The present investigation was undertaken to isolate, screen and evaluate a selected promising PGPR Brevibacillus brevis on cotton crop. Out of 156 bacterial isolates one of the most promising isolate was analyzed for the various PGP traits. A seed germination analysis was conducted with cotton seeds to evaluate the potential of the isolate to promote plant growth. The bacterial isolate was checked for its growth and survival at high temperatures. The isolate was also analyzed for the PGP traits exhibited after the heat treatment. To identify the isolate morphological, biochemical and molecular characterization was performed. The isolate was found positive for many of the PGP attributes like IAA, ARA, anti-fungal activity and ammonia production. Effect of seed bacterization on various plant growth parameters was used as an indicator. The isolate showed significant growth and exhibited various PGP traits at high temperature making it suitable as an inoculant for cotton crop. Isolate was identified as Brevibacillus brevis [SVC(II)14] based on phenotypic as well as genotypic attributes and after conducting this research we propose that the B. brevis which is reported for the first time for its PGP potential in cotton, exerts its beneficial effects on cotton crop through combined modes of actions.
Plant growth promoting rhizobacteria are the soil bacteria inhabiting around/on the root surface and are directly or indirectly involved in promoting plant growth and development via production and secretion of various regulatory chemicals in the vicinity of rhizosphere. There has been much research interest in PGPB and there is now an increasing number of PGPB being commercialized for various crops. Today a lot of efforts have been made for searching and investigating the PGPB and their mode of action, so that they can be exploited commercially as biofertilizers. Because of the various challenges faced in screening, formulation, and application, PGPB have yet to fulfill their promise and potential as commercial inoculants. Recent progress in our understanding of their diversity, colonization ability, mechanisms of action, formulation, and application should facilitate their development as reliable components in the management of sustainable agricultural systems. Several reviews have discussed specific aspects of PGPB as bioinoculants. We have tried to critically evaluate the current status of bacterial inoculants for contemporary agriculture in developed and developing countries. This review focuses on some important information regarding the biofertilizing potential of some important group of microbes, their formulations, their application for the development of sustainable technology, scope of improvement by genetic engineering, steps to be undertaken for their commercialization and their future prospects.
Purpose In North West-Indo Gangetic Plains (NW-IGP) of India in situ burning of crop residues is practiced by majority of farmers' which deteriorates soil and environmental quality. Fungi have the potential for lignocellulose degradation and can be used for the in situ decomposition of crop residues. Lignocellulose degrading fungal spp. were isolated and evaluated for the activity of lignocellulolytic enzymes. Method The lignocellulose degrading fungi were isolated by appearance of zone on carboxy methyl cellulose (CMC) agar media and tannic acid (TA) media. Carboxy methyl cellulase, filter paperase, cellobiase, xylanase and laccase activity were estimated in submerged, as well as solid state fermentation using a mixture of rice and wheat straw in the ratio of 4:1. rice-wheat straw as substrate. The residue left after solid state fermentation was evaluated for carbon/nitrogen ratio, dry mass loss, and loss of cellulose, hemicellulose and lignin. Selected potential isolates were further tested in pot experiment for their effect on wheat plants. The interaction among isolates was also studied. Result After primary screening, 19 out of a total of 72 fungal isolates were selected based on their enzymatic activity profile and potential to degrade lignocellulosic residues in submerged fermentation. Out of these 19 isolates, 11 were further selected based on their enzymatic secretions in solid state fermentation. All the 11 strains were identified morphologically. Four fungal isolates (RPW 1/3, RPW 1/6, RPWM 2/2 and RZWM 3/2) showed higher enzymatic activities and more loss of dry mass and cell wall constituents over the other isolates. These isolates were identified by ITS region sequencing as Aspergillus flavus, Aspergillus terreus, Penicillium pinophilum and Alternaria alternata. Conclusion This study revealed that fungal isolates may be used for managing crop residues in conservation agriculture based rice-wheat system of NW-IGP to eliminate ill effects of residue burning.
