The present study was undertaken to explore the effect of administration of high doses of sodium selenite on the expression of Bcl-2 in patients with non-Hodgkin's lymphoma (NHL). Fifty patients with newly diagnosed NHL were randomly divided into two groups. Group A-I received standard chemotherapy whereas group A-II received adjuvant sodium selenite 0.2 mg kg-1 day-1 for 30 days in addition to chemotherapy. Enzyme-linked immunosorbent assay was used to assess Bcl-2 at the time of diagnosis and after therapy in the two groups. Sodium selenite administration resulted in significant decline of Bcl-2 level after therapy in group A-II (8.6 +/- 6.9 ng/ml vs 3 6.9 +/- 7.9 ng/ml, P < 0.05). Also, complete response reached 60% in group A-II compared to 40% in group A-I. Significant increase in CD4/CD8 ratio was noticed in group A-II compared to group A-I after therapy (1.45 +/- 0.36 vs 1.10 +/- 0.28 p 0.04). Overall survival time in months was significantly longer in complete remission patients in group A-II (21.87 +/- 1.41) compared to group A-I (19.70 +/- 1.95) (p = 0.01). It is concluded that sodium selenite administration at the dosage and duration chosen acts as a down regulator of Bcl-2 and improves clinical outcome.
Hysterothylacium aduncum (Nematoda, Anisakidae) was isolated from the intestine of the common sole Solea solea (Family, Soleidae) collected from coasts along Alexandria City at the Mediterranean Sea in Egypt, during the period from May to September 2013. Light and scanning electron microscopy revealed that this nematode parasite belongs to the family Anisakidae in the genus Hysterothylacium. The type species is named H. aduncum, based on the presence of three interlocked lips with the interlabium in between, the presence of cephalic papillae, and large numbers of caudal papillae in males. Body measurements showed that the male worms were smaller than females measuring 13.9-18 mm (16.2 ± 0.2) in length and 0.26-0.34 mm (0.30 ± 0.01) in width. Females measured 20.5-24.5 mm (22.7 ± 0.2) in length and 0.41-0.52 mm (0.45 ± 0.01) in width. The morphological characteristics of this species was confirmed by molecular analysis of 18S rDNA for these parasites followed by comparison between sequence data for them with those obtained from the Genbank showing that H. aduncum is deeply embedded in the genus Hysterothylacium with a sequence similarity between 95.5-94.3 % with close relationships to other H. aduncum specimens and Hysterothylacium sp.. Furthermore, it was shown that this parasitic nematode is able to accumulate larger concentrations of heavy metals such as Fe, Cu, Cd, and Ni within its tissues than of its host fish and thus it can be used as a useful bio-indicator of water pollution.
Aspiculuris tetraptera is a heteroxynematid nematoda infecting most of the laboratory animals, occasionally mice which represent the mostly used animal for biological, medical, and pharmacological studies. The present study aimed to investigate the prevalence of nematode parasites infection in the laboratory mice Mus musculus in Egypt. Morphologically, this oxyurid possessed four distinct cephalic papillae on a cephalic plate, with three small rudimental lips carrying two sessile poorly developed labial papillae and one pair of amphidial pores. Esophagus divided into cylindrical corpus and globular bulb. Distinct cervical alae interrupted at the level of esophago-intestinal junction forming an acute angle. At the caudal end, twelve caudal papillae in male worms while an ovijector apparatus opening and a vulva surrounded by protruded lips in females were observed. The general morphological criteria include this nematode with other Aspiculuris species which were compared in the present study. Molecular characterization based on 18SSU rDNA sequencing performed to confirm the taxonomic position of this species and to documents the morphological data. Sequence alignment detects a percent of identity up to 88.0% with other Heteroxynematidae species. Phylogenetic analysis showed that the present recorded is a putative sister taxon to A. tetraptera recorded in a previous study. The SSU rDNA sequence has been deposited in the GenBank under the accession no. MG019400.
In an aquatic environment, there is a profound and inverse relationship between environmental quality and disease status of fish. Parasites are one of the most serious limiting factors in aquaculture. Therefore, the present investigation was carried out during the period of February-December 2014 to determine the parasitic infections in the African sharptooth catfish Clarias gariepinus, relative to the capability of internal parasites to accumulate heavy metals. Up to 100 catfish were examined for gastrointestinal helminths and 38% of fish were found to be infected with the cestode Polyonchobothrium clarias. The morphology of this parasite species, based on light and scanning electron microscopy, revealed that the adult worm was characterized by a rectangular scolex measuring 0.43-0.58 (0.49 ± 0.1) mm long and 0.15-0.21 (0.19 ± 0.1) mm wide, with a flat to slightly raised rostellum armed with a crown with two semicircles each bearing 13-15 hooks, followed by immature, mature and gravid proglottids which were about 29-55 (45), 16-30 (24) and 15-39 (28) in number, respectively. The mature proglottid contained a single set of genitalia in which medullary testes measured 0.09-0.13 (0.11 ± 0.01) mm long and 0.05-0.08 (0.06 ± 0.01) mm wide; a bi-lobed ovary was situated near the posterior margin of the proglottid, extending laterally up to the longitudinal excretory canals; the tubular uterus arose from the ootype up to the anterior margin of the proglottid; and vitelline follicles were cortical. The greater portion of the gravid proglottid was occupied by a uterus filled with unoperculate and embryonated eggs. Chemical analysis confirmed that the concentrations of heavy metals (Zn, Cu, Mn, Cd, Ni and Pb) accumulated in P. clarias were higher than in fish tissues and values recommended by FAO/WHO, with the exception of Zn, which was found to be higher in fish kidneys than in the cestode. This supports the hypothesis that cestodes of fish can be regarded as useful bioindicators when evaluating the environmental pollution of aquatic ecosystems by heavy metals.
The present study described a new species of the genus Echinocephalus that was isolated from the intestine of the common carp Cyprinus carpio inhabiting Burullus Lake, Kafr el-Sheikh Governorate, Egypt. Thirty-two out of 70 examined specimens (45.71 %) were found to be naturally infected. The adult male worms were elongated and measured 0.79-0.85 mm (0.81 ± 0.02) in length and 0.21-0.32 mm (0.28 ± 0.002) in width, while females reached 0.89-0.98 mm (0.9 ± 0.02) long and 0.25-0.38 mm (0.28 ± 0.002) wide. The reported species are investigated microscopically, genetically, and compared with previous related ones. The present study recorded this new nematode parasite as Echinocephalus carpiae with accession no. KC493258, inhabiting new host and locality records in Egypt. It is especially characterized by short body length, cephalic bulb with few (six to eight) multiple rows of spines, clear annulated cuticle, and new genetic sequences, which identify the paraphyly of Gnathostomatinae.
