In this study, we examine the linkages between abusive supervision, psychological distress, and turnover intentions. In addition, we aim to investigate whether continuance commitment moderates the effects of abusive supervision and psychological distress on turnover intentions. The sample of the study comprised 250 police personnel from one of the largest cities of Pakistan. Data were collected using a self-administered questionnaire and hypotheses were tested with linear regressions using the PROCESS macro. We found that abusive supervision positively predicts psychological distress that, in turn, positively predicts turnover intentions. Furthermore, continuance commitment attenuates the impacts of abusive supervision and psychological distress on turnover intentions. We contribute to the literature by demonstrating that sustained exposure to abusive supervision affects subordinates' turnover intentions directly as well as indirectly through psychological distress, and that the positive impacts of abusive supervision and associated psychological distress on turnover intentions are weaker among individuals who are high in continuance commitment compared with those who are low. A number of practical implications are also discussed.
In this study, the effect of multi strain probiotics was studied on antibody response, blood chemistry and hematological parameters in combination with vaccination. A total of sixty day-old broiler chicks were equally divided into four groups: Control, Probiotic, Vaccine and Probiotic + Vaccine. Probiotics were offered at 2g/500ml of water for 5 days/week up to 6 weeks. NDV vaccine was given by eye drops at 0.025ml/bird at day 7. At 21 st day NDV Lasota vaccine was offered in drinking distil water at 0.50ml /bird. IBDV vaccine was administered by drinking water at 10 th , 22 nd and 30 th day of experiment. Blood samples were collected at 15, 30 and 45 days to measure antibody response, blood chemistry and hematological parameters. Antibody titer against NDV vaccine and IBD vaccine showed decline in control and probiotic groups. In case of NDV vaccine, there was no significant difference in Vaccine group (GMT=log2 7.33 ) and Probiotic + Vaccine group (GMT=log2 7.66 ) at day 45 th of experiment. For IBDV vaccine, there was significant difference at 45 th day in ELISA titer of vaccine group (2233) and Probiotic+Vaccine group (4206). Cholesterol and ALT showed significant decrease in PV group at 45 th day. Significant increase in WBC count was observed in PV group. Level of significance was P≤0.05. Probiotics when administered along with vaccine showed improvement in antibody titer but no harmful increase or decrease in blood chemistry and hematological parameters was observed.
Background. The term “persisters” refers to a small bacterial population that persists during treatment with high antibiotic concentration or dose in the absence of genetic resistance. The present study was designed to investigate the transcriptional response in indigenous Klebsiella pneumoniae under the ciprofloxacin stress. Methods. Isolation and identification of K. pneumoniae were carried out through standard microbiological protocols. The characterization of quinolone resistance was performed by estimating the quinolone susceptibility testing, MIC estimation, and detecting the QRDR and PMQR. Transcriptional response of the isolates to ciprofloxacin was determined using qPCR. Results. Among 34 isolates, 23 (67%) were resistant to ciprofloxacin. Both QRDR (gyrA and gyrB) and PMQR (qnrA, qnrB, and qnrS) were detected in the isolates, and all were found resistant to ciprofloxacin. The mRNA levels of both mutS and euTu under the influence of ciprofloxacin were significantly increased. On ciprofloxacin exposure, the mRNA levels of the DNA damage response element (mutS) were raised in a time-dependent fashion. K. pneumoniae showed high-level resistance to ciprofloxacin in the presence of mutations in QRDR and PMQR genes. Conclusion. The transcriptional response revealed the upregulation of DNA repair and protein folding elements (mutS and euTu) in ciprofloxacin stress and delayed cell division. The ciprofloxacin was found to trigger various stress responses in a time- and concentration-dependent manner.
The emergence of SARS-CoV-2 variants has posed a challenge to disease control efforts worldwide. This study explored the genomic diversity and phylogenetic relationship of SARS-CoV-2 variants reported in Pakistan. Our objective was to understand the transmission dynamics of different lineages within the country. We retrieved and analyzed spike protein sequences from Pakistan and compared them with reference sequences reported worldwide. Our analysis revealed the clustering of Pakistan-origin isolates in nine different clades representing different regions worldwide, suggesting the transmission of multiple lineages within the country. We found 96 PANGO lineages of SARS-CoV-2 in Pakistan, and 64 of these corresponded to 4 WHO-designated variants: Alpha, Beta, Delta, and Omicron. The most dominant variants in Pakistan were Alpha (B.1.1.7), Beta (B.1.351), Delta (B.1.617.2, AY.108), and Omicron (BA.2.75, BA.5.2), and the N-terminal domain and receptor binding regions were the most hypervariable regions of the spike gene. Compared to the reference strain, characteristic substitutions were found in dominant variants. Our findings emphasize the importance of continuously monitoring and assessing nucleotide and residue substitutions over time to understand virus evolutionary trends better and devise effective disease control interventions.
Background: Antibiotic resistance is a major health hazard around the globe. Hypervirulent Klebsiella Pneumoniae (hvKp) is associated with hospital-acquired and community-acquired infections. Since there is a lack of new antibiotics against multidrug-resistant (MDR) pathogens, phage therapy might provide an alternative approach to confer antibiotic resistance. Objectives: This study aimed to estimate the occurrence of hvKp and characterize the bacteriophage against the hvKp prevalence in clinical settings, which might be used as an alternative to antibiotics. Methods: Different clinical samples (n = 50) were collected to isolate K. pneumoniae, and the assessment of multidrug resistance was carried out based on the Clinical and Laboratory Standards Institute guidelines (2020). The bacteriophage was isolated from hospital waste, and the double agar overlay method was used for phage purification and propagation. Spot test and one-step curve were performed to determine host-phage interactions. For the evaluation of phage stability in environmental conditions, the phage was incubated at various ranges of temperature, pH, and chloroform. Results: Out of the collected samples, 22 (44%) isolates were confirmed as K. pneumoniae. Among confirmed K. pneumoniae isolates, a total of 11 (50%) isolates were detected as hvKp. Moreover, 14 (64%) isolates were detected as MDR, out of which 5 (35%) isolates were among hvKp phenotypes. Maximum resistance was observed against ampicillin (86%) followed by ceftriaxone (81%) which was the highest among cephalosporins. The isolated bacteriophage showed a broad host range, short latent period, and stability. Overall, 16 isolates (85%) of K. pneumoniae were susceptible to phage infection, among which 12 isolates were MDR (75%); however, all 5 (100%) hvKp isolates were susceptible to phage infection. One-step growth analysis revealed a burst size of 190 phages/host bacterial cells with a short latent period of 24 minutes. Conclusions: Altogether, the significant prevalence of hvKp was estimated in clinical settings, and the isolated bacteriophage showed significant lytic activity as it killed all the hvKp strains. Phage therapy might be exploited and used as a potential alternative therapeutic approach against infections caused by this resistant pathogen.
