Genetic engineering strategy in chilli peppers (Capsicum annuum L.) has been applied so far to improve its genetic capacity for instance against some important diseases. This strategy needs an established system for calli preparation as well as transformant regeneration. In most cases, this necessity is genotype dependent especially for some local genotypes. Based on this, we optimized media for calli induction, shoot and root regeneration of our three local cultivars namely Kampung, Kopay, and Trisula. Media combinations basically were composed of MS and combined with some growth factors, for instance, IAA, BAP, NAA and TDZ with different combinations for each calli induction, shoot and root regeneration. All media compositions were supplemented with 30 g/L sucroses. The best medium for calli induction was MS media supplemented with 4 mg/L BAP, 0.5 mg/L IAA and L2 vitamin (Thiamin-HCl and Pyrodoxine-HCl). While the best medium for shoot induction and elongation was MS supplemented with 1mg/L BAP, 5 mg/L AgNO 3 , 2 mg/L GA 3 , 2 mg/L Calcium Pantothenate and L2 vitamin. Furthermore, the best medium for root induction and elongation was WPM (Woody Plant Medium) supplemented with 0.4 mg/L NAA. This finding should provide optimal media for calli induction and shoot-root regeneration for the three local cultivars Kampung, Kopay, and Trisula.
Penelitian tentang pertumbuhan dan hasil tanaman bawang merah pada berbagai konsentrasi pupuk organik cair fermentasi urine sapi telah dilaksanakan di Jorong Kopi, Nagari Bukit Sileh, Kecamatan Lembang Jaya, Kabupaten Solok yang terletak pada ketinggian ± 1400 m dpl. Penelitian bertujuan mendapatkan konsentrasi pupuk organik cair fermentasi urine sapi yang terbaik untuk pertumbuhan dan hasil bawang merah yang optimal. Rancangan penelitian yang digunakan adalah rancangan acak kelompok (RAK) faktor tunggal dengan perlakuan yang diuji adalah pupuk organik cair urine sapi. Terdapat 6 taraf perlakuan yang diaplikasikan yaitu 0 ml/1 liter air, 10 ml/1 liter air, 20 ml/1 liter air, 30 ml/1 liter air, 40 ml/1 liter air, 50 ml/1 liter air. Setiap perlakuan terdiri dari 4 ulangan. Data hasil pengamatan dianalisis ragam dan dilanjutkan dengan uji lanjut Duncan’s Multiple Range Test (DMRT) pada taraf nyata 5%. Peubah yang diamati adalah tinggi tanaman, jumlah daun, jumlah umbi/rumpun, bobot umbi/rumpun, bobot kering umbi/rumpun dan hasil/hektar. Hasil penelitian menujukkan bahwa aplikasi pupuk organik cair fermentasi urine sapi memberikan hasil tertinggi pada dosis 50 ml/1 liter air. Hal ini terlihat pada tinggi tanaman, bobot segar umbi per rumpun, bobot kering umbi per rumpun dan hasil per hektar.
Organic fertilizers can be used in cauliflower cultivation as liquid organic fertilizer from Paitan. Paitan is a weed with organic matter and nutrients that can increase soil and plant fertility. The response of growth and production of cauliflower was examined with different doses of paitan liquid organic fertilizer. The randomized block design in this research is five groups with five treatments and then tested applying the Duncan New Multiple Range Test at a significance level of 5 % and 1 % utilizing SAS. Paitan liquid organic fertilizer doses given include treatment 20 ml, 40 ml, 60 ml, and 80 ml paitan/plants. Parameters observed on cauliflower were plant height, amount of leaf, leaf width, flower diameter, flower production weight, and flower weight. The findings of the research indicated no significant effect of liquid organic fertilizer on cauliflower plant height, the amount of leaf, leaf width, and flower diameter. The best impact of this research on cauliflower is the weight of flower production and flower weight treatment doses of 80 ml paitan liquid organic fertilizer.
