Cây Giảo cổ lam (Gynostemma pubescens) phân bố chủ yếu tại các tỉnh phía Bắc, là một trong những loại thảo dược quí và tốt cho sức khỏe của con người. Trong nghiên cứu này, chúng tôi nghiên cứu nhân giống in vitro và nuôi trồng đánh giá sự sinh trưởng phát triển cây Giảo cổ lam cấy mô trong nhà kính tại Đà Lạt, Lâm Đồng. Kết quả cho thấy, môi trường MS (Murashige Skoog) bổ sung 1mg/l BA, 30g/l sucrose, 8g/l agar, pH 5.8 là tốt nhất đến sự tái sinh chồi in vitro, với chiều cao chồi 1.84 cm, số chồi 10.50 chồi/mẫu. Môi trường MS bổ sung 0.5 mg/l TDZ, 30g/l sucrose, 8 g/l agar, pH 5.8 là tốt nhất đến sự tái sinh chồi in vitro, với chiều cao chồi 1.99cm, số chồi 13.80 chồi/mẫu. Nồng độ IBA từ 0 - 1 mg/l đều thích hợp đến sự tái sinh rễ in vitro của cây Giảo cổ lam, với tỉ lệ tái sinh rễ đạt 100%. Vụn xơ dừa là giá thể thích hợp nhất để chuyển cây Giảo cổ lam cấy mô ra ngoài vườn ươm, với tỷ lệ sống đạt 100%, chiều cao cây 9.73cm, chiều dài rễ 6.45cm. Đất mùn là giá thể tốt nhất đến sự sinh trưởng phát triển của cây, với tỷ lệ sống đạt 100%, chiều cao cây 82.08cm, chiều dài rễ 36.57cm, khối lượng tươi 57.32g/cây. Tưới 100 ml phân Nitrophoska (2 g/l) theo định kỳ mỗi tuần một lần là tốt nhất đến sự sinh trưởng phát triển của cây, với tỷ lệ sống đạt 100%, chiều cao cây 94.22cm, chiều dài rễ 37.71cm, khối lượng tươi 59.38g/cây.
This study grafted monomer acrylamide onto cellulose (from Corchorus capsularis powder) using trigger system including sodium hydrosulfite (SB) and ammonium peroxodisulfate (APS) to remove the macrolide group (Spiramycin -SPI, Erythromycin -ERY, Tylosin -TYL) from water. Corchorus capsularis powder was pre-treated by sodium hydroxide 15 % (COC), then modified by a mixture of sodium bisulfite (SB) 0.15 mol/L and ammonium peroxydisulfate (APS) 0.2 mol/L. The material was grafted with monoacrylamide (AM) at ratio AM/COC = 1/1 in 2.5 hours at 60 o C. The adsorbent was characterized by FTIR and SEM. After modification with AM, the adsorbent (BTA) was used to adsorb SPI, ERY, and TYL from water. Effects of pH, temperature and adsorbent dose to adsorption performance were studied. Concentration of macrolides were determined by LC/MS-MS. Maximum adsorption capacities of SPI, ERY, TYL following Langmuir isotherm model were 50.00, 33.33, 25.00 mg/g, respectively. The adsorption constant Kp of SPI, ERY, and TYL following the Freundlich isotherm model were 5383.53, 1082.79 and 1004.76, respectively. The n constant ranged from 1 to 10. The Optimal adsorption condition of SPI, ERY, and TYL to BTA material were fixed at 30 o C, pH 7.
Paphiopedilum x dalatense is a beautiful orchid species with large flowers in variable colors and leaves covered with stripes and beautiful unseen mosaic spots. Recently, many people exploit this species, causing it becomes very rare. In this study, we studied the effects of various organic matter: potato, banana and tryptone, yeast powder, peptone on the growth and development of P. dalatense shoots as well as the effects of NAA and humic acid on in vitro rooting of this orchid were investigated. The research results showed that MS medium supplemented with 100 g/L banana in combination with 100 g/L potato (5,4 shoots/sample, 18,8 mm/shoot, 4,5 leaves/shoot, and shoots survival rate of 100%) or MS medium supplemented with 1 g/L peptone (4,19 shoots/sample, 15 mm/shoot, 4 leaves/bud, and 92% of shoots survival rate) were the best response for the shoot formation and development. In addition, the half strength MS culture medium supplemented with 1 mg/L NAA (5,2 leaves/sample, 4,6 roots/buds, 3,56 cm/root, and 100% rate for rooting) was the suitable medium for the in vitro rooting of P. dalatense. Being cultured on half strength MS medium supplemented with 2 mg/L humic acid, the rooting rate reached 100% with the greatest root number and the longest root (5 roots/shoots, 5,5 cm/root). The obtained results on the in vitro propagation on this orchid helps contribute to the conservation and increases the genotic pool of this precious wild orchid species, as well as the rapid multiplication of healthy plantlets serving the commercialization of precious orchid species.
