SignificanceThe completion of seed germination is an irrevocable event for plants, determining, for most plants, the site of the remainder of their life cycle. One environmental cue important to the completion of seed germination is light, which, in Arabidopsis thaliana, can influence a host of transcription factors, including PHYTOCHROME-INTERACTING FACTOR1 (PIF1), a negative regulator of the completion of germination and seedling de-etiolation. The KELCH F-BOX protein COLD TEMPERATURE GERMINATING10 (CTG10) can recognize and bind to PIF1, negatively influencing PIF1 stability, stimulating the completion of germination, and promoting a de-etiolated seedling morphology. PIF1, in turn, can downregulate CTG10 expression, revealing a complex coregulation orchestrated by light presence and quality that dictates whether the seed completes germination.
Purpose:Strabismus and anisometropia are the most common causes of amblyopia. It can be easily prevented or treated if detected early. With the changing socio-cultural-economic milieu of the society, the perspectives of strabismus in society are gradually changing but still adequate knowledge, awareness, and attitude of parents toward strabismus will help in preventing amblyopia and aid in the proper psychosocial adaptation of such children. This study aimed to assess knowledge and attitude of parents toward children suffering from strabismus.Methods:A prospective study was carried out from January 1 to February 29, 2016, through a structured questionnaire to assess the level of knowledge and attitude of parents of children suffering from strabismus.Results:One hundred and twenty parents of children with strabismus were interviewed through a questionnaire. Education level of 78 parents was less than graduation (60%) and of 42 parents (40%) was graduation or higher. The majority of the parents, i.e., 116 (96.67%) were bothered due to strabismus. One hundred and one (84.17%) parents felt that their child's strabismus was noticed by others during interaction. Seventy-four (61.67%) parents felt that their children will have difficulty in making friends. Ninety (75%) parents felt uncomfortable if someone asked something about their child's strabismus. One hundred and ten (91.67%) parents considered strabismus as cosmetic stigma.Conclusion:Some parents, especially from the lower educated segment, had poor understanding of strabismus, thus resulting in late presentation and ineffective countermeasures. The key to prevent strabismic amblyopia and its psychosocial impacts is to provide health education regarding strabismus.
Arabidopsis FY, a homologue of the yeast RNA 3' processing factor Pfs2p, regulates the autonomous floral transition pathway through its interaction with FCA, an RNA binding protein. It is demonstrated here that FY also influences seed dormancy. Freshly-harvested seed of the Arabidopsis fy-1 mutant germinated readily in the absence of stratification or after-ripening. Furthermore, the fy-1 mutant showed less ABA sensitivity compared with the wild type, Ler, under identical conditions. Freshly-harvested seed of fy-1 had significantly higher ABA levels than Ler, even though Ler was dormant and fy-1 germinated readily. The PPLPP domains of FY, which are required for flowering control, were not essential for the ABA-influenced repression of germination. FLC expression analysis in seeds of different genotypes suggested that the effect of FY on dormancy may not be elicited through FLC. No significant differences in CYP707A1, CYP707A2, NCED9, ABI3, and ABI4 were observed between freshly-harvested Ler and fy-1 imbibed for 48 h. GA3ox1 and GA3ox2 rapidly increased over the 48 h imbibition period for fy-1, with no significant increases in these transcripts for Ler. ABI5 levels were significantly lower in fy-1 over the 48 h imbibition period. The results suggest that FY is involved in the development of dormancy and ABA sensitivity in Arabidopsis seed.
As a crop, flax holds significant commercial value for its omega-3 rich oilseeds and stem fibres. Canada is the largest producer of linseed but there exists scope for significant yield improvements. Implementation of mechanisms such as male sterility can permit the development of hybrids to assist in achieving this goal. Temperature sensitive male sterility has been reported in flax but the leakiness of this system in field conditions limits the production of quality hybrid seeds. Here, we characterized a 2,588 bp transcript differentially expressed in male sterile lines of flax. The twelve intron gene predicted to encode a 368 amino acid protein has five WD40 repeats which, in silico, form a propeller structure with putative nucleic acid and histone binding capabilities. The LuWD40-1 protein localized to the nucleus and its expression increased during the transition and continued through the vegetative stages (seed, etiolated seedling, stem) while the transcript levels declined during reproductive development (ovary, anthers) and embryonic morphogenesis of male fertile plants. Knockout lines for LuWD40-1 in flax failed to develop shoots while overexpression lines showed delayed growth phenotype and were male sterile. The non-viable flowers failed to open and the pollen grains from these flowers were empty. Three independent transgenic lines overexpressing the LuWD40-1 gene had ∼80% non-viable pollen, reduced branching, delayed flowering and maturity compared to male fertile genotypes. The present study provides new insights into a male sterility mechanism present in flax.
