A number of Escherichia coli strains isolated from patients suffering from diarrhea were characterized and isolated. They were classified according to their activity of recognition of specific glycosphingolipid-receptors separated on thin-layer chromatogram (TLC) and those incorporated into liposomes. Our finding revealed that 20% of tested strains (group 1) bound Galα1-4Gal-containing glycosphingolipids found in total natural fractions of human meconium and human stomach as well as reference glycosphingolipids. The majority of EPEC strains (group 2) recognized clearly GalNAcβ4Gal-containing receptors within these fractions. Using lipid extracts from total natural fractions of the stomach and meconium, group 1 bound an actively large number of glycosphingolipid receptors demonstrated by intensified black bands, whereas two positive bindings in the 2-and 4-sugar-regions were only detected when EPEC group 2 was used. Application of liposomeaggregation binding assay, the two selected strains EPEC 37 and EPEC 57, representing group 1 and group 2, respectively, recognized well Galα1-4Gal and GalNAcβ4Gal-containing glycosphingolipids incorporated into liposomes receptors. In the present study, lactosylceramideglycosphingolipid (Galβ4Glcβ1Cer) with nonhydroxylated ceramide showed no binding activity in both assay-systems, whereas the same receptorcontaining hydroxylated fatty acid of ceramide was bound by both strain groups.
