This study aims to investigate the effects and underlying mechanisms of overexpression microRNA-9-5p (miR-9-5p) on the Aβ-induced mouse hippocampal neuron cell line HT22. Different concentrations of Aβ25-35 (10, 20, 40, 80, and 160 μM) treatment were used to establish AD model in HT22 cells. The CCK-8 assay was used to measure the cell viability. The mRNA expression levels of miR-9-5p and glycogen synthase kinase-3β (GSK-3β) were determined by RT-qPCR. HT22 cell apoptosis was analyzed flow cytometry. MiR-9-5p was down-regulated in Aβ25-35-induced HT22 cells. GSK-3β is a functional target for miR-9-5p. MiR-9-5p overexpression inhibited Aβ25-35-induced mitochondrial dysfunction, cell apoptosis, and oxidative stress by regulating GSK-3β expression in HT22 cells. Furthermore, through targeting GSK-3β, overexpression of miR-9-5p partly activated nuclear factor Nrf2/Keap1 signaling, including part increases of Nrf2, HO-1, SOD-1, GCLC expression and slight decrease of Keap1 expression. Our results showed miR-9-5p may play a powerful role in the pathogenesis of AD.
DNA methyltransferase 3A (DNMT3A) mutations occurred in 18%~23% of acute myeloid leukemia (AML) patients, and were considered to be an adverse prognostic factor for adult de novo AML cases. However, the relevant molecular mechanism of the mutation in AML pathogenesis remains obscure. In this study, we established K562 and SKM1 cell model carrying the DNMT3A R882H mutation via transcription activator-like effector nuclease (TALEN) and Clustered regularly interspaced short palindromic repeats (CRISPR/Cas9) technology, and discovered that mutated DNMT3A could promote the proliferative capability of malignant cell clones. Further RNA microarray analysis revealed that some genes crucial for glutathione (GSH) synthesis, including CTH, PSPH, PSAT1 and especially SLC7A11 (the cysteine/glutamate transporter) were significantly up-regulated, which resulted in significant elevation of intracellular GSH levels. A subsequent experiment demonstrated that the mutant clones are resistant to chemotherapy as well as SLC7A11-inhibitorsBy shRNA induced SLC7A11 silencing, we discovered profoundly decreased cellular GSH and cell proliferative ability of DNMT3A mutated clones. Our results provided novel insight into the role of the DNMT3A R882H mutation in AML pathogenesis and suggested that targeting the cellular GSH synthetic pathway could enhance the current therapy for AML patients with the DNMT3A R882H mutation.
. The local HIF-2a/EPO pathway in the bone marrow is associated with excessive erythrocytosis and the increase in bone marrow microvessel density in chronic mountain sickness. High Alt Med Biol. 16:318-330, 2015.-Aim: Chronic mountain sickness (CMS) is characterized by excessive erythrocytosis, and angiogenesis may be involved in the pathogenesis of this disease. The bone marrow niche is the primary site of erythropoiesis and angiogenesis. This study was aimed at investigating the associations of the levels of hypoxia-inducible factors (HIFs), erythropoietin (EPO), and erythropoietin receptor (EPOR), as well as microvessel density (MVD) in the bone marrow with CMS. Results: A total of 34 patients with CMS and 30 control subjects residing in areas at altitudes of 3000-4500 m were recruited for this study. The mRNA and protein expression of HIF-2a and EPO in the bone marrow cells was significantly higher in the CMS patients than in the controls. Moreover, changes in HIF-2a expression in CMS patients were significantly correlated with EPO and hemoglobin levels. In contrast, the expression of mRNA and protein expression of HIF-1a and EPOR did not differ significantly between the CMS and control patients. Increased MVD was observed in the bone marrow of the patients with CMS and it was significantly correlated with hemoglobin. Conclusions: Bone marrow cells of CMS patients may show enhanced activity of the HIF-2a/EPO pathway, and EPO may regulate the erythropoiesis and vasculogenesis through autocrine or/and paracrine mechanisms in the bone marrow niche. The increased MVD in the bone marrow of CMS patients appears to be involved in the pathogenesis of this disease.
Abstract. Gastric cancer is a lethal disease with a high mortality rate. Studies have suggested that prostate stem cell antigen (PSCA) rs2294008 polymorphism is associated with gastric cancer (GC). In this case-control study, we investigated rs2294008 polymorphism in the Tibet, Hui and Han nationalities in the Qinghai area of China. Genomic DNA was extracted from the peripheral blood of 286, 315 and 350 healthy volunteers and from 219, 233 and 265 Helicobacter pylori-negative non-cardia GC patients from the Tibet, Hui and Han populations, respectively. The rs2294008 polymorphism was analyzed by denaturing high-performance liquid chromatography. rs2294008 CT and TT genotypes were associated with GC both in the Tibet and Han populations (adjusted OR=1.51, 1.47, 2.01, 1.85; 95% CI, 1.04-2.19, 1.05-2.06, 1.04-3.88, 1.03-3.34; P=0.030, 0.025, 0.039, 0.040, respectively). rs2294008 TT genotype was associated with GC in the Hui population (adjusted OR=2.14; 95% CI, 1.29-3.55; P=0.003). Furthermore, when stratified by histopathology, the rs2294008 CT and TT genotypes were associated with diffuse GC in the Tibet and Han nationalities (adjusted OR=1.93, 1.73, 2.69, 2.86; 95% CI, 1.09-3.44, 1.01-2.95, 1.06-6.84, 1.27-6.46; P=0.025, 0.045, 0.038, 0.011, respectively). However, the rs2294008 TT genotype was associated with both intestinal and diffuse types of GC (adjusted OR=2.10, 2.21; 95% CI, P=0.012, 0.023, respectively) and the rs2294008 CT genotype was only associated with intestinal-type GC in the Hui nationalitiy group (adjusted OR=1.60; 95% CI, 1.04-2.47; P=0.034). The results therefore showed that rs2294008 may differentially contribute to GC among different nationalities in one area and its role is independent from Helicobacter pylori-infection.
