Records from 2105 calves born over 2 yr on a large dairy in a subtropical climate were examined to delineate factors that influence passive transfer of colostral immunoglobulins and the effect of passive transfer of immunoglobulin on calf mortality. Seasonal effects on immunoglobulin absorption were detected with highest serum total protein occurring in February and March. Lower total protein concentrations were associated with elevated environmental temperatures in the summer months. Maternal antibody was highest in calves from second parity cows; and dystocia appeared to decrease the amount of immunoglobulin absorbed by the neonatal calf. Calves that died of infectious disease during the first 14 wk of life had significantly lower serum total protein concentrations than those that lived. Heritability of serum protein concentration in calves was .02 +/- .03.
Pancreatic ductal adenocarcinoma (PDAC) is a fatal disease due to early metastatic spread, late diagnosis and the lack of efficient therapies. A major driver of cancer progression and hurdle to successful treatment is transforming growth factor (TGF)-β. Recent data from pancreatic cancer mouse models showed that transcriptionally active p73 (TAp73), a p53 family member, inhibits tumor progression through promoting tumor suppressive canonical TGF-β/Smad signaling, while preventing non-canonical TGF-β signaling through extracellular signal-regulated kinases (ERK)1/2. Here, we studied whether this mechanism also operates in human PDAC. Using the PDAC-derived tumor cell lines PANC-1, HPAFII and L3.6pl, we showed that TAp73 induces the expression of the epithelial marker and invasion suppressor E-cadherin and the common-mediator Smad, SMAD4, while at the same time suppressing expression of the EMT master regulator SNAIL and basal and TGF-β1-induced activation of ERK1 and ERK2. Using dominant-negative and RNA interference-based inhibition of SMAD4 function, we went on to show that inhibition of ERK activation by TAp73 is mediated through SMAD4. Intriguingly, both SMAD4 and the α isoform of TAp73—but not the β isoform—interfered with cell migration, as shown by xCELLigence technology. Our findings highlighted the role of TAp73-SMAD4 signaling in tumor suppression of human PDAC and identified direct inhibition of basal and TGF-β-stimulated pro-invasive ERK activation as an underlying mechanism.
Results strongly suggest that there is a high prevalence of Salmonella spp in cull dairy cows at slaughter, which could burden Hazard Analysis Critical Control Point programs implemented in slaughter establishments. Procedures to reduce Salmonella load at the dairy farm and during transport to slaughter could reduce the risk of spread during the slaughter process.
Aims Biopsy of the periprosthetic tissue is an important diagnostic tool for prosthetic joint infection (PJI) as it enables the detection of the responsible microorganism with its sensitivity to antibiotics. We aimed to investigate how often the bacteria identified in the tissue analysis differed between samples obtained from preoperative biopsy and intraoperative revision surgery in cases of late PJI; and whether there was a therapeutic consequence. Methods A total of 508 patients who required revision surgery of total hip arthroplasty (THA) (n = 231) or total knee arthroplasty (TKA) (n = 277) because of component loosening underwent biopsy before revision surgery. The tissue samples collected at biopsy and during revision surgery were analyzed according to the criteria of the Musculoskeletal Infection Society (MSIS). Results In total, 178 (113 THA, 65 TKA) were classified as infected. The biopsy procedure had a sensitivity of 93.8%, a specificity of 97.3%, a positive predictive value (PPV) of 94.9%, a negative predictive value (NPV) of 96.7%, and an accuracy of 96.1%. Of the 178 infected patients, 26 showed a difference in the detected bacteria from the biopsy and the revision surgery (14.6%). This difference required a change to antibiotic therapy in only two cases (1.1%). Conclusion Biopsy is a useful tool to diagnose PJI, but there may be a difference in the detected bacteria between the biopsy and revision surgery. However, this did not affect the choice of antibiotic therapy in most cases, rendering the clinical relevance of this phenomenon as low. Cite this article: Bone Joint J 2020;102-B(3):329–335
Data were from 492 Holstein cows calved between October, 1976, and May, 1977, in eight herds in Michigan. Diameter of the cervix of each cow was estimated by palpation at 8 to 21 days postpartum and again 2 wk later. Parturition and postpartum discharge from the genital tract were classified as normal or abnormal. Size of the cervix between 12 and 26 days postpartum was classified as small, medium, or large within two parity groups. Incidence of abnormal parturition and incidence of abnormal postpartum discharge were 14.1 and 38.0%. Involution of the cervix was affected by parity and type of postpartum discharge. Involution of the cervix occurred sooner postpartum in primiparous than in multiparous cows and sooner postpartum in cows with normal discharge than in cows with abnormal discharge. The greatest difference in cervix diameter between cows with normal or abnormal discharges was 10 mm at 3 wk postpartum. Days from parturition to first detected estrus and first service were extended by abnormal postpartum discharge but not by abnormal parturition. First-service conception rate was lower and days from parturition to conception were higher in cows with medium or large cervices compared to cows with small cervices. Type of postpartum discharge and size of cervix early postpartum may be useful for predicting subsequent reproductive performance in dairy cows.
