A number of studies were undertaken in the territory of the “Syrovatka river basin of the Emerald Network of Ukraine (UA0000428 Syrovatka river basin). Based on the data gathered from field work surveys of the area, the characteristics of the habitats (C1.222, C1.224, C1.32, C1.33, C2.33+, С2.34, C3.4, C3.51, D5.2, E1.2, E2.2, E3.4, E5.4, F3.247, F9.1+G1.11, G1.21+G1.41, G1.A1, G1.7, G1.8, G1.A4) are presented according to the area covered and their ecological features. Phytosociological characteristics of habitats were given. As a result of the analysis of the distribution of certain species of sozophytes, a brief description of the status of the identified populations was presented. Populations of vascular plant species protected in Europe (Ostericum palustre, Iris hungarica), Ukraine (Epipactis helleborine, Lilium martagon, Platanthera bifolia), and at the regional level (Carpinus betulus, Campanula persicifolia, Calla palustris, Dryopteris cristata, Inula helenium, Utricularia vulgaris) have been studied in detail. The main threats to the existence of the natural complex of this area, which may appear in the situation of increasing economic impact are drainage and ploughing of floodplains, terracing, and afforestation of meadow-steppe slopes of the ravine. The typological characteristics of artificial and natural forest plantations of the territory were described based on the generalization of details gathered from information on their local management.
Raw materials derived from the species Potentilla alba L. have been widely used in medicine. It has been shown that roots and rhizomes contain compounds with a number of antioxidant properties that may support therapeutic, namely anti-inflammatory and adaptogenic effects. The species is also promising in the complex treatment of thyroid pathology. Analysis of the obtained raw materials for the presence of the required plant species for the manufacture of medicines is essential procedure. The purpose of the research is to identify the anatomical features of the root and rhizome of Potentilla alba. It will help to identify these plants in the raw material. Comparison of the anatomical structure of the roots and rhizomes of five members of the genus Potentilla L.: P. alba L., P. anserinа L., P. argentea L., P. pilosa Willd., P. reptans L. has been held. All species have been grown in the collection of the Fomin Botanical Garden. The cross sections of the root and rhizome with a thickness of 15-20 μm were made by the means of a freezing microtome. The inclusions were identified in the resulting cross sections stained with safranin, KI and sudan. The species differ in the development and location of xylem elements, and by localization of inclusions. The most effective anatomical markers of P. alba, compared to other species of the genus, are the development and the location of rhizome xylem elements. The vessels of the secondary xylem are small, placed with uneven radial strands. The thicknes of strands is 1-3 vessels, delimited by the rays of the xylem parenchyma. The number of inclusions depends to a greater extent on the conditions of plant growth, so it is a less important factor during the analysis of plant raw materials.
Our research goal has been to find the optimal nutrient media for initiation of the primary callus in the species of the Cactaceae family. Common methods of plant biotechnology were used. Primary explants of the cacti were cultivated on Murashige and Skoog medium (MS medium). The content of macro- and microelements has been diluted twice (½ MS) and the vitamins (B1 and B6 – 0.5 mg/l, PP – 1 mg/l) were added, as well as 100 mg/l meso-isonitol and 20 g/l of sucrose. It was determined that callus formation formed efficiently when cultivated on half MS media with 20 g/l sucrose, 3 mg/l 6- benzylaminopurine, 0,2 mg/l indole-3-acetic acid,0,1 mg/l α-napthaleneacetic acid and 5 mg/l ascorbic acid. It was discovered, that for initiation of tissue differentiation and cacti callus formation, high concentrations of cytokinine-active growth regulators are required.
