The 'saguiru' Steindachnerina insculpta, a small teleost detritivorous fish species in Furnas reservoir, upper rio Paraná basin, Minas Gerais, Brazil, has a prolonged period of reproductive activity that extends from September to March, with fractionated spawning. During gonadal maturation, the females showed a low hepatosomatic index, suggesting that hepatic substances are possibly transferred from liver to ovaries. During the reproductive period, this fish had lower stomach repletion and coelomic fat indexes, indicating that the species eat less and consume more fat reserves. The weight of the ovaries influenced positively the condition factor (K1), since the lowest K2 values (without the gonads weight), were obtained during maturation, confirming the influence of the weight of the ovaries upon the physiological condition of the females.
Fasciola hepatica is a parasite that affects the hepatic ducts of several species of domestic and wild vertebrates, causing huge economic losses to livestock rearing worldwide. Reports on occurrences of F. hepatica in capybaras are an important epidemiological aspect of this disease, since these rodents can be a source of contamination for other animals and humans. In the present study, conducted in a rural area of the municipality of Confins, Minas Gerais, fresh feces from capybaras were collected from the ground near a lagoon at the edge of the Ribeirão da Mata river. These were examined using the technique of four metal sieves. F. hepatica eggs were recovered. This trematode species was confirmed by observing morphological characteristics and measuring the eggs recovered from the capybara feces, and through experimental infection of Lymnaea columella (Say, 1817) by miracidia from these eggs and subsequent infection of C57/BL06 mice with metacercariae originating from these infected mollusks. The data suggest the occurrence of natural cycle of F. hepatica in this region and provide a warning that expansion of the geographical distribution of this parasite by means of this rodent is possible. It is therefore important to adopting measures for epidemiological control of this helminthiasis.
During gonadal maturation the females of Steindachnerina insculpta, a teleost of the Furnas reservoir, Minas Gerais, Brazil, show low hepatosomatic and high gonadossomatic indexes, suggesting possible transference of hepatic substances to the ovaries. In comparison to the resting phase, in the maturation/ mature one the hepatocytes show greater development of both smooth and rough endoplasmic reticula, increased electron density in the mitochondria, and glycogen scattered throughout the cytoplasm. In both resting and maturation/mature phases, the hepatic parenchyma contains melanomacrophagic centers that consist of degenerating cellular debris and are surrounded by capsules formed by cells, resembling fibroblasts, joined by desmosomes. Comparando-se fêmeas no estádio de repouso com aquelas no estádio maturação/maduro, observaramse nos hepatócitos de fêmeas no estádio maturação/maduro maior desenvolvimento do retículo endoplasmático rugoso e liso, mitocôndrias com densidade eletrônica aumentada e glicogênio espalhado por todo citoplasma. No estádio de repouso, assim como no estádio de maturação/maduro, observaram-se, no parênquima hepático, centros melanomacrofágicos constituídos de restos celulares em degeneração delimitados por cápsula contendo células semelhantes a fibroblastos unidas por desmosomas. The lymnaeids are important in the epidemiology of Fasciola hepatica, a neglected and endemic zoonosis. The interaction between the internal defense system of Pseudosuccinea columella and F. hepatica has been little studied. In the present study the effect of infection by F. hepatica on P. columella circulating haemocytes was investigated. Changes in the average number of total circulating haemocytes have been observed at 30 minutes post-infection and 1, 7, 10, 14, 21, 28 and 50 days post-infection (dpi). Miracidia were observed head-foot and mantle at 30 minutes post-infection. Miracidia/Sporocysts in the mantle skirt 1 dpi, and fully formed sporocysts were observed in the head-foot at 7 dpi. Rediae became evident at 10 dpi and were located between the haemocoel and the muscles from 14 dpi; 50 dpi, the rediae in the digestive gland contained cercariae. The statistical analysis of the total haemocytes of P. columella infected by F. hepatica showed significant differences on the 30 minutes post-infection and 1, 14, 21, and 28 dpi in comparison to uninfected molluscs (0 dpi). Therefore, the interference observed on the internal defence system of P. columella may have direct association with the development of F. hepatica.Keywords: Fasciola hepatica, haemocytes, Pseudosuccinea columella. ResumoOs limnaeideos são importantes na epidemiologia de Fasciola hepatica, uma zoonose negligenciada e endêmica. A interação entre o sistema interno de defesa de Pseudosuccinea columella e F. hepatica tem sido pouco estudada. No presente estudo, investigou-se o efeito da infecção por F. hepatica nos hemócitos circulantes de P. columella. Alterações no número médio de hemócitos circulantes foram observadas aos 30 minutos e 1, 7, 10, 14, 21, 28 e 50 dias após a infecção (dpi). Miracídios foram observados na região cefalopodal e manto aos 30 minutos após a infecção. Miracídio/esporocistos foram observados no colar do manto ao 1 dpi, e esporocistos totalmente formados na região cefalopodal aos 7 dpi. Rédias tornam-se evidentes aos 10 dpi entre a hemocele e músculos a partir de 14 dpi; e rédias com cercárias próximas a glândula digestiva aos 50 dpi. A análise estatística dos hemócitos totais de P. columella infectados por F. hepatica demonstrou diferenças significativas nos 30 minutos pós-infecção e 1, 14, 21 e 28 dpi em comparação aos moluscos não infectados (0 dpi). Portanto, a interferência observada no sistema de defesa interna de P. columella pode ter associação direta com o desenvolvimento de F. hepatica.
