Oneof the timber plants family which is abundant in Leuser Mountain is Dipterocarpaceae. However, the data of species and relationship ofDipterocarpaceae in Ketambe Research Station is lack and not available. The aim of the study is to analyze the phylogenetic ofDipterocarpaceae based on rbcL and matK gene in Ketambe Research Station. This research was carried out from July 2015 to August2016 in Ketambe Research Station and Forestry and Forest Genetics Laboratory of Molecular, Bogor Agricultural University. Themethod used quadrat sampling technique with purposive sampling and experimental laboratory consisting of DNA extraction, PCR,electrophoresis, and sequencing. The data analysis was done using BioEdit and MEGA6. The results showed that based onmorphological identification, there were five Dipterocarpaceae species found namely; Parashorea lucida, Shorea parvifolia, Shorealepidota, Shorea johorensis, and Hopea dryobalanoides. The phylogenetic tree based on rbcL gene showed that there were twomonophyletic groups, the first group was S. johorensis, S. lepidota, and H. dryobalanoides; and the second group consisted of S.parvifolia and P. lucida. The phylogenetic tree reconstruction based on matK gene showed that Shorea parvifolia and S. johorensis wereseparated in two different monophyletic groups.
Shorea johorensis is one of the red meranti plants in Ketambe Research Station, Gunung Leuser National Park, Aceh Tenggara. Currently, Shorea johorensis also is well known as a major source of valuable commercial timber. This research aimed to analyze the phylogenetic of Shorea johorensis based on chloroplast and nuclear DNA in Ketambe Research Station so that it can be known the relationship of Shorea johorensis with other species Dipterocarpaceae in GenBank database. The research was conducted from July 2015 to August 2016 in Ketambe Research Station and Forestry and Forest Genetics Laboratory of Molecular, Bogor Agricultural University. The method used quadrat sampling technique with purposive sampling and experimental laboratory that consisted of DNA extraction, PCR, electrophoresis, and sequencing. The data analysis was done using BioEdit, MEGA6, and BLAST. This study examines the relationship of Dipterocarpaceae in Ketambe Research Station Gunung Leuser National Park. This is based on the study of morphological diversity of vegetative organs which was carried out in July 2015 to June 2016 (one year). Quadrat sampling technique and purposive sampling technique methods were employed in this research and the data was analyzed using R2.10.0 program. Morphology identification and preservation of samples were done at Acehnese Herbarium, Department of Biology, Faculty of Mathematics and Natural Sciences, Syiah Kuala University. The results showed that there were five Dipterocarpaceae species in Ketambe Research Station which are Hopea dryobalanoides, Shorea parvifolia, Shorea lepidota, Shorea johorensis, and Parashorea lucida. These five species were grouped into three clusters, the first clusters consisted of Hopea dryobalanoides, the second cluster consisted of Parashorea lucida, and the third cluster consisted of Shorea johorensis, Shorea parvifolia, and Shorea lepidota. The closest relationship was Shorea parvifolia and Shorea lepidota with a distance of 3.464102. The furthest relationship was Hopea dryobalanoides and Parashorea lucida with a distance of 6.633250.
This research was about Dipterocarpaceae inventory at Ketambe Research Station, Gunung Leuser National Park. The research was done on July 2015. Determination of sampling plot was using purposive sampling. The parameter observed was species as well as number of species. The result showed that there were 5 species of Dipterocarpaceae found in the sampling plot. All the species found were belongs to tribe of Shoreae namely; Shorea johorensis, Shorea parvifolia, Hopea dryobalanoides, Shorea lepidota, and Parashorea lucida.Key words: Dipterocarpaceae, inventory, Gunung Leuser National Park
