The role of blood monocytes in HIV-1 infection is a relatively new field of interest. What happens to HIV-1 in monocytes and their relationship to CD4+ T cells before, during, and after suppressive antiretroviral therapy (ART) is largely unstudied. Here, considering that diversity is a good indicator of continued replication over time, we evaluated the effect of ART on HIV-1 in blood monocytes and CD4+ T cells by examining the diversity of HIV-1 from 4 infected patients who underwent and stopped therapy. We determined diversity and compartmentalization of HIV-1 between blood monocytes and CD4+ T cells in each patient in relationship to their ART regimens. Our data indicate that the rate of HIV-1 diversity increase in monocytes during therapy was significantly higher than in CD4+ T cells (P<0.05), suggesting that HIV-1 present in monocytes diversify more during therapy than in CD4+ T cells. Increased rates of HIV-1 compartmentalization between monocytes and CD4+ T cells while on therapy were also observed. These results suggest that ART inhibits HIV-1 replication in CD4+ T cells more than in blood monocytes and that better treatments to combat HIV-1 in monocytes/macrophages may be needed for a more complete suppression of HIV replication.
This article analyses the work of issuing tickets to queuing customers, thereby contributing to the literature on interactive service work. It draws analytical attention to artful practices through which employees infer ticket orders from local configurations of talk, gesture and bodily movement. It reveals not only the practical reasoning deployed by the service worker, but also the agency of the customer in the course of encounters. Drawing upon video recordings of over 200 separate transactions, the demands of remedying problem orders are analysed to reveal how staff infer and clarify social ‘facts’, such as the customer’s age, their nationality, employment status and willingness to pay the higher ‘Gift Aid’ price. An image of interactive service work emerges that emphasizes the peculiar and skilful articulation of sociological categories in the course of apparently routine low-level work. The concept of ‘inferential labour’ is introduced to capture these processes, which resonates with studies of categorization and emotional labour in interactive service work.
This paper analyses how a Big Issue vendor approached passers-by and how they responded, how recognizable courses of social and economic activity were interactionally produced from initiation through to some conclusion. The paper recovers how the vendor's work was contextually embedded in the urban landscape, how it was constrained by, and actively shaped, the social order of the street. Drawing on video-audio recordings the paper contributes to a growing body of ethnographic and ethnomethodological research which has emphasized the embodied, contingent and interactional character of economic activity. By examining such materials, the paper is well positioned to describe how the vendor found his market on the street, social interventions that propelled passers-by into buying behaviour. The paper sheds light on now familiar encounters which occur millions of times each week in the UK and beyond.
This paper explores the relevance of Harvey Sacks' work for contemporary organization studies. Sacks encourages analysts to tether their studies to real-time workplace activities; ordinary scenes of work are recorded, slowed down and made the central object of study. Something of Sacks' analytic mentality and style are illustrated through the analysis of two data extracts: an emergency 999 call and a face-to-face sales encounter. A distinctive way of doing organizational analysis is discussed that foregrounds knowledgeability and agency via the examination of sequence and method. Sacks raises the possibility that organization might be recoverable from the fine-grained detail of actual episodes. The idea that order, intelligibility and the constitution of social scenes might have a basis aside from the more general notion of discourse is discussed.
Drawing on ethnomethodology and conversation analysis, this article explores the relation between practice and activity; between recruitment practice and the ordinary activities of the job interview. Job interviews are recognizably and accountably different from other interview-formats, such as broadcast news or academic research interviews. Such differences are instantly hearable because ordinary activities are built systematically so as to reveal an orientation to 'practice', distinctive purposes, entitlements, presuppositions, identities and definitions of acceptable conduct. The article illustrates analytic procedures for recovering such orientations and thus for understanding how people embed and reveal practice, with and for one another, in interaction. It is argued that the practice-turn should not overlook the fact that practice is, in the first instance, a members' phenomenon, something that members draw upon, monitor and orient to in real time interaction.
