Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disorder characterized by the selective death of motor neurons. Causative mutations in the global RNA-processing proteins TDP-43 and FUS among others, as well as their aggregation in ALS patients, have identified defects in RNA metabolism as an important feature in this disease. Lethal congenital contracture syndrome 1 and lethal arthrogryposis with anterior horn cell disease are autosomal recessive fetal motor neuron diseases that are caused by mutations in another global RNA-processing protein, hGle1. In this study, we carried out the first screening of GLE1 in ALS patients (173 familial and 760 sporadic) and identified 2 deleterious mutations (1 splice site and 1 nonsense mutation) and 1 missense mutation. Functional analysis of the deleterious mutants revealed them to be unable to rescue motor neuron pathology in zebrafish morphants lacking Gle1. Furthermore, in HeLa cells, both mutations caused a depletion of hGle1 at the nuclear pore where it carries out an essential role in nuclear export of mRNA. These results suggest a haploinsufficiency mechanism and point to a causative role for GLE1 mutations in ALS patients. This further supports the involvement of global defects in RNA metabolism in ALS.
Despite numerous regulations, methamphetamine consumption persists; its availability has even increased in the United States. Methamphetamine is produced in small labs and super labs that are differentiated by the quantity of drug they generate and by how they are embedded in trafficking networks. The stagnant statistics regarding methamphetamine consumption and lab seizures suggest that laws have been ineffective, partly due to the producers’ adaptability. To understand this adaptation, methamphetamine recipes collected online will be analyzed through a qualitative methodology. Emphasis will be placed on the impact of the American legislation toward synthetic drug production. This article describes how methamphetamine producers have adapted to get around the regulations. The producers synthesize the regulated precursors by extracting them from processed products. To comply with the quotas imposed by law, the producers limit their quantities used. This article suggests that producers keep abreast of legislations and perfect the recipes accordingly.
According to the criminal career paradigm, the link between past and future criminal activities is important and the desistance process may vary for individuals whose criminal trajectories were punctuated by failure compared with those who achieved a certain level of success. This study, based on the life narratives of 27 individuals who maintained a state of nonoffending for more than a year, examines how criminal achievement modulates the desistance process. The aim is to understand whether criminal achievement acts as a barrier or a facilitator in the desistance process. A short questionnaire based on the life-history calendar method was used to classify individuals according to the parameters of their criminal careers. Narrative life stories were then used to look at the obstacles and frustrations encountered during desistance. The results show the relation between criminal achievement and desistance is complex: success in criminal activities is not always hindering desistance.
Cryptomarkets are online illicit marketplaces where drug dealers advertise the sale of illicit drugs. Anonymizing technologies such as the Tor network and virtual currencies are used to hide cryptomarket participants' identity and to limit the ability of law enforcement agencies to make arrests. In this paper, our aim is to describe how herbal cannabis dealers and buyers in the United States have adapted to the online sale of herbal cannabis through cryptomarkets. To achieve this goal, we evaluate the size and scope of the American herbal cannabis market on cryptomarkets and compare it to other drug markets from other countries, evaluate the impact of cryptomarkets on offline sales of herbal cannabis, and evaluate the ties between the now licit herbal cannabis markets in some States and cryptomarkets. Our results suggest that only a small fraction of herbal cannabis dealers and drug users have transitioned to cryptomarkets. This can be explained by the need for technical skills to buy and sell herbal cannabis online and by the need to have access to computers that are not accessible to all. The slow rate of adoption may also be explained by the higher price of herbal cannabis relative to street prices. If cryptomarkets were to be adopted by a larger portion of the herbal cannabis market actors, our results suggest that wholesale and regional distributors who are not active on cryptomarkets would be the most affected market's participants.
La recherche scientifique cherche la plupart du temps à comprendre la nature mais se tourne aussi à l’occasion vers elle-même pour mieux comprendre la genèse des connaissances scientifiques. Cette quête s’est traduite par une littérature importante portant sur les réseaux de collaborations entre auteurs pour produire des textes scientifiques. L’objectif général de cette branche de recherche est d’expliquer la structure des collaborations et de déterminer les corollaires de la performance dans le monde universitaire. Cet article vise à combler un manque de connaissance sur les réseaux de collaboration francophones en étudiant 40 ans de collaborations entre auteurs de la revue Criminologie. La méthodologie adopte une approche égocentrique et sociométrique et caractérise les liens entre les coauteurs de la revue. Nos résultats démontrent une augmentation des collaborations dans le temps, mais sans nécessairement la création d’une seule et unique communauté d’auteurs qui contrôlerait la diffusion des connaissances dans la revue. Les acteurs clés sont présentés mais ne montrent aucune stabilité dans le temps.
