.
Introduction: Waddlia chondrophila and Parachlamydia acanthamoebae are well-known and best-studied representatives of Сhlamydia-related bacteria carrying a potential zoonotic threat. These bacteria are associated with miscarriage, ectopic pregnancy, diseases of the respiratory system in both humans and animals. Despite the importance of these Сhlamydia-like organisms for human medicine along with veterinary medicine, studies on their prevalence in Ukraine were not conducted due to the lack of available tests. The aim of our work was to create relatively cheap and easy method for detection Waddlia chondrophila and Parachlamydia acanthamoebae. Materials and methods: GenBank database was used to find nucleotide sequences of the 16S rRNA gene of bacteria Chlamydiales’ order. Alignment was performed using the MEGA7 software, in order to detect the presence of polymorphic hybridization sites specifically attributed to Waddlia chondrophila and Parachlamydia acanthamoebae. Primer- BLAST software was used to design oligonucleotide primers, to evaluate the critical parameters of the primer, in particular, the melting temperature, difference between melting temperatures for the primer pairs, the GC content, the self-complementarity, etc. Results and conclusions: The amplification of control DNA of Parachlamydia acanthamoebae and Waddlia chrondophila in single PCR using the corresponding primers and subsequent gel electrophoresis of PCR products determined the size of the amplified DNA fragments 88 b.p. and 123 b.p, respectively; the fragments were in line with the expected sizes. The analytical specificity test was performed by amplifying the control DNA of 15 species of the order Chlamydiales.
The paper reveals a comprehensive approach to economic efficiency assessment of large-scale investment projects in the energy sector. Authors consider scientific approaches to integrated assessment and substantiate its advantages when applying it to the assessment of effectiveness for the energy sector. At present, the accuracy of the estimates of the efficiency of investments in the energy sector is of particular importance in terms of the limited financial resources and the increasing capital intensity of deposit development. However, existing methods of determining investment efficiency for complex projects often tend to lead to incorrect results. The integrated assessment approach proposed in the article aims to overcome the shortcomings of existing approaches. Its peculiarity is to consider investment in a complex project as a single system consisting of subsystemsindividual projects. The whole set of projects is classified according to the nature of the mutual influence, which makes the assessment process much easier. The implementation of an integrated approach requires special care in the performance evaluation and is possible in cases of simultaneous implementation of the projects included in the complex, aimed at achieving the same effects and having synchronized time frames of entry. The positive effect of increasing the accuracy of the assessment and, as a consequence, of significantly reducing the risk of economically unjustified investments, far exceeds the cost of making the evaluation procedure more complex under the developed method.
