ABSTRACT. The herpes simplex virus 2 (HSV-2) is one of the most important sexually transmitted pathogens, and can facilitate the spread of human immunodeficiency virus. The currently available antiviral drugs have certain limitations. Nanosilver has received increasing attention recently with respect to its antibacterial and antiviral properties. The purpose of this study was to determine the inhibiting effect and mechanism of silver nanoparticles (Ag-NPs) on HSV-2. The cytotoxicity of Vero cells induced by different Ag-NP concentrations was investigated by using the methyl thiazolyl tetrazolium (MTT) assay. The inhibiting effect of Ag-NPs on HSV-2 at various times was also evaluated by using a plaque assay. The toxicity of 100 μg/mL Ag-NPs on Vero cells was very low. The mixture of Ag-NP suspension and HSV-2 prior to infecting cells could significantly inhibit the production of progeny viruses. AgNPs also inhibited the replication of HSV-2 for 24 h before infecting cells with HSV-2. Therefore, 100 μg/mL Ag-NPs could completely inhibit HSV-2 replication. Ag-NPs at nontoxic concentrations were capable of inhibiting HSV-2 replication when administered prior to viral infection or soon after initial virus exposure. PurposeThe aim of our study was to assess, for the first time, the validity, reliability, and acceptability of the European Organization for Research and Treatment of Cancer (EORTC) Quality of Life questionnaire (QLQ) cervical cancer module (CX24) in Chinese cervical cancer patients.Patients and methodsOne hundred fifteen outpatients with cervical cancer in the First Affiliated Hospital of Xinxiang Medical University from May 2013 to July 2013 were included in this study. All participants self-administered the EORTC QLQ-CX24 and the core questionnaire (EORTC QLQ-C30), and the Karnofsky Performance Scale was performed to evaluate scores. Data were analyzed with Cronbach’s α coefficient, Pearson correlation test, multitrait scaling analysis, and Mann–Whitney U test.ResultsScale reliability was confirmed by Cronbach’s α coefficients for internal consistency, which ranged from 0.71 to 0.82. Convergent and discriminant validity were confirmed by multitrait scaling analysis, which revealed three (3.4%) scaling errors for symptom experience scales and zero (0%) for body image as well as sexual/vaginal functioning scales. Higher missing value rate occurred in sexuality-related items. The clinical validity of the Chinese version of the EORTC QLQ-CX24 was demonstrated by the ability to discriminate among patients in different International Federation of Gynecology and Obstetrics stages.ConclusionThe EORTC QLQ-CX24 was proved to be a reliable and valid instrument with which to measure the quality of life in cervical cancer patients in the People’s Republic of China.
Abstract. MicroRNA serve crucial roles in a variety of human cancer types. The miR-302-367 cluster has been reported to suppress the proliferation of cervical carcinoma cells through the novel target AKT1; however, the molecular mechanism of miR-302 in cervical cancer metastasis remains unclear. The present study aimed to investigate the clinical significance of miR-302-3p expression in cervical cancer, and to examine the regulatory mechanism of miR-302-3p in the malignant phenotypes of cervical cancer cells. The present data indicated that miR-302-3p was significantly downregulated in cervical cancer tissues compared with the level in adjacent non-tumor tissues, and low expression of miR-302-3p was significantly associated with node metastasis, advanced clinical stage, and poor prognosis in patients with cervical cancer. Restoration of miR-302-3p expression caused a significant reduction in cervical cancer cell migration and invasion. Defective in cullin neddylation 1 domain containing 1 (DCUN1D1) was identified as a novel target gene of miR-302-3p, and miR-302-3p negatively regulated the mRNA and protein expression of DCUN1D1 in cervical cancer HeLa cells. Additionally, overexpression of DCUN1D1 rescued the effects of miR-302-3p on the migration and invasion of cervical cancer cells. Furthermore, DCUN1D1 was upregulated in cervical cancer tissues compared with the levels in adjacent tissues, and its high expression was associated with node metastasis, advanced clinical stage, and shorter survival time in patients with cervical cancer. Notably, a negative correlation between miR-302-3p and DCUN1D1 expression in cervical cancer tissues was observed. Taken together, the present study suggests that miR-302-3p serves a suppressive role in cervical cancer metastasis, partly at least, via directly targeting DCUN1D1. Therefore, miR-302-3p/DCUN1D1 may be a potential therapeutic target for cervical cancer treatment.
Oxygen dissociation and activation on surfaces play a crucial role in heterogeneous catalysis and oxidation processes. In this study, we have conducted a series of scanning tunneling microscopy (STM) experiments combined with density functional theory calculation to investigate the oxidation process in a single crystal Co(0001) surface. For the first time, we show a comprehensive in situ STM study of the oxidation process of Co(0001) from an atomic point of view. With low O 2 exposure at 90 K, chemisorbed oxygen pairs are observed showing a dumbbell-like STM feature. At a relatively higher temperature range of 160−250 K, a large-scale p(2 × 2)−O adlayer forms and the O adatoms show surprisingly high mobility. With the temperature of Co(0001) kept at ≥300 K, adsorption of oxygen leads to fast oxidation of the surface to amorphous cotton-like protrusions, which occur initially at the step/edge sites and interstitial defect sites.
According to our preliminary results, simple USgHIFU is a safe and effective single-treatment method of treating uterine leiomyoma greater than 10 cm in diameter and is an almost innocuous alternative therapeutic strategy. Advances in knowledge: The conclusions indicate simple USgHIFU is safe and effective as one-time treatment of uterine leiomyoma greater than 10 cm in diameter, it could be a promising therapeutic strategy.
