This article discusses OpenAI's ChatGPT, a generative pre-trained transformer, which uses natural language processing to fulfill text-based user requests (i.e., a "chatbot"). The history and principles behind ChatGPT and similar models are discussed. This technology is then discussed in relation to its potential impact on academia and scholarly research and publishing. ChatGPT is seen as a potential model for the automated preparation of essays and other types of scholarly manuscripts. Potential ethical issues that could arise with the emergence of large language models like GPT-3, the underlying technology behind ChatGPT, and its usage by academics and researchers, are discussed and situated within the context of broader advancements in artificial intelligence, machine learning, and natural language processing for research and scholarly publishing.
We numerically study heat conduction in a few one-dimensional Fermi-Pasta-Ulam (FPU)-type lattices by both nonequilibrium heat bath and equilibrium Green-Kubo algorithms. In those lattices, heat conductivity κ is known to diverge with length N as N α . It is commonly expected that the running exponent α should monotonously decreases with N and a recent study has shown that α for the FPU-β lattice saturates to 1/3 as N ∼ 10 4 . However, our calculations clearly show that α changes its behaviour, increasing towards the asymptotic value 2/5 for yet larger N values. As for the purely quartic lattice, α = 2/5 is clearly observed in four orders of magnitude of N ranging from 10 2 to 10 6 . This unexpected reversal phenomenon can be observed more clearly in a much shorter FPU-αβ lattice.
Peripheral nerve injury results in limited nerve regeneration and severe functional impairment. Mesenchymal stem cells (MSCs) are a remarkable tool for peripheral nerve regeneration. The involvement of human umbilical cord MSC‐derived extracellular vesicles (hUCMSC‐EVs) in peripheral nerve regeneration, however, remains unknown. In this study, we evaluated functional recovery and nerve regeneration in rats that received hUCMSC‐EV treatment after nerve transection. We observed that hUCMSC‐EV treatment promoted the recovery of motor function and the regeneration of axons; increased the sciatic functional index; resulted in the generation of numerous axons and of several Schwann cells that surrounded individual axons; and attenuated the atrophy of the gastrocnemius muscle. hUCMSC‐EVs aggregated to rat nerve defects, down‐regulated interleukin (IL)‐6 and IL‐1β, up‐regulated IL‐10 and modulated inflammation in the injured nerve. These effects likely contributed to the promotion of nerve regeneration. Our findings indicate that hUCMSC‐EVs can improve functional recovery and nerve regeneration by providing a favourable microenvironment for nerve regeneration. Thus, hUCMSC‐EVs have considerable potential for application in the treatment of peripheral nerve injury.
BackgroundCircular RNAs (circRNAs) are a class of novel RNAs with important biological functions, and aberrant expression of circRNAs has been implicated in human diseases. However, the feasibility of using blood circRNAs as disease biomarkers is largely unknown.MethodsWe explored the potential of using human peripheral blood mononuclear cell (PBMC) circRNAs as marker molecules to diagnose active pulmonary tuberculosis (TB).FindingsFirst, we demonstrated that circRNAs are widely expressed in human PBMCs and that many are abundant enough to be detected. Second, we found that the magnitude of PBMC circRNAs in TB patients was higher than that in the paired healthy controls. Compared with host linear transcripts, the circRNAs within several pathways are disproportionately upregulated in active TB patients, including “Cytokine-cytokine receptor interaction”, “Chemokine signaling pathway”, “Neurotrophin signaling pathway”, and “Bacterial invasion of epithelial cells”. Based on the differentially expressed circRNAs within these pathways, we developed a PBMC circRNA-based molecular signature differentiating active TB patients from healthy controls. We validated the classification power of the PBMC circRNA signature in an independent cohort with the area under the receiver operating characteristic curve (AUC) at 0.946.InterpretationOur results suggest that PBMC circRNAs are potentially reliable marker molecules in TB diagnosis.
