: This study was carried out to obtain morphological and physiological characteristics of ear mushrooms for an artificial cultivation. Eighteen strains were cultivated with bag culture and classified into mainly five groups such as brown, black, white, purple and others group. The highest yield was shown in 43007 strain as 98.3 g/bag and 43009, 43016, 43025 and 44035 strains were more than 60 g/bag. Among collected strains, 43007, 43009 and 43035 were selected in this study as superior strains. Three selected strains were investigated for optimal mycelial growth conditions. MCM and GPYM media were selected for the favorable culture medium. The carbon sources of 43007, 43009 and 43035 on mycelial growth were maltose, fructose and glucose, respectively and peptone was selected as a nitrogen source. Highest mycelial growth was observed when the C/N ratio was 10 for 43007 and 20 for 43009 and 43035.
To select the viable alternative substrates among the variable organic substrates for productivity enhancement and production cost-reduction of oyster mushroom in bottle culture, this study was carried out at mushroom research institute of GGRDA in 2007. In bottle culture of oyster mushroom (Plerutus ostreatus), the seedcakes of rape (RS), soybean (SS), coconut (CCS), and kapok (KS) were examined as substitute of cotton seedcake which was primary nutritive material of mushroom growing substrate. The chemical properties of substrate mixed with kapok seedcake is similar to the mixture with cotton seedcake in T-C, T-N, C/N ratio, and other nutrients. Mixed growing substrate containing cotton seedcake and kapok seedcake was superior to other mixtures 99.2% and 99.5%, respectively in spawning ratio and was faster mycellium growth in column test than that of soybeen seedcake, cotton + soybeen seedcake, and coconut seedcake. The period required in first pin-heading was 1-2 days longer in rape and soybeen seedcake mixture. Also there wad no primodia and fruitbody formation at soybeen seedcake mixture which had highest T-N content among the other mixed substrates. Yield per bottle and biological efficiency were highest of 144.6 g and 75.4%, respectively at kapok seedcake mixture. As a result, this study found that cotton seedcake can be replaced with kapok seedcake in bottle culture of oyster mushroom.
To determine sugars content of agricultural products and foods, simultaneous quantitative analysis was carried out on fructose, glucose, sucrose, maltose, and lactose by HPLC-RI. Analysis conditions were set as column ZORBAX carbohydrate (4.6 mm ID×250 mm, 5 μm), the mobile phase of 75% ACN, the column temperature of 35°C, sample injection amount of 10 μL and the flow rate of 1 mL/min. Five standard solutions were isolated without interfering peak within 30 minutes and the calibration curves of standard were confirmed excellent linearity from 0.10% to 1.00% with R2≥0.999. Based on the chromatogram of the standard solution, the limit of quantification (LOQ) and the limit of detection (LOD) values were calculated. The accuracy of the analytical values were highest at 100% water extraction method to the fructose 95.7%, sucrose 98.7%, lactose 102.7% respectively, compared with reference value of a certified reference material (BCR644), by applying the four solvent extraction methods. Using an in-house quality control material (infant formula), repeatability and reproducibility values of this experiment were verified on the basis of AOAC guideline and reference values were set up at 1.17 g/100g of glucose, 0.85 g/100g of maltose, and 45.54 g/100g of lactose. Quality control charts were drawn up and used for sugars analysis of agricultural products.
