The main aim of this work was to study the leaf secondary metabolite profiles of artificially induced tetraploids (2n=4x=48) of Solanum commersonii, a diploid (2n=2x=24) wild potato species. The tetraploid genotypes of S. commersonii were produced by oryzalin treatment. Both HPLC-UV and LC/MS analyses revealed that there were no qualitative differences in the metabolite profiles between the diploid S. commersonii and its tetraploids. By contrast, the results showed that the phenylpropanoid content was generally significantly higher in the tetraploids than in the diploid S. commersonii. Concerning the glycoalkaloids (GAs), the results provided evidence that the content of minor GAs (solanidenediol triose, solanidadienol lycotetraose, and solanidenol lycotetraose) was higher in tetraploids than in the diploid progenitor, while the content of major GAs (dehydrodemissine and dehydrocommersonine) was significantly higher in diploid S. commersonii than in its tetraploid genotypes. The results are discussed from the practical perspective of potato biodiversity enhancement.
The evolutionary diversity of wild potato species makes them excellent materials for improving the narrow genetic basis of the cultivated potato Solanum tuberosum. Understanding their genetic diversity is important not only to choose the best parents for breeding, but also to design proper crossing schemes and selection strategies. The objectives of this study were to determine the resistance response to Ralstonia solanacearum, Potato virus Y and low temperatures of 21 clones of 12 potato species, and to determine their genetic diversity through simple sequence repeat (SSR) markers. Sources of resistance have been found for all the investigated traits, with high resistance variability not only between but also within species. Combined resistances were also identified, with positive implications for efficient breeding. SSR analysis allowed the detection of 12 loci and 46 alleles across all genotypes, with an average value of 3.8 alleles per locus. Both unique and rare alleles useful for marker-assisted selection were found. SSR-based cluster analysis revealed that resistant genotypes were distributed among all clusters, suggesting that genetically different resistant genotypes were identified. Wild potatoes, as dynamic resource adapted to various environmental conditions, represent a powerful and informative reservoir of genes useful for breeding efforts. WRKY transcription factors (TFs) are encoded by one of the largest families in plants and are involved in several biological processes such as growth and development, signal transduction, and plant defence against stress. In this study, 79 and 84 genes encoding putative WRKY TFs have been identified in two wild potato relatives, Solanum commersonii and S. chacoense. Phylogenetic analysis of WRKY proteins divided ScWRKYs and SchWRKYs into three Groups and seven subGroups. Structural and phylogenetic comparative analyses suggested an interspecific variability of WRKYs. Analysis of gene expression profiles in different tissues and under various stresses allowed to select ScWRKY045 as a good candidate in woundingresponse, ScWRKY055 as a bacterial infection triggered WRKY and ScWRKY023 as a multiple stressresponsive WRKY gene. Those WRKYs were further studied through interactome analysis allowing the identification of potential co-expression relationships between ScWRKYs/SchWRKYs and genes of various pathways. Overall, this study enabled the discrimination of WRKY genes that could be considered as potential candidates in both breeding programs and functional studies.Plants experience environmental constrains and pathogen attacks during their life. Being sessile organisms, their survival depends on the ability to properly and promptly reprogram cellular networks. Several and different classes of transcription factors (TFs) work as "master regulators" and "selector genes", being able to control processes that specify cell types and developmental patterning and modulate specific pathways. Among them, WRKY factors are drawing a great deal of interest in the scientific community due to their ability to simultaneously cope with multiple stresses 1,2 . They are notorious for coordinating signals in plant immunity response against several pathogens and pest attacks 3,4 . More recently, it has been confirmed that WRKYs also base defence mechanism to abiotic stresses and play a key role in cross-talk pathway networks between plant response and development 5,6 . Their involvement into multiple stress response and in plant growth regulation is evidenced by their W-box specific DNA binding 7,8 . Potato is a staple food crop and an important source of dietary energy. Its tubers contain several essential amino acids, vitamins, minerals and phytochemicals that contribute to the nutritional value of this important product. Recently, scientific interest has focused on purple and red potatoes that, due to the presence of anthocyanins, may be considered as natural powerful functional food. The aim of this study was to evaluate the characteristics of pigmented varieties, the types of anthocyanins accumulated and the level of both beneficial phytochemicals (vitamin C and chlorogenic acids, CGAs) and anti-nutritional compounds (glycoalkaloids) following various cooking methods. The analyses described the presence of a mix of several acylated anthocyanins in pigmented tubers along with high level of CGA. The amount of antioxidants was differently affected by heat treatments according to the type of molecule and the cooking methods used. In some cases, the beneficial compounds were made more available by heat treatments for the analytical detection as compared to raw materials. Data reported here describe both the agronomic properties of these pigmented varieties and the effects of food processing methods on bioactive molecules contained in this natural functional food. They may provide useful information for breeders aiming to develop new varieties that could include desirable agronomical and industrial processing traits.
Development of local plant genetic resources grown in specific territories requires approaches that are able to discriminate between local and alien germplasm. In this work, three potato (Solanum tuberosum L.) local accessions grown in the area of Majella National Park (Abruzzo, Italy) and five commercial varieties cultivated in the same area were characterized using 22 morphological descriptors and microsatellite (SSR) DNA markers. Analysis of the DNA and of the plant, leaf, flower, and tuber morpho-agronomic traits allowed for a reliable discrimination of the local potato accessions, and provided a clear picture of their genetic relationships with the commercial varieties. Moreover, infrared spectroscopy was used to acquire a fingerprint of the tuber flesh composition. A total of 279 spectra, 70% of which were used in calibration and the remaining 30% for prediction, were processed using partial least squares discriminant analysis. About 97% of the calibration samples and 80% of the prediction samples were correctly classified according to the potato origin. In summary, the combination of the three approaches were useful in the characterization and valorization of local germplasm. In particular, the molecular markers suggest that the potato accession named Montenerodomo, cultivated in Majella National Park, can be considered a local variety and can be registered into the Regional Voluntary GR Register and entered into the foreseen protection scheme, as reported by the Italian regional laws.
