Hewan peliharaan eksotik seperti baning cokelat (Manouria emys) dan baning Sulawesi (Indotestudo forstenii) tidak lepas dari ancaman penyakit infeksi nematoda parasit gastrointestinal. Tujuan dari kajian ini adalah untuk mendeskripsikan nematoda beserta sifat patogenitas parasit yang ditemukan pada pemeriksaan bangkai M. emys dan I. forstenii. Bangkai kura-kura dinekropsi dan perubahan patologis yang ditemukan didokumentasikan. Nematoda dikoleksi dan dipreservasi dalam alkohol 70%. Nematoda diamati dengan mikroskop cahaya dan mikroskop elektron. Organ yang diduga mengalami perubahan patologis (sekum) dilakukan pengambilan dua sampel untuk proses pembuatan preparat histopatologi guna melihat struktur mikro jaringan terinfeksi dan mikroskop elektron guna melihat struktur ultra jaringan terinfeksi. Berdasarkan hasil pemeriksaan parasit, ditemukan nematoda Cissophyllus sp. sebanyak 318 individu dan Tonaudia sp. sebanyak enam individu pada sekum M. emys. Pada I. forstenii, empat individu Spironoura sp. dikoleksi dari organ intestinum. Berdasarkan hasil pemeriksaan histopatologi serta pemeriksaan mikroskop elektron pada organ sekum M. emys ditemukan adanya potongan cacing dan peradangan pada tunika submukosa dan muskularis. Morfologi kompleks mulut Cissophyllus sp. dan Tonaudia sp. diduga berkontribusi pada perubahan patologis dan menyebabkan peradangan pada sekum M. emys. Perubahan patologis yang ditemukan menunjukkan bahwa nematoda parasit Cissophyllus dan Tonaudia sp. memiliki sifat patogenitas untuk hospes M. emys. Kajian lebih lanjut perlu dilakukan untuk melihat patogenesis infeksi jenis-jenis nematoda tersebut.Kata kunci: Kura-kura darat, mikroskop elektron, nematoda, parasit gastrointestinal, patologi anatomi Abstract. Asian grass frog Fejervarya limnocharis is being utilized as pets, for laboratory experiments, for a mixture of traditional medicine and for cuisine. The harvest of F. limnocharis in high volume can threat its population. Biological data such as the age when the specimens are harvested is valuable information to manage the harvesting system in sustainable way. We conducted the skeletochronology technique using paraffin methods and hematoxylin staining from 69 samples (46 males, 21 females, 2 juveniles). The results showed that the age harvested male ranged from 1 to 3 years old, while the female ranged from 2 to 3 years old. The snout-vent length (SVL) of harvested specimens ranges between 39.84−52.37 mm for both sexes. We propose an intervention in the harvesting system by limitation of the size for harvested specimens to at least 46 mm. In this minimum size, individuals of F. limnocharis have reproduced several times and have contributed to the population in the wild.
Tokay gecko, Gekko gecko (Linnaeus, 1758) is highly exploited in Java for human consumption in China. Using skeletochronology, the age and body growth of Tokay Geckos from local populations in Central Java (18 females, 21 males) and in East Java (19 females, 42 males) were studied for the first time. A total of 100 frozen specimens (37 females, 63 males) were used. Sex was determined by observing the presence (male) or absence of pre-anal pores (female). Snout to vent length (SVL) of the smallest female and male with the line of arrested growth (LAG) was 127.1 and 160.4 mm, respectively. Both males and females reach sexual maturity before one year old with 98.3 mm SVL for females and 111.6 mm SVL for males. Analysis of the LAG counts indicated that the age of individuals that were harvested was ranged 0 – 2 years for females and males, with SVL ranging 110 – 182 mm. Growth rates in SVL are much faster in males as compared to females, as well as increasing of MP (width of bone from the edge of marrow cavity to the periosteal outer margin) inside femur bone. The mean SVL of males was larger than that of females. The Sexual Dimorphism Index (SDI) was calculated as -0.28, indicating a sexual size dimorphism in favor of males.
Abstract. Phadmacanty NLPR, Kurniati H. 2019. Short Communication: Determination of the age of the Paddy Field Frog, Fejervarya cancrivora (Anura: Dicroglossidae) by using skeletochronology. Biodiversitas 20: 2739-2743. The age of 102 individuals of paddy field frog (Fejervarya cancrivora) from Karawang District, West Java, Indonesia was skeletochronological determined. Hematoxylinophilic lines interpreted as lines of arrested growth (LAG) were observed clearly in the femur bone suggesting that this technique can be used to estimate the age of the frog, although the level of accuracy is likely to be strongly influenced by external and internal factor. In general, age estimates look close to accuracy at mature age levels; which one LAG equals the age of the frog that has passed one year old. The age structure of F. cancrivora from a paddy field in Karawang District has the maximum age of five years in females and four years in males. In general, adult females were not significantly larger than adult males.
