RNA polymerase IV and V are principal players in the RdDM pathway, where their current study has shown interaction of several factors that control DNA silencing of intergenic regions and siRNA production. DNA silencing is an important process during cell differentiation, nuclear structure and viral control. However, RNA pol IV and V are yet to be study in model monocot systems like Oryza sativa that can provide further information on genetic silence mechanism in plats. We show the expression pattern of these polymerases in tissues extracts of Oryza sativa. Detectable amounts of these polymerases are found in specific adult plant tissues and particularly expressed during somatic embryogenesis but not during early stages of normal embryo development. The use of synthetic auxin leads to an induction of both RNA pol IV and V in scutellum tissue where nuclear localization may be required for genome reorganization and gene silencing.
Large multipeptide protein complexes have provided a challenge for epitope selection, which is required for immunological protocols where native conformations are needed. Immunolocalization requires native conformation of the proteins, which is essential for further understanding of biological activity. RNA polymerase IV and V are multisubunit proteins that interact with other factors in the RNA-directed DNA methylation pathway for control of DNA silencing by small interfering RNA and DNA methylation. DNA silencing is an important process during cell differentiation, nuclear structure and viral control among others. RNA polymerase IV and V are yet to be studied in model monocot systems like Oryza sativa, which may provide further data to define if the genetic silencing mechanism has diverged over time as compared to dicots. Here we show an in silico selection process of exposed sequences and their use for obtaining antibodies against native RNA polymerase IV and V in O. sativa.
El presente trabajo explica la relevancia de los objetos del pasado, de origen doméstico, como materiales estratégicos para la reflexión pretérita y como alternativa a la crisis que enfrenta la enseñanza de la historia de carácter nacionalista. Nuestro objetivo es reflexionar sobre una experiencia didáctica a través del programa piloto titulado El Museo Móvil, que ha sido llevado a diversas escuelas secundarias y preparatorias del Estado de Yucatán, en México. Este ejercicio nos ha demostrado que el empleo de la didáctica del objeto que incorpora materiales con determinada carga simbólica y discursiva de carácter local y regional tiene potencial para articular, con mucho éxito, el proceso de enseñanza-aprendizaje que facilita asumir el pensamiento histórico, en su dimensión de consciencia histórica, que contribuye a formar una ciudadanía crítica y reflexiva.
