A +s-$sorption-elution (viradel) method using electropositive microporous filters (Virosorb 1 -MDS) was used for &e concentration and detection of infectious pancreatic necrosis virus (IPNV) at a hatchery with a past history of IPNV infection. Samples of fish tissue and unconcentrated water were also examined for the presence of IPNV. Virus was isolated from three of the nine concentrated water samples taken from various locations at the hatchery. Virus was also isolated from pooled fish tissues, but not from unconcentrated water samples. Representative numbers of viruses isolated from water and fish tissues were examined for virus neutralization in the presence of anti-IPNV antiserum; resistance to acid, ether, and heat; and replication in the presence of 5-bromo-2'-deoxyuridine (BUDR). When tested by dot-immunobinding assay using a panel of monoclonal antibodies (MABs), the viral isolates were found to belong to the West Buxton serotype of IPNV.

Evaluierung einer Wasseranreuherungsmethode fur den Nachweis des infektiosen Pankreas-Nekrose-Virus in einer FischbrutanstaltEine Virus-Adsorptions-Eluationsmethode (Viradel) wurde unter Verwendung eines efektropositiven MikroToren!ters (Virosox 1 -MDS) eingesetzt, urn den infektiosen Pankreas-Nekrose-Virus (IPNV) anzureichern und in einer Brutanstdt mir friiherer IPNV-Infektion nachzuweisen. Gewebeproben der Fische und Wasserproben wurden ebenfalls auf die Anwesenheit von IPNV hin ge riift. Der Virus wurde aus drei von neun aufkonzentrierten Wasserproben isoliert, die von verschidnen SteUen der Brutanstalt entnommen wurden. Der Virus wurde auch aus zusammengefatken Fischgewebeproben isoliert, jedoch nicht in unbehandelten Wasserproben. Re risentative Viszahlen aus dem Wasser und den Fischgeweben wurden auf die mogliche VirusneutrJsation in Gegenwart von Anti-IPNV-Antiserum hin untersucht sowie auf Widerstandsfahigkeit gegenuber Sauren, Estern und W h e . A virus adsorption-elution (viradel) method for use with positively charged microporous filters was previously developed in our laboratory to concentrate waterborne infectious pancreatic necrosis virus (IPNV). In the present study, this method was evaluated for the detection of IPNV in water of laboratory aquaria containing IPNV-infected rainbow trout Onchorhynchus mykiss. Waterborne IPNV samples concentrated by the viradel method were compared with samples of sediment, fish tissue homogenate, and untreated water to determine how much the viradel method could improve rapidity and efficiency of virus detection. Results of three separate experiments indicated a significant, positive correlation between the virus liters in fish tissue and in the viradel method samples and between the sediment and untreated water samples.
