Laccases are multi-copper oxidase enzymes that catalyze the oxidation of different compounds (phenolics and non-phenolics). The scientific literature on laccases is quite extensive, including many basic and applied research about the structure, functions, mechanism of action and a variety of biotechnological applications of these versatile enzymes. Laccases can be used in various industries/sectors, from the environmental field to the cosmetics industry, including food processing and the textile industry (dyes biodegradation and synthesis). Known as eco-friendly or green enzymes, the application of laccases in biocatalytic processes represents a promising sustainable alternative to conventional methods. Due to the advantages granted by enzyme immobilization, publications on immobilized laccases increased substantially in recent years. Many patents related to the use of laccases are available, however, the real industrial or environmental use of laccases is still challenged by cost–benefit, especially concerning the feasibility of producing this enzyme on a large scale. Although this is a compelling point and the enzyme market is heated, articles on the production and application of laccases usually neglect the economic assessment of the processes. In this review, we present a description of laccases structure and mechanisms of action including the different sources (fungi, bacteria, and plants) for laccases production and tools for laccases evolution and prediction of potential substrates. In addition, we both compare approaches for scaling-up processes with an emphasis on cost reduction and productivity and critically review several immobilization methods for laccases. Following the critical view on production and immobilization, we provide a set of applications for free and immobilized laccases based on articles published within the last five years and patents which may guide future strategies for laccase use and commercialization.
Este trabalho buscou relatos recentes na literatura sobre a utilização da casca do arroz (CA) em processos de queima visando a produção de sílica. Em primeiro momento, avaliou-se a caracterização da casca de arroz para quantificações de seus componentes e a partir disso foi possível identificar elementos presentes na rede que compõe este resíduo, que são de extrema importância para escolha do processo de queima a ser adotado. Dois diferentes grupos de processos de combustão da CA foram analisados: queima controlada e não controlada. A utilização de um pré-tratamento precedente ao processo de queima também foi analisado objetivando a obtenção de cinzas com maior grau de pureza em relação ao percentual de sílica amorfa. A transformação das cinzas em energia e biomateriais que podem ser aplicados na construção civil, indústria de vidros, cerâmicas etc, reduzem os danos ambientais causados pela emissão de gases do efeito estufa, descarte em aterro sanitário ou despejo indevido no solo. A extração de subprodutos com valor agregado, é uma maneira de reduzir o consumo de matéria prima proveniente do meio ambiente colaborando para o desenvolvimento econômico sustentável.
This study aimed to evaluate the waste cooking oil (WCO) hydrolysis in ultrasonic system using lipase as catalyst. Lipase was produced by the fungus Aspergillus niger via solid state fermentation (SSF) using canola meal as substrate. Prior to the hydrolysis reaction, the lipase behavior when subjected to ultrasound was evaluated by varying the temperature of the ultrasonic bath, the exposure time and the equipment power. Having optimized the treatment on ultrasound, the WCO hydrolysis reaction was carried out by evaluating the oil:water ratio and the lipase concentration. For a greater homogenization of the reaction medium, a mechanical stirrer at 170rpm was used. All steps were analyzed by experimental design technique. The lipase treatment in ultrasound generated an increase of about 320% in its hydrolytic activity using 50% of ultrasonic power for 25min. at 45°C. The results of the experimental design conducted for ultrasound-assisted hydrolysis showed that the best condition was using an oil:water ratio of 1:3 (v:v) and enzyme concentration of 15% (v/v), generating 62.67μmol/mL of free fatty acids (FFA) in 12h of reaction. Thus, the use of Aspergillus niger lipase as a catalyst for hydrolysis reaction of WCO can be considered as a possible pretreatment technique of the oil in order to accelerate its degradation.
