Summary We need to determine the best use of soil vis–NIR spectral libraries that are being developed at regional, national and global scales to predict soil properties from new spectral readings. To reduce the complexity of a calibration dataset derived from the Chinese vis–NIR soil spectral library (CSSL), we tested a local regression method that combined geographical sub‐setting with a local partial least squares regression (local‐PLSR) that uses a limited number of similar vis–NIR spectra (k‐nearest neighbours). The central idea of the local regression, and of other local statistical approaches, is to derive a local prediction model by identifying samples in the calibration dataset that are similar, in spectral variable space, to the samples used for prediction. Here, to derive our local regressions we used Euclidean distance in spectral space between the calibration dataset and prediction samples, and we also used soil geographical zoning to account for similarities in soil‐forming conditions. We tested this approach with the CSSL, which comprised 2732 soil samples collected from 20 provinces in the People's Republic of China to predict soil organic matter (SOM). Results showed that the prediction accuracy of our spatially constrained local‐PLSR method (R2 = 0.74, RPIQ = 2.6) was better than that from local‐PLSR (R2 = 0.69, RPIQ = 2.3) and PLSR alone (R2 = 0.50, RPIQ = 1.5). The coupling of a local‐PLSR regression with soil geographical zoning can improve the accuracy of local SOM predictions using large, complex soil spectral libraries. The approach might be embedded into vis–NIR sensors for laboratory analysis or field estimation.
MicroRNAs (miRNAs) are endogenous small non-coding RNAs regulating gene expression in animals and plants. To find some differentially expressed miRNAs that may be associated with age-dependent behavioural changes in honey bees (Apis mellifera), we applied next-generation high-throughput sequencing technology to detect small RNAs in nurses and foragers. Our results showed that both nurses and foragers had a complicated small RNA population, and the length of small RNAs varied, 22 nucleotides being the predominant length. Combining deep sequencing and bioinformatic analysis, we discovered that nine known miRNAs were significantly different between nurses and foragers (P < 0.01; absolute value of fold-change ≥ 1). Some of their target genes were related to neural function. Moreover, 67 novel miRNAs were identified in nurses and foragers. Ame-miR-31a and ame-miR-13b were further validated using quantitative reverse-transcription PCR assays. The present study provides new information on the miRNA abundance of honey bees, and enhances our understanding of miRNA function in the regulation of honey bee development.
An efficient Agrobacterium-mediated durum wheat transformation system has been developed for the production of 121 independent transgenic lines. This improved system used Agrobacterium strain AGL1 containing the superbinary pGreen/pSoup vector system and durum wheat cv Stewart as the recipient plant. Acetosyringone at 400 μM was added to both the inoculation and cultivation medium, and picloram at 10 mg l−1 and 2 mg l−1 was used in the cultivation and induction medium, respectively. Compared with 200 μM in the inoculation and cultivation media, the increased acetosyringone concentration led to significantly higher GUS (β-glucuronidase) transient expression and T-DNA delivery efficiency. However, no evident effects of acetosyringone concentration on regeneration frequency were observed. The higher acetosyringone concentration led to an improvement in average final transformation efficiency from 4.7% to 6.3%. Furthermore, the concentration of picloram in the co-cultivation medium had significant effects on callus induction and regeneration. Compared with 2 mg l−1 picloram in the co-cultivation medium, increasing the concentration to 10 mg l−1 picloram resulted in improved final transformation frequency from 2.8% to 6.3%, with the highest frequency of 12.3% reached in one particular experiment, although statistical analysis showed that this difference in final transformation efficiency had a low level of significance. Stable integration of foreign genes, their expression, and inheritance were confirmed by Southern blot analyses, GUS assay, and genetic analysis. Analysis of T1 progeny showed that, of the 31 transgenic lines randomly selected, nearly one-third had a segregation ratio of 3:1, while the remainder had ratios typical of two or three independently segregating loci.
