This study aimed to develop a TaqMan Real-Time Polymerase Chain Reaction method, using a novel primer for detection of pork adulteration in meatballs. The study is important as it described a TaqMan method for product adulteration analysis. TaqMan is known to have a more specific result compared to SYBR green analytical method. Assay in the study combined species-specific primers and TaqMan probes to targeting 153 bp fragment of D-loop mitochondrial region of pork. A specificity test was conducted on fresh tissues of pork, beef, chicken, wild boar, dog, and mouse. Meatballs as samples were prepared from a mixture of pork-beef and wild boar-beef with concentrations as follows: 5%, 10%, 25%, 75%, 90%, and 100%. The linearity and sensitivity of the method were performed by measuring the amplification curve from the dilution series, namely 1000, 200, 100, 10, 5, 1, and 0.5 pg/μL of DNA, extracted from 100% pork meatballs. A repeatability test was conducted as many as six repetitions on 100% pork and 100% wild boar meatballs. This study showed that mitochondrial D-loop species-specific primers and TaqMan probes could identify the DNA of pork and wild boar on the fresh tissues. Additionally, it also resulted in a threshold cycle (Ct) of 17.02 and 17.95 for pork, 22.22 for wild boar, while the negative result for others. The detection limit has shown 5 pg in the meatball formulation. The Relative Standard Deviation (RSD) of repeatability was 1.936% for pork, while 2.140% for wild boar. The developed method was also applied to analyzing commercial meatballs. A TaqMan real-time PCR analytical method using specific primer targeting on 153 bp fragment of the D-loop mitochondrial region could be applied as a standard method for identifying pork and wild boar in food samples intended for halal authentication studies
Objective: This study aimed to design specific primers derived from mitochondrial cytb of Sus Scrofa (1F1R primer) used in the pork meatball analysis using real time polymerase chain reaction (RT-PCR) method. Materials and Methods: Such designed primers were validated and these included specificity of primer, linearity, and sensitivity of the method as well as the repeatability test. The primers were specifically affirmed in the fresh tissue of chickens, cows, pigs, and goats. The linearity and sensitivity of the method was conducted by measuring the amplification curve from a series of dilution (0, 1, 1, 10, 100, 1,000, and 10,000 pg/μl of DNA) extracted from 100% pork meatball formulation. The repeatability test was conducted by determining the cycle threshold ( Ct ) values of RT-PCR amplification from 100% pork meatball formulation as many as six times. Results: Primer of 1F1R (forward: 5′-ACG CGA TAT AAG CAG GTA AA-3′; reverse: 5′-CTG CTT TCG TAG CAC GTA TT-3′) was specific in analyzing the presence of pork in meatball formulation at 47.1°C, which was optimum annealing temperature. The DNA identification was able to use the primers by RT-PCR with 1 pg as the limit of detection, efficiency value was 242.58%, and the coefficient of determination value ( R 2 ) was 0.956. The coefficient of variance was 4.13%. The developed method was also fruitfully applied to analyze commercial meatballs. Conclusion: RT-PCR method using specific primers targeting on mitochondrial gene (1F1R primer) could be used as the standard method for identification of pork in food samples intended for halal authentication studies.
Unawareness of the roles and competencies of other health professions has led to many medical and medication errors in the treatment of patients in the hospital. It is considered important to give Interprofessional Education (IPE) to students at pre-clinic and clinical stage in order to have a good understanding about roles of other health professionas. IPE is an interprofessional collaborative learning to support promotive, preventive, curative, rehabilitative and other approches related to health aspect. Perception is among the most important and highly needed skill in the implementation of IPE. This research aimed to determine the effect of IPE learning toward the perception among the students in Faculty of Medicine and Health Science of Universitas Muhammadiyah Yogyakarta (FMHS UMY). This is an observational study adopting a cross sectional design. The sample of students was collected through purposive sampling technique. A number of 94 students met the inclusion criteria, comprised of 21 students of Medical Profession Education, 25 of Dentistry Profession Education, 25 of Nursing Profession Education and 21 Undergraduate students of Pharmacy. Samples were given questionnaire of<em> Interdiciplinary Education Perception Scale</em>. Comparative test<em> </em>was done by<em> Kruskall-Wallis test.</em> From 94 students of FMHS, 75.5% had good perception toward IPE. No significant differences appeared in perception (p=0.285) among the students of FMHS UMY. The <em>Interprofessional Education</em> (IPE) had significant effects toward the perception among the students of FMHS UMY.
Pengetahuan tentang halal dalam segala aspek, khususnya makanan-minuman semestinya tidak hanya menjadi domain orang dewasa. Anak-anak mulai usia dini juga sudah layak untuk dikenalkan dengan terminologi-terminologi dalam syariat Islam terkait dengan masalah halal-haram. Edukasi terhadap prinsip-prinsip yang benar, pemahaman terhadap titik kritis dan mengenali jenis-jenis bahan pangan yang meragukan status kehalalannya dapat meningkatkan daya kritis anak terhadap produk yang beredar. Dalam kegiatan ini dilakukan usaha edukasi pengenalan terminologi-terminologi halal dan haran terhadap anak usia dini dengan menggunakan media buku 3 dimensi/ pop-up book. Buku tiga dimensi berwarna (pop-up book) yang yang digunakan berisi informasi/ pengetahuan tentang halal dan haram. Konten informasi yang diajukan merujuk kepada ayat Al-Qur’an dan Hadis Nabi Saw. yang berderajat sahih dan atau hasan. Buku didesain dengan tampilan yang menarik dengan harapan dapat memudahkan bagi anak-anak terhadap informasi yang disajikan. Program ini dijalankan dengan kolaborasi antara IRPC Farmasi UMY dengan Halalan Thayyiban Center (HTC) UMY. Penggunakan media yang diaplikasi dengan model belajar sambal bermain terbukti dapat menarik minat peserta didik dalam hal anak TK/ usia dini. Model buku pop-up dapat memudahkan bagi anak untuk mengingat dan bahkan menghafal poin-poin yang ada dalam buku.
