Natural and edible materials have attracted increasing attention in food packaging, which could overcome the serious environmental issues caused by conventional non-biodegradable synthetic packaging. In this work, gelatin nanofibers incorporated with two kinds of essential oil (EO), peppermint essential oil (PO) and chamomile essential oil (CO), were fabricated by electrospinning for potential edible packaging application. Electron microscopy showed that smooth and uniform morphology of the gelatin/EOs was obtained, and the diameter of nanofibers was mostly enlarged with the increase of the EO content. The proton nuclear magnetic resonance spectrum confirmed the existence of PO and CO in nanofibers after electrospinning. The addition of EOs led to an enhancement of the water contact angle of nanofibers. The antioxidant activity was significantly improved for the nanofibers loaded with CO, while the antibacteria activity against Escherichia coli and Staphylococcus aureus was better for the fibers with PO addition. The combination of half PO and half CO in nanofibers compensated for their respective limitations and exhibited optimum bioactivities. Finally, the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay with NIH-3T3 fibroblasts demonstrated the absence of cytotoxicity of the gelatin/EO nanofibers. Thus, our studies suggest that the developed gelatin/PO/CO nanofiber could be a promising candidate for edible packaging.
The stabilities of three natural antioxidants, vitamin C (VC), (-)-epigallocatechin gallate (EGCG), and curcumin, in silk films were examined and mechanisms of stabilization were elucidated. The antioxidants were physically incorporated into three types of silk films: as-cast, dried from hydrogels, and methanol-treated. Films were stored at 4, 37, and 45 °C for 30 days in phosphate-buffered saline, pH 7.4, along with controls consisting of free antioxidants. Incorporation of antioxidants did not significantly change film morphology or secondary structure. When stored at 4 °C, all samples showed similar antioxidant activities (percent scavenging) at different time points, determined by the colorimetric 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. At higher temperatures, VC in the as-cast film, EGCG in the as-cast and dried hydrogel films, and curcumin in the methanol-treated films retained more than 50% scavenging activity after 14 days of storage, significantly higher than the other samples. Interaction between antioxidants and silk, as well as degradation of the antioxidants, was investigated by fast-performance liquid chromatography (FPLC) and high-pressure liquid chromatography (HPLC), with an aim of understanding the mechanisms of silk-based stabilization. Binding of antioxidant molecules to hydrophobic or to hydrophilic/hydrophilic boundary regions of silk, depending on the chemical properties of the antioxidant, may account for the observed stabilization effects. The data can help guide further engineering of antioxidant-functionalized silk biomaterials.
Curcumin, a natural phenolic compound derived from the plant Curcuma longa, was physically entrapped and stabilized in silk hydrogel films and its influence on human bone marrow-derived mesenchymal stem cells (hBMSCs) was assessed related to adipogenic differentiation. The presence of curcumin significantly reduced silk gelation time and changed the porous morphology of gel matrix, but did not change the formation of silk beta-sheet structure. Based on spectrofluorimetric analysis, curcumin likely interacted with hydrophobic residues in silk, interacting with the beta-sheet domains formed in the hydrogels. The antioxidant activity of silk film-associated curcumin remained functional over at least one month in both the dry and hydrated state. Negligible curcumin was released from silk hydrogel films over 48 hours incubation in aqueous solution. For hBMSCs cultured on silk films containing more than 0.25 mg/mL curcumin, cell proliferation was inhibited while adipogenesis was significantly promoted based on transcripts as well as oil red O staining. When hBMSCs were cultured in media containing free curcumin, both proliferation and adipogenesis of hBMSCs were inhibited when curcumin concentrations exceeded 5 μM, which is more than 1,000-times higher than the level of curcumin released from the films in aqueous solution. Thus, silk film-associated curcumin exhibited different effects on hBMSC proliferation and differentiation when compared to curcumin in solution.
The development of food packaging possessing bioactivities which could extend the shelf life of food has gained increased interest in recent years. In this study, gelatin nanofibers with encapsulated angelica essential oil (AEO) were fabricated via electrospinning. The morphology of gelatin/AEO nanofibers was examined by scanning electron microscopy (SEM) and the addition of AEO resulted in the increase of fiber diameter. The proton nuclear magnetic resonance (1H-NMR) spectra were measured to confirm the presence of AEO in nanofibers. The hydrophobic property of gelatin nanofibers was also found to be improved with the addition of AEO. The nanofibers incorporated with AEO showed significant antioxidant activity and inhibitory effect against both Gram-negative and Gram-positive bacteria in a concentration dependent manner. Furthermore, the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) assay demonstrated that the developed gelatin/AEO nanofibers revealed no cytotoxicity effect. Thus, gelatin nanofibers incorporated with AEO can be used as potential food packaging.
In this work, an energetic composite fiber, in which 2,6-diamino-3,5-dinitropyrazine-1-oxide (LLM-105) nanoparticles intimately incorporated with a nitrocellulose/glycidyl azide polymer (NC/GAP) fiber, was prepared by the electrospinning method. The morphology and structure of the nanofiber was characterized by scanning electron microscopy (SEM), energy dispersive X-Ray (EDX), fourier transform infrared spectroscopy (IR), X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), and Brunauer–Emmett–Teller (BET). The nanofibers possessed a three-dimensional (3D) net structure and a large specific surface area. Thermal analysis, energetic performance, and sensitivities were investigated, and they were compared with NC/GAP and LLM-105 nanoparticles. The NC/GAP/nano-LLM-105 nanofibers show higher decomposition rates and lower decomposition temperatures. The NC/GAP/nano-LLM-105 decomposed to CO2, CO, H2O, N2O, and a few NO, -CH2O-, and -CH- fragments, in the thermal-infrared spectrometry online (TG-IR) measurement. The NC/GAP/nano-LLM-105 nanofibers demonstrated a higher standard specific impulse (Isp), a higher combustion chamber temperature (Tc), and a higher specialty height (H50). The introduction of nano-LLM-105 in the NC/GAP matrix results in an improvement in energetic performance and safety.
